    Control capacity of the LPSc 1067 strain of Beauveria bassiana (Ascomycota: Hypocreales) on different species of grasshoppers (Orthoptera: Acrididae: Melanoplinae), agricultural pests in Argentina

    Grasshoppers affect agriculture worldwide, causing serious economic damage. Currently, the application of chemical insecticides against grasshoppers is the only effective strategy, even considering the significant environmental concern. This study aimed to test the entomopathogenic fungi Beauveria bassiana (LPSc 1067) as biocontrol agent on six harmful grasshopper species in Argentina. Significant differences were observed (DF= 5; F= 9.93; P<0.0001) when considering B. bassiana pathogenicity on third-instar nymphs of the different grasshopper species. The highest mortality (100%) was registered on Trimerotropis pallidipennis and Dichroplus maculipennis nymphs while the lowest mortality (48.6 ±3.5%) was observed on Scotussa lemniscata nymphs. The lowest mean survival time (MST) was recorded for T. pallidipennis (3.5 ±0.15 days) and the highest MST was observed on Dichroplus pratensis nymphs (7.48 ±0.28 days). Results suggest that B. bassiana LPSc 1067 may constitute an excellent candidate to be further studied as biological control agent of T. pallidipennis and D. maculipennis.     Potential use of cardoon oil for biodiesel production and its comparison with soybean oil

    El cardo (silvestre y cultivado) es una especie con potencial para ser utilizado como cultivo energético con aprovechamiento integral para producción de biocombustible sólidos (pellets) y líquidos (bioetanol y biodiesel). Los objetivos del presente trabajo fueron estudiar el perfil de ácidos grasos del aceite de cardo mediante técnicas químicas específicas, y la comparación con el perfil de ácidos grasos del aceite de soja. Los resultados obtenidos demuestran que el aceite de cardo presenta características adecuadas para ser destinado a la producción de biodiesel lo que permitiría la incorporación de un nuevo cultivo alternativo al circuito productivo-comercial con fines energéticos.Cardoon (wild and cultivated) is a species with potential to be fully used as an energy crop for solid (pellets) and liquid (bioethanol and biodiesel) biofuels production. The objective of this work was to study the fatty acids profile of cardoon oil by specific chemical techniques and its comparison with the fatty acids profile of Soybean oil. En la actualidad, los insecticidas químicos siguen siendo el único medio utilizado para el control de acridios, pero los efectos de su utilización son ambientalmente preocupantes. El objetivo de este trabajo fue probar la eficacia de la cepa Beauveria bassiana (LPSc 1067) sobre seis especies de tucuras consideradas plagas de Argentina. En cuanto a la patogenicidad de B. bassiana sobre ninfas de tercer estadio de las diferentes especies tratadas, se encontraron diferencias significativas (DF= 5; F= 9.93; P<0.0001). Los valores de mortalidad más altos (100%) se registraron en ninfas de Trimerotropis pallidipennis y Dichroplus maculipennis y la mortalidad más baja se observó en ninfas de Scotussa lemniscata con una mortalidad de 48,6 +3,5%. El tiempo medio de supervivencia (MST) más bajo se registró para T. pallidipennis (3,5 +0,15 días) y el MST más alto se observó en ninfas de Dichroplus pratensis (7,48 +0,28 días). Los resultados sugieren que B. bassiana LPSc 1067 constituye un excelente candidato para ser estudiado en profundidad como agente de control biológico de T. pallidipennis y D. maculipennis.Fil: Pelizza, Sebastian Alberto.     In this work the development and the results of a practical work are shown. The practical work has been designed for the teaching of organic chemistry in a high school teaching course. The selected theme is related to the perfume industry. Its main goal was related to promoting the study of the chemical discipline. This work was carried out within five weeks. It contemplates the fundamental abilities of reading, writing, orality and critical thinking. The results obtained indicate that it was possible to consolidate contents, relate them to others previously developed, improve the writing and presentation of reports, develop collaborative work, and improve skills in the handling of laboratory material.Fil: Guntero, Vanina Alejandra.     Development of a modified transformation platform for apomixis candidate genes research in Paspalum notatum (bahiagrass)

    Abstract The aim of this work was to improve existing transformation protocols and to transform specific genotypes of Paspalum notatum (bahiagrass) for functional analyses of candidate genes involved in reproduction. Three different explants were assayed for in vitro plant regeneration: mature seeds, mature embryos, and shoot meristems. Plant regeneration was achieved with all explant types, but mature seeds produced the optimal rate (78.0%) and were easiest to manipulate. A method based on serial re-induction of calli from meristems of the regenerated lines was also developed, which could be useful in plant breeding strategies pursuing somaclonal variation. Transient transformation experiments were performed on calli obtained from mature seeds using a compressed helium gene gun. Transient transformation constructs included anthocyanin-synthesis genes cloned under the CAMV 35S promoter and an enhanced green fluorescent protein gene (egfp) driven by the rice actin1 (act1) promoter. Selection curves for ammonium glufosinate were developed in order to determine the optimal selective pressure for stable transformation (1.0 mg/L). Stable co-transformation experiments were carried out with two different constructs containing: (1) the reporter egfp gene cloned under the rice act1 promoter and (2) the selector bar gene driven by the ubiquitin promoter. A total of 27 (64.2%) transgenic plants out of 42 resistant plants analyzed were obtained. The presence of the transgenes in regenerated plants was confirmed by polymerase chain reaction and DNA gel blot analysis. Gene expression was demonstrated by eGFP fluorescence detection and in vivo assays for ammonium glufosinate tolerance.     Characterization and expression analysis of Somatic Embryogenesis Receptor Kinase (SERK) genes in sexual and apomictic Paspalum notatum

    The Somatic Embryogenesis Receptor Kinase (SERK) gene plays a fundamental role in somatic embryogenesis of angiosperms, and is associated with apomixis in Poa pratensis. The objective of this work was to isolate, characterize and analyze the expression patterns of SERK genes in apomictic and sexual genotypes of Paspalum notatum. A conserved 200-bp gene fragment was amplified from genomic DNA with heterologous primers, and used to initiate a chromosomal walking strategy for cloning the complete sequence. This procedure allowed the isolation of two members of the P. notatum SERK family; PnSERK1, which is similar to PpSERK1, and PnSERK2, which is similar to ZmSERK2 and AtSERK1. Phylogenetic analyses indicated that PnSERK1 and PnSERK2 represent paralogous sequences. Southern-blot hybridization indicated the presence of at least three copies of SERK genes in the species. qRT-PCR analyses revealed that PnSERK2 was expressed at significantly higher levels than PnSERK1 in roots, leaves, reproductive tissues and embryogenic calli.Moreover, in situ hybridization experiments revealed that PnSERK2 displayed a spatially and chronologically altered expression pattern in reproductive organs of the apomictic genotype with respect to the sexual one. PnSERK2 is expressed in nucellar cells of the apomictic genotype at meiosis, but only in the megaspore mother cell in the sexual genotype.     Porte de Montmartre at the rhythm of the 15-Minute City

    openThis research revolves around the investigation of the claim that the 15-Minute City facilitates urban development towards a more sustainable development, providing more accessible services, fostering an ecological use of facilities, and creating occasions for people’s interactions. It does so by investigating the case study of the neighbourhood of Porte de Montmartre, in the 18th arrondissement of Paris. It qualifies as a single case study and a research of qualitative nature, for it pursues an investigation of officially released material, supported by semi-structured interview conducted with particularly engaged citizens i.e. those representing cultural and social association in the neighbourhood. Central to this study is the ‘use’ of the territory, for time and space in the urban dynamics are understood as socially constructed, and therefore continuously shaped by relations immersed in a geography of power. After having contextualised the neighbourhood of Porte de Montmartre, the findings are organised combining information emerging from the desk research and the interviews in categories relevant for identifying and presenting relevant themes. These categories revolves around different sphere of life e.g. commercial, related to health care and safety, and two main tools central to the 15-Minute City i.e. the use of school yards and the Kiosque Citoyen. Finally, it is revealed how the 15-Minute City does not present a revolutionary character, nor is to be considered inclusive or democratic as a policy. In fact, as a policy the 15-Minute City has to be understood as an orientation, while the real change is expected to come from the local actors. The local reality of Porte de Montmartre appears to functioning in meeting its residents’ needs and desire, especially though collaboration - although improvements can be made to encourage different social groups to mingle. Interestingly, the cultural and social associations seems to be the strength of the territory, instrumental in maintaining and stimulating social life
