miR-375 gene dosage in pancreatic β-cells: implications for regulation of β-cell mass and biomarker development

MicroRNAs play a crucial role in the regulation of cell growth and differentiation. Mice with genetic deletion of miR-375 exhibit impaired glycemic control due to decreased β-cell and increased α-cell mass and function. The relative importance of these processes for the overall phenotype of miR-375KO mice is unknown. Here, we show that mice overexpressing miR-375 exhibit normal β-cell mass and function. Selective re-expression of miR-375 in β-cells of miR-375KO mice normalizes both, α- and β-cell phenotypes as well as glucose metabolism. Using this model, we also analyzed the contribution of β-cells to the total plasma miR-375 levels. Only a small proportion (≈1 %) of circulating miR-375 originates from β-cells. Furthermore, acute and profound β-cell destruction is sufficient to detect elevations of miR-375 levels in the blood. These findings are supported by higher miR-375 levels in the circulation of type 1 diabetes (T1D) subjects but not mature onset diabetes of the young (MODY) and type 2 diabetes (T2D) patients. Together, our data support an essential role for miR-375 in the maintenance of β-cell mass and provide in vivo evidence for release of miRNAs from pancreatic β-cells. The small contribution of β-cells to total plasma miR-375 levels make this miRNA an unlikely biomarker for β-cell function but suggests a utility for the detection of acute β-cell death for autoimmune diabetes

Model based methodology development for energy recovery in ash heat exchange systems

Flash tank evaporation combined with a condensing heat exchanger can be used when heat exchange is required between two streams and where at least one of these streams is difficult to handle (in terms of solid particles content, viscosity, pH, consistency etc.). To increase the efficiency of heat exchange, a cascade of these units in series can be used. Heat transfer relationships in such a cascade are very complex due to their interconnectivity, thus the impact of any changes proposed is difficult to predict. In this report, a mathematical model of a single unit ash tank evaporator combined with a condensing heat exchanger unit is proposed. This model is then developed for a chain of the units. The purpose of this model is to allow an accurate evaluation of the effect and result of an alteration to the system. The resulting model is applied to the RUSAL Aughinish Alumina digester area

The Penguin: a Low Reynolds Number Powered Glider for Station Keeping Missions

    Get PDF
The Penguin is a low Reynolds number (approx. 100,000) remotely piloted vehicle (RPV). It was designed to fly three laps indoors around two pylons in a figure-eight course while maximizing loiter time. The Penguin's low Reynolds number mission is an important one currently being studied for possible future flights in the atmospheres of other planets and for specialized military missions. Although the Penguin's mission seemed quite simple at first, the challenges of such low Reynolds number flight have proven to be quite unique. In addition to the constraint of low Reynolds number flight, the aircraft had to be robust in its control, highly durable, and it had to carry a small instrument package. The Penguin's flight plan, concept, performance, aerodynamic design, weight estimation, structural design, propulsion, stability and control, and cost estimate is detailed

Counts and Colors of Faint Galaxies in the U and R Bands

    Get PDF
Ground-based counts and colors of faint galaxies in the U and R bands in one field at high Galactic latitude are presented. Integrated over flux, a total of 1.2x10^5 sources per square degree are found to U=25.5 mag and 6.3x10^5 sources per square degree to R=27 mag, with d log N/dm ~ 0.5 in the U band and d log N/dm ~ 0.3 in the R band. Consistent with these number-magnitude curves, sources become bluer with increasing magnitude to median U-R=0.6 mag at 24<U<25 mag and U-R=1.2 mag at 25 < R < 26 mag. Because the Lyman break redshifts into the U band at z~3, at least 1.2x10^5 sources per square degree must be at redshifts z<3. Measurable U-band fluxes of 73 percent of the 6.3x10^5 sources per square degree suggest that the majority of these also lie at z < 3. These results require an enormous space density of objects in any cosmological model.Comment: 17 pages, MNRAS in pres

Extremely metal-poor stars from the SDSS

We give a progress report about the activities within the CIFIST Team related to the search for extremely metal-poor stars in the Sloan Digital Sky Survey's spectroscopic catalog. So far the search has provided 25 candidates with metallicities around or smaller -3. For 15 candidates high resolution spectroscopy with UVES at the VLT has confirmed their extremely metal-poor status. Work is under way to extend the search to the SDSS's photometric catalog by augmenting the SDSS photometry, and by gauging the capabilities of X-shooter when going to significantly fainter targets.Comment: 6 pages, 6 figures, Proceedings paper of the conference "A stellar journey: A symposium in celebration of Bengt Gustafsson's 65th birthday
