    Invasive pleural malignant mesothelioma with rib destruction and concurrent osteosarcoma in a dog

    A 7-year-old Dachshund was clinically examined because of a 10-day history of lameness in the left hind limb. On the basis of radiological and cytological findings, an osteosarcoma of the left acetabular region was suspected. The dog underwent a hemipelvectomy and osteosarcoma was diagnosed by subsequent histopathological examination. An immovable subcutaneous mass was noted on the left chest wall during the physical examination and non-septic neutrophilic inflammation was diagnosed by cytology. Forty days later, the dog showed signs of respiratory distress with an in-diameter increase of the subcutaneous mass up to 4 cm. Thoracic radiography and ultrasonography revealed pleural effusion and a lytic process in the fourth left rib. Furthermore, ultrasound examination revealed a mixed echogenic mobile structure with a diameter of around 2 cm floating within the pleural fluid of the left hemithorax close to the pericardium. The dog underwent surgery for an en bloc resection of the subcutaneous mass together with the fourth rib and the parietal pleura. Moreover, the left altered lung lobe, corresponding to the mobile structure detected by ultrasound, was removed. Based on cytological, histopathological, and immunohistochemical examinations, an invasive epithelioid pleural malignant mesothelioma was diagnosed

    Cyclic Peptides Acting as Allosteric Inhibitors of Human Thymidylate Synthase and Cancer Cell Growth

    Thymidylate synthase (TS) is a prominent drug target for different cancer types. However, the prolonged use of its classical inhibitors, substrate analogs that bind at the active site, leads to TS overexpression and drug resistance in the clinic. In the effort to identify anti-TS drugs with new modes of action and able to overcome platinum drug resistance in ovarian cancer, octapeptides with a new allosteric inhibition mechanism were identified as cancer cell growth inhibitors that do not cause TS overexpression. To improve the biological properties, 10 cyclic peptides (cPs) were designed from the lead peptides and synthesized. The cPs were screened for the ability to inhibit recombinant human thymidylate synthase (hTS), and peptide 7 was found to act as an allosteric inhibitor more potent than its parent open-chain peptide [Pro3]LR. In cytotoxicity studies on three human ovarian cancer cell lines, IGROV-1, A2780, and A2780/CP, peptide 5 and two other cPs, including 7, showed IC50 values comparable with those of the reference drug 5-fluorouracil, of the open-chain peptide [d-Gln4]LR, and of another seven prolyl derivatives of the lead peptide LR. These promising results indicate cP 7 as a possible lead compound to be chemically modified with the aim of improving both allosteric TS inhibitory activity and anticancer effectiveness

    Duration of Untreated Disorder and Cannabis Use: An Observational Study on a Cohort of Young Italian Patients Experiencing Psychotic Experiences and Dissociative Symptoms

It is an important predictor of several disease-related outcomes in psychotic disorders. The aim of this manuscript is investigating the influence of cannabis on the DUP and its clinical correlates. Methods: During years 2014−2019, sixty-two FEP patients with and without cannabis use disorder (CUD) were recruited from several Italian psychiatric hospitals. The subjects were then divided into two groups based on the duration of the DUP and assessed at the beginning of the antipsychotic treatment and after 3 and 6 months, using the Positive and Negative Syndrome Scale (PANSS), the Global Assessment of Functioning (GAF) scale, and the Dissociative Experiences Scale (DES-II). Results: As expected, a longer DUP was associated with worse symptoms and cannabis use did not seem to affect the DUP, but both were related with more dissociative symptoms at onset and over time. Discussion: According to our study, cannabis use can be a predictor of FEP and DUP, and of disease outcome. However, several factors might influence the relationship between cannabis use and DUP. Preventing cannabis use and early diagnosis of psychotic disorders might impact the disease by reducing the persistence of symptoms and limiting dissociative experiences.Peer reviewedFinal Published versio

    SATURN: A Technological Demonstration Mission for Distributed SAR Imaging

    The OHB-Italia S.p.A-led consortium is in the midst of Phase B of SATURN (Synthetic AperTure radar cUbesat foRmation flyiNg), part of ALCOR, an Italian Space Agency (ASI) programme promoting the development of the next generation Italian CubeSats. SATURN is a demonstration mission that features Multiple-Input-Multiple-Output (MIMO) technology applied to a Swarm of CubeSats equipped with Synthetic Aperture Radar (SAR) for Earth Observation. MIMO is based on cooperative active sensors, where each one transmits signals and receives the illuminated common area backscatter related to the entire swarm, increasing measurement performances with a trend approximatively equal to the square of the number of sensors. The complete SATURN constellation features 16 mini-swarms, each of 3 CubeSats, spread over 4 SSOs equally spaced by 3 hours of local time. The constellation is designed to provide an average revisit time of 1.5 h and an interferometric revisit time of 1 day worldwide. The aim of this demonstration mission is to verify MIMO technology applied to SAR on a mini-swarm of 3 CubeSats in close formation on a Low Earth Down-Dusk Sun Synchronous Orbit. Using OHB-I’s M3Multi Mission Modular platform equipped with a miniaturized SAR Instrument, developed by ARESYS S.r.l. and Airbus Italia S.p.A., our mission is able to achieve a resolution of 5x5 m over a 30 km swath. Thus, SATURN enables low-cost, scalable SAR missions for affordable access to space for public and private entities, overcoming the single point of failure of one large and complex satellite. Subsequent swarms, deploying from 3 to 48 CubeSats, are expected to bring technological innovations and improve Italy’s competitiveness in the European and global Earth Observation scenario

    Phase II Randomized Study of Vandetanib Plus Gemcitabine or Gemcitabine Plus Placebo as First-Line Treatment of Advanced Non–Small-Cell Lung Cancer in Elderly Patients

    Introduction:The aim of the present study was to evaluate the efficacy and tolerability of vandetanib plus gemcitabine (V/G) compared with gemcitabine alone in elderly patients with untreated advanced non–small-cell lung cancer.Methods:This was a phase II, randomized, double-blind study. A total of 124 elderly patients (mean age, 75 yr; age range, 70–84 yr; 73% men) received V/G (n = 61) or placebo plus gemcitabine (n = 63). Progression-free survival (PFS) was the primary endpoint. Secondary endpoints were overall survival, objective response rate, duration of response, disease control rate, time to deterioration of performance status, and safety outcomes.Results:PFS was significantly prolonged with V/G (median, 183 days; 95% confidence interval, 116–214) compared with placebo plus gemcitabine (median, 169 days; 95% confidence interval, 95–194; p = 0.047). No statistically significant differences between arms were observed in all secondary endpoints, including overall survival. The addition of vandetanib to gemcitabine was well tolerated. The rate of patients with ≥1 treatment-related adverse event was comparable in the two arms, pyrexia, dyspnea, and neutropenia being the most common adverse events.Conclusions:V/G combination was associated with a statistically significant prolongation of PFS compared with gemcitabine alone in untreated elderly patients with advanced non–small-cell lung cancer, with an acceptable safety profile

    Proposal for the Global Boundary Stratotype Section and Point (GSSP) for the Priabonian Stage (Eocene) at the Alano section (Italy)

    The base of the Priabonian Stage is one of two stage boundaries in the Paleogene that remains to be formalized. The Alano section (NE Italy) was elected by consensus as a suitable candidate for the base of the Priabonian during the Priabonian Working Group meeting held in Alano di Piave in June 2012. Further detailed research on the section is now followed by a formal proposal, which identifies the base of a prominent crystal tuff layer, the Tiziano bed, at meter 63.57 of the Alano section, as a suitable candidate for the Priabonian Stage. The choice of the Tiziano bed is appropriate from the historical point of view and several bio-magnetostratigraphic events are available to approximate this chronostratigraphic boundary and guarantee a high degree of correlatability over wide geographic areas. Events which approximate the base of the Priabonian Stage in the Alano section are the successive extinction of large acarininids and Morozovelloides (planktonic foraminifera), the Base of common and continuous Cribrocentrum erbae and the Top of Chiasmolithus grandis (nannofossils), as well as the Base of Subchron C17n.2n and the Base of Chron C17n (magnetostratigraphy). Cyclostratigraphic analysis of the Bartonian-Priabonian transition of the Alano section as well as radioisotopic data of the Tiziano tuff layer provide an absolute age (37.710 – 37.762 Ma, respectively) of this bed and, consequently, of the base of the Priabonian Stage
