    Mucormicose rino-orbito-cerebral num doente oncológico pediátrico

    Aim: Mucormycosis infections are highly lethal in immunosuppressed patients. The authors present a rare case of successful treatment of the condition. Case description: A 14-year-old male with acute lymphoblastic lymphoma in remission was diagnosed with pansinusitis, cerebral osteomyelitis, and encephalitis with an abscess caused by Rhizopus sp. Neurosurgical drainage, endoscopic sinus surgery, and left eye enucleation were performed, and the patient was simultaneously treated with lipossomal-amphotericin B and posaconazol (later replaced by isavuconazol). However, complications (left frontal lobe herniation and cerebrospinal fluid fistula) ensued, requiring surgical reoperation. Chemotherapy was restarted four months later. Comments: This is a rare case report, in which early and aggressive surgery, antifungal treatment, and multidisciplinary team work were crucial for the successful outcome.info:eu-repo/semantics/publishedVersio

    Purification of a lectin with antibacterial activity from Bothrops leucurus snake venom

    A novel lectin was isolated from Bothrops leucurus snake venom using a combination of affinity and gel filtration chromatographies. the lectin (BIL) agglutinated glutaraldehyde-treated rabbit and human erythrocytes with preference for rabbit erythrocytes. Galactose, raffinose, lactose, fetal bovine serum and casein inhibited lectin-induced rabbit erythrocyte agglutination. BIL, with a molecular mass of 30 kDa and composed of two subunits of 15 kDa, showed dependence on calcium. BIL is an acidic protein with highest activity over the pH range of 4.0-7.0 and stable under heating to 70 degrees C. Fluorescence emission spectra showed tryptophan residues partially buried within the lectin structure. the percentages of secondary structure revealed by circular dichroism were 1% alpha-helix, 44% beta-sheet, 24% beta-turn and 31% unordered. BIL showed effective antibacterial activity against Gram-positive bacteria Staphylococcus aureus, Enterococcus faecalis and Bacillus subtilis with minimal inhibitory concentrations of 31.25, 62.25 and 125 mu g/mL, respectively. in conclusion, B. leucurus snake venom contains a galactoside-binding lectin with antibacterial activity.     Evaluation of the insecticidal potential of leaf extracts of Anadenanthera colubrina var. cebil (Griseb) Altschul against Tetranychus urticae Koch and Sitophilus zeamais Mots

    The deterioration and degradation of crops by insects causes economic losses of several billion dollars and affects food security. Sitophilus zeamais and Tetranychus urticae are two economically important species of agricultural crops around the world. The high toxicity of synthetic insecticides and the development of insect resistance to currently used chemicals stimulate the investigation of plant-derived insecticides as new alternatives for pest control. The insecticidal properties of secondary metabolites have been recognized, and have been confirmed scientifically in the last decades. The extract in cyclohexane (Ch) and ethyl acetate (Ea) of the leaves of Anadenanthera colubrina var. cebil collected in the dry and rainy seasons were evaluated for the survival and nutritional physiology of S. zeamais (maize weevil) and for the acaricidal effect front T. urticae (two-spotted spider). All extracts induced mortality of S. zeamais; however, only cyclohexane dry season (Chd) and ethyl acetate in the rainy season (Ear) caused significant mortality (p <0.05), more than 50%, promoting physiological damage to insects in all concentrations tested. In the mortality tests with the spider mite, extracts Chd and Ear showed an LC50 of 52.75 and 156.42 mg.mL-1. Still, in the same test, it can be observed that oviposition was also susceptible to extracts. Therefore, the extracts of A. colubrina are a source of compounds capable of interfering with and impairing the metabolism of the two studied pests

    Terapia cognitiva : aplicações de uma técnica para qualidade de vida e saúde

    Esta pesquisa teve por objetivo geral aplicar e avaliar uma técnica específica de terapia cognitiva - organizada em 12 sessões grupais e denominada Tomada de Decisão e Qualidade de Vida -, destinada a promover saúde e incrementar qualidade de vida. No total, participaram 18 servidores de uma instituição pública de ensino superior. Nas etapas de admissão e de encerramento, aplicaram-se : Questionário de Qualidade de Vida, Inventário Beck de Ansiedade e Inventário Beck de Depressão. Foram identificadas melhoras significativas nos domínios físico, psicológico, meio ambiente, geral e saúde, relacionados à qualidade de vida. Não se verificaram alterações significantes nos escores de ansiedade (p=0,26). Em contrapartida, os escores de depressão indicaram melhora (p=0,02). Os resultados sugerem que a técnica pode ser empregada para promover saúde e qualidade de vida.In this study we implemented and assessed a specific cognitive therapy technique - Decision Making and Quality of Life, which is used to promote health and improve quality of life. Eighteen employees from a higher education institution participated in the study, which was organized into 12 group sessions. At the admission and concluding phases, we asked participants to complete the World Health Organization Quality of Life - Bref Questionnaire, the Beck Anxiety Inventory and the Beck Depression Inventory. Results showed significant improvement in five of the domains that measure quality of life: physical, psychological, environmental, general, and health. There were no significant changes (p=0.26) in anxiety scores. In contrast, the depression scores got significantly better (p=0.02). The results suggest that the proposed technique is conducive to health promotion and quality of life

    Caracterização fitoquímica e avaliação do potencial acaricida e inseticida do óleo essencial de Hymeneae courbaril L. var. courbaril sobre o ácaro-rajado e o gorgulho do milho

    As propriedades inseticidas dos óleos essenciais são geralmente reconhecidas ao longo dos séculos e confirmadas cientificamente nas últimas décadas. Tais produtos podem ser usados na agricultura como uma alternativa para o controle de pragas. O óleo essencial derivado das folhas de Hymenaea courbaril L. var. courbaril foi obtido por hidrodestilação com arraste a vapor e caracterizado fitoquimicamente por cromatografia gasosa/espectrometria de massa (CG/EM); seu potencial acaricida e inseticida foi avaliado contra Tetranychus urticae e Sitophilus zeamais, respectivamente, pragas que ocorrem em uma ampla variedade de culturas economicamente importantes em todo o mundo. Após a análise por CG/EM, foram identificados 36 compostos, sendo 46,42% dos monoterpenos e 51,41% dos sesquiterpenos. Os bioensaios inseticidas avaliaram a interferência do óleo essencial na fisiologia nutricional do gorgulho e no processo de fumigação do ácaro. Os ensaios indicaram que o óleo essencial causou uma mortalidade significativa, promovendo danos fisiológicos ao S. zeamais. O valor de CL50 para T. urticae foi de 35,57 ?L/mL de ar; além disso, também foi observado que a oviposição foi afetada negativamente pelo óleo essencial. O óleo essencial de H. courbaril demonstrou ser um potencial controle de pragas por afetar o metabolismo e a reprodução de ambas as espécies de praga testadas

    Prediction of protein assemblies, the next frontier: The CASP14-CAPRI experiment

    We present the results for CAPRI Round 50, the fourth joint CASP-CAPRI protein assembly prediction challenge. The Round comprised a total of twelve targets, including six dimers, three trimers, and three higher-order oligomers. Four of these were easy targets, for which good structural templates were available either for the full assembly, or for the main interfaces (of the higher-order oligomers). Eight were difficult targets for which only distantly related templates were found for the individual subunits. Twenty-five CAPRI groups including eight automatic servers submitted ~1250 models per target. Twenty groups including six servers participated in the CAPRI scoring challenge submitted ~190 models per target. The accuracy of the predicted models was evaluated using the classical CAPRI criteria. The prediction performance was measured by a weighted scoring scheme that takes into account the number of models of acceptable quality or higher submitted by each group as part of their five top-ranking models. Compared to the previous CASP-CAPRI challenge, top performing groups submitted such models for a larger fraction (70–75%) of the targets in this Round, but fewer of these models were of high accuracy. Scorer groups achieved stronger performance with more groups submitting correct models for 70–80% of the targets or achieving high accuracy predictions. Servers performed less well in general, except for the MDOCKPP and LZERD servers, who performed on par with human groups.     Sarcoma de Ewing ou Linfoma Ósseo

    Apresenta-se o caso clínico de uma criança de 4 anos, sexo masculino com história de dor no membro inferior direito, claudicação da marcha e exame objectivo sem outras alterações. Oa exames imagiológicos mostraram uma lesão osteolítica desde o colo até ao terço distai da diáfise femural. O doente foi referenciado para a nossa Instituição com o diagnóstico provável de Sarcoma de Ewing versus linfoma, Com o objectivo de estadiar o tumor foi realizada uma tomografia axial computorizada (TAC) abdominal que revelou uma massa sólida intra-renal á esquerda. Foram realizadas duas biópsias (fémur e rim) que confirmaram o diagnóstico de linfoma linfoblástico de células da linhagem B. Este caso demonstra que o diagnóstico de linfoma ósseo deve ser incluído no grupo de diagnósticos diferenciais de tumores ósseos, ainda que a ocorrência deste tipo de tumor numa criança seja rara.Palavras-Chave - Tumor ósseo, linfoma de células   sarcoma de Ewin
