    Redefining effect size interpretations for psychotherapy RCTs in depression

    Introduction: Effect sizes are often used to interpret the magnitude of a result and in power calculations when planning research studies. However, as effect size interpretations are context-dependent, Jacob Cohen’s suggested guidelines for what represents a small, medium, and large effect are unlikely to be suitable for a diverse range of research populations and interventions. Our objective here is to determine empirically-derived effect size thresholds associated with psychotherapy randomized controlled trials (RCTs) in depression by calculating the effect size distribution. Methods: We extracted effect sizes from 366 RCTs provided by the systematic review of Cuijpers and colleagues (2020) on psychotherapy for depressive disorders across all age groups. The 50th percentile effect size, as this represents a medium effect size, and the 25th (small) and 75th (large) percentile effect sizes were calculated to determine empirically-derived effect size thresholds. Results: After adjusting for publication bias, 0.27, 0.53, and 0.86 represent small, medium, and large effect sizes, respectively, for psychotherapy treatment for depressive disorders. Discussion: The effect size distribution for psychotherapy treatment of depression indicates that observed effect size thresholds are larger than Cohen’s suggested effect size thresholds (0.2, 0.5, and 0.8). These results have implications for the interpretation of study effects and the planning of future studies via power analyses, which often use effect size thresholds.publishedVersio

    Mitochondrial fatty acid β-oxidation disorders: from disease to lipidomic studies—a critical review

    Fatty acid oxidation disorders (FAODs) are inborn errors of metabolism (IEMs) caused by defects in the fatty acid (FA) mitochondrial β-oxidation. The most common FAODs are characterized by the accumulation of medium-chain FAs and long-chain (3-hydroxy) FAs (and their carnitine derivatives), respectively. These deregulations are associated with lipotoxicity which affects several organs and potentially leads to life-threatening complications and comorbidities. Changes in the lipidome have been associated with several diseases, including some IEMs. In FAODs, the alteration of acylcarnitines (CARs) and FA profiles have been reported in patients and animal models, but changes in polar and neutral lipid profile are still scarcely studied. In this review, we present the main findings on FA and CAR profile changes associated with FAOD pathogenesis, their correlation with oxidative damage, and the consequent disturbance of mitochondrial homeostasis. Moreover, alterations in polar and neutral lipid classes and lipid species identified so far and their possible role in FAODs are discussed.     Kinetics of the pollutant photocatalytic conversion in a Photo-CREC-Air Reactor

    This research reports the kinetic studies for acetone and acetaldehyde photoconversion in the gas phase utilizing a scaled-up Photo-CREC-Air unit with TiO2. The inclusion of intermediate species in the reaction network is required for the kinetic modeling of a wide range of oxygenate pollutant concentrations. The proposed ‘‘in parallel-series” reaction network encompasses a Langmuir-Hinshelwood (L-H) kinetics including species adsorption and intrinsic reaction parameters. The estimated kinetic parameters provide a successful prediction of various measurable chemical species. It is shown that the proposed kinetic model can be simplified, under low initial model pollutant concentrations. This is critical to avoid model overparameterization. The proposed kinetic model while being restricted to two model pollutants (ace- tone and acetaldehyde) and one specific photoreactor (e.g. 55.1 L Photo-CREC-Air), provides a framework to establish the photodegradation kinetics of other organic species in air, for larger photoreactor scales

    U–Pb ages and Hf-isotope data of detrital zircons from the late Neoarchean-Paleoproterozoic Minas Basin, SE Brazil

    Because of its world-class iron ore deposits and promising Au and U mineralizations, the late Neoarchean to Paleoproterozoic Minas Basin (Minas Supergroup, SE of Brazil) is one of the best-studied basins in South America. However, the lack of datable interlayered volcanic rocks prevented discourse over ages of the strata, the sources and the nature of its ore deposits. In this paper, we present detrital zircon U–Pb age patterns coupled with Lu–Hf data for 18 samples, representing different stages of the Minas Basin evolution (∼2000 analyzed zircons). Age spectra for the main basal unit (Moeda Formation) show a classic rift-related detrital zircon pattern, characterized by multiple autochthonous sources, which in turn are much older than the age of deposition. Maximum age for the rifting event is constrained at ca. 2600 Ma. Detritus accumulated at the base of the Minas Supergroup were derived from Archean source rocks and their sedimentation was marked by differential uplift of the Archean crust, shortly after the 2730–2600 Ma high-K calc-alkaline magmatism (Mamona Event). The age of the BIF deposits is younger than 2600 Ma, most likely coinciding with the great oxygenation event between 2400 and 2200 Ma and the precipitation of banded iron deposits worldwide. Detrital zircons from the topmost units of the Minas strata suggest that tectonic inversion and closure of the basin took place at ca. 2120 Ma with the deposition of the synorogenic Sabará Group. Rhyacian zircon supply showing juvenile Hf signatures gives evidence of a late Rhyacian amalgamation between the Mineiro Belt and the craton. The εHf signatures support the hypothesis that the Archean crystalline crust of the craton was mostly built by crust–mantle mixing processes, with a successive decrease of εHf values in zircons crystallized after 3250 Ma and minor mantle-like additions after Paleoarchean times. Regionally, our dataset supports previous interpretations of a long-lived evolution of the southern São Francisco Craton comprising a succession of convergent island arcs, small microplate collisions, and development of Archean convergent and divergent basins that evolved between Archean and Paleoproterozoic times.Fil: Martínez Dopico, Carmen Irene.     Dried blood spots in clinical lipidomics: optimization and recent findings

    Dried blood spots (DBS) are being considered as an alternative sampling method of blood collection that can be used in combination with lipidomic and other omic analysis. DBS are successfully used in the clinical context to collect samples for newborn screening for the measurement of specifc fatty acid derivatives, such as acylcarnitines, and lipids from whole blood for diagnostic purposes. However, DBS are scarcely used for lipidomic analysis and investigations. Lipidomic stud ies using DBS are starting to emerge as a powerful method for sampling and storage in clinical lipidomic analysis, but the major research work is being done in the pre- and analytical steps and procedures, and few in clinical applications. This review presents a description of the impact factors and variables that can afect DBS lipidomic analysis, such as the type of DBS card, haematocrit, homogeneity of the blood drop, matrix/chromatographic efects, and the chemical and physi cal properties of the analyte. Additionally, a brief overview of lipidomic studies using DBS to unveil their application in clinical scenarios is also presented, considering the studies of method development and validation and, to a less extent, for clinical diagnosis using clinical lipidomics. DBS combined with lipidomic approaches proved to be as efective as whole blood samples, achieving high levels of sensitivity and specifcity during MS and MS/MS analysis, which could be a useful tool for biomarker identifcation.     Validación psicométrica del instrumento audio guiado rBVQ para la evaluación del bullying

    The revised Olweus Bully/Victim Questionnaire (rBVQ) is an internationally used instrument to identify bullying, but due to Brazilian students’ low proficiency in reading and writing, it had to be adapted for audio-guided use through smartphones. We investigated construct validity, using Confirmatory Factor Analysis (CFA), under a two-correlated factor solution. Participated 1,742 fifth graders and 2,316 seventh graders from 30 public schools in the city of São Paulo. We found excellent fit indices models for both grades (fifth grade: CFI=0.985, RMSEA=0.020; seventh grade: CFI=0.990, RMSEA=0.015), as well as factor loading higher than 0.4 for all items, indicating their relevance to the construct. The rBVQ demonstrated excellent psychometric properties and may help future studies that aim to investigate bullying in populations with similar profiles. In addition, this study innovates by evaluating an audio-guided instrument and using mobile technology.El Cuestionario Revisado de Bullying de Olweus (rBVQ) es un instrumento utilizado internacionalmente para identificar bullying, pero debido a las dificultades de estudiantes brasileños en la lectura y la escritura, se ha adaptado su uso por medio de audioguías en smartphones. Se investigó la validez del constructo mediante el Análisis Factorial Confirmatorio, bajo un modelo de dos factores. Participaron 1742 y 2316 estudiantes de quinto y séptimo curso de 30 escuelas públicas de São Paulo. Excelentes tasas de adecuación fueron encontradas para ambos casos (quinto año: CFI=0,985, RMSEA=0,020; séptimo año: CFI=0,990, RMSEA=0,015), además de cargas factoriales superiores a 0,4 para todos los ítems, lo que indica su relevancia para el constructo. El instrumento rBVQ ha presentado excelentes propiedades psicométricas y puede ayudar a futuros estudios pretendan investigar el bullying en poblaciones con perfiles similares.     Brazilian cross-cultural adaptation and validation of the List of Threatening Events Questionnaire (LTE-Q)

    Objective: To perform a construct validation of the List of Threatening Events Questionnaire (LTE-Q), as well as convergence validation by identifying its association with drug use in a sample of the Brazilian population. Methods: This is a secondary analysis of the Second Brazilian National Alcohol and Drugs Survey (II BNADS), which used a cross-cultural adaptation of the LTE-Q in a probabilistic sample of 4,607 participants aged 14 years and older. Latent class analysis was used to validate the latent trait adversity (which considered the number of events from the list of 12 item in the LTE experienced by the respondent in the previous year) and logistic regression was performed to find its association with binge drinking and cocaine use. Results: The confirmatory factor analysis returned a chi-square of 108.341, weighted root mean square residual (WRMR) of 1.240, confirmatory fit indices (CFI) of 0.970, Tucker-Lewis index (TLI) of 0.962, and root mean square error approximation (RMSEA) score of 1.000. LTE-Q convergence validation showed that the adversity latent trait increased the chances of binge drinking by 1.31 time and doubled the chances of previous year cocaine use (adjusted by sociodemographic variables). Conclusion: The use of the LTE-Q in Brazil should be encouraged in different research fields, including large epidemiological surveys, as it is also appropriate when time and budget are limited.     The polar lipidome of cultured Emiliania huxleyi: a source of bioactive lipids with relevance for biotechnological applications

    Polar lipids from microalgae have aroused greater interest as a natural source of omega-3 (n-3) polyunsaturated fatty acids (PUFA), an alternative to fish, but also as bioactive compounds with multiple applications. The present study aims to characterize the polar lipid profile of cultured microalga Emiliania huxleyi using hydrophilic interaction liquid chromatography coupled with high-resolution mass spectrometry (HILIC-MS) and fatty acids (FA) analysis by gas chromatography (GC-MS). The lipidome of E. huxleyi revealed the presence of distinct n-3 PUFA (40% of total FA), namely docosahexaenoic acid (22:6n-3) and stearidonic acid (18:4n-3), which give this microalga an increased commercial value as a source of n-3 PUFA present in the form of polar lipids. A total of 134 species of polar lipids were identified and some of these species, particularly glycolipids, have already been reported for their bioactive properties. Among betaine lipids, the diacylglyceryl carboxyhydroxymethylcholine (DGCC) class is the least reported in microalgae. For the first time, monomethylphosphatidylethanolamine (MMPE) has been found in the lipidome of E. huxleyi.     Potential of Coccolithophore microalgae as fillers in starch-based films for active and sustainable food packaging

    Coccolithophore microalgae, such as Emiliania huxleyi (EHUX) and Chrysotila pseudoroscoffensis (CP), are composed of calcium carbonate (CaCO3) and contain bioactive compounds that can be explored to produce sustainable food packaging. In this study, for the first time, these microalgae were incorporated as fillers in starch-based films, envisioning the development of biodegradable and bioactive materials for food packaging applications. The films were obtained by solvent casting using different proportions of the filler (2.5, 5, 10, and 20%, w/w). For comparison, commercial CaCO3, used as filler in the plastic industry, was also tested. The incorporation of CaCO3 and microalgae (EHUX or CP) made the films significantly less rigid, decreasing Young’s modulus up to 4.7-fold. Moreover, the incorporation of microalgae hydrophobic compounds as lipids turned the surface hydrophobic (water contact angles > 90°). Contrary to what was observed with commercial CaCO3, the films prepared with microalgae exhibited antioxidant activity, increasing from 0.9% (control) up to 60.4% (EHUX 20%) of ABTS radical inhibition. Overall, the introduction of microalgae biomass improved hydrophobicity and antioxidant capacity of starch-based films.     State, trait, and accumulated features of the Alzheimer's Disease Assessment Scale Cognitive Subscale (ADAS-Cog) in mild Alzheimer's disease

    Background The Alzheimer's Disease Assessment Scale Cognitive Subscale (ADAS-Cog) is used to assess decline in memory, language, and praxis in Alzheimer's disease (AD). Methods A latent state–trait model with autoregressive effects was used to determine how much of the ADAS-Cog item measurement was reliable, and of that, how much of the information was occasion specific (state) versus consistent (trait or accumulated from one visit to the next). Results Participants with mild AD (n = 341) were assessed four times over 24 months. Praxis items were generally unreliable as were some memory items. Language items were generally the most reliable, and this increased over time. Only two ADAS-Cog items showed reliability >0.70 at all four assessments, word recall (memory) and naming (language). Of the reliable information, language items exhibited greater consistency (63.4% to 88.2%) than occasion specificity, and of the consistent information, language items tended to reflect effects of AD progression that accumulated from one visit to the next (35.5% to 45.3%). In contrast, reliable information from praxis items tended to come from trait information. The reliable information in the memory items reflected more consistent than occasion-specific information, but they varied between items in the relative amounts of trait versus accumulated effects. Conclusions Although the ADAS-Cog was designed to track cognitive decline, most items were unreliable, and each item captured different amounts of information related to occasion-specific, trait, and accumulated effects of AD over time. These latent properties complicate the interpretation of trends seen in ordinary statistical analyses of trials and other clinical studies with repeated ADAS-Cog item measures
