    COAUTHOR - a MoU to create a COnsortium of Academics from Universities promoting the use of THORium

    This paper describes the Memorandum of Understanding (MoU) signed by the authors to create a future consortium of academics from universities to promote the utilization of thorium (COAUTHOR). Besides the description of the MoU, also results of the research conducted in each participating partner or collaborative work performed among them will be described. Finally, the future work planned in the framework of the MoU, will be discussed

    Nuclear Energy, primarily to produce electricity and other use, and the enveloping Nuclear Technology, as inherited from the XX Century, constitutes a controversial issue for political and economic reasons. On the one hand, the energy source is promoted in several Countries and an unavoidable mean to ensure growth for the human civilization ad suitable living standard with reduced or no impact upon the environment, on the other hand it is abandoned or going to be abandoned in other Countries which did benefit of stable economic growth. Thorium is an emblem for such a situation: huge reserves are available all over the world (primarily India, Turkey, and Brazil, but not only) and its technological worth for exploitation in current generation of thermal fission reactors is demonstrated, on the other hand no industrial use is ongoing or planned for the near future (with an exception constituted by situation in India). Moreover, research on thorium utilization in nuclear reactors and associated fuel cycles has been of academic interest for many researchers around the world. These researches are being conducted to increase the natural resource utilization, reduces the radiotoxicity, and other criteria of sustainability, by using thorium in the present time advanced reactors (Generation III), as well for the future Generation IV, mainly in Molten Salt Reactors (MSR) and in hybrid fusion/ accelerators driven system. Here we are going to describe a MoU signed by the authors to promote the utilization of thorium as nuclear fuel, and shortly describe the research activities conducted by the MoU partners

    Intracranial V. cholerae Sialidase Protects against Excitotoxic Neurodegeneration

    Converging evidence shows that GD3 ganglioside is a critical effector in a number of apoptotic pathways, and GM1 ganglioside has neuroprotective and noötropic properties. Targeted deletion of GD3 synthase (GD3S) eliminates GD3 and increases GM1 levels. Primary neurons from GD3S−/− mice are resistant to neurotoxicity induced by amyloid-ÎČ or hyperhomocysteinemia, and when GD3S is eliminated in the APP/PSEN1 double-transgenic model of Alzheimer's disease the plaque-associated oxidative stress and inflammatory response are absent. To date, no small-molecule inhibitor of GD3S exists. In the present study we used sialidase from Vibrio cholerae (VCS) to produce a brain ganglioside profile that approximates that of GD3S deletion. VCS hydrolyzes GD1a and complex b-series gangliosides to GM1, and the apoptogenic GD3 is degraded. VCS was infused by osmotic minipump into the dorsal third ventricle in mice over a 4-week period. Sensorimotor behaviors, anxiety, and cognition were unaffected in VCS-treated mice. To determine whether VCS was neuroprotective in vivo, we injected kainic acid on the 25th day of infusion to induce status epilepticus. Kainic acid induced a robust lesion of the CA3 hippocampal subfield in aCSF-treated controls. In contrast, all hippocampal regions in VCS-treated mice were largely intact. VCS did not protect against seizures. These results demonstrate that strategic degradation of complex gangliosides and GD3 can be used to achieve neuroprotection without adversely affecting behavior

    E-Cadherin Downregulation is Mediated by Promoter Methylation in Canine Prostate Cancer

    E-cadherin is a transmembrane glycoprotein responsible for cell-to-cell adhesion, and its loss has been associated with metastasis development. Although E-cadherin downregulation was previously reported in canine prostate cancer (PC), the mechanism involved in this process is unclear. It is well established that dogs, besides humans, spontaneously develop PC with high frequency; therefore, canine PC is an interesting model to study human PC. In human PC, CDH1 methylation has been associated with E-cadherin downregulation. However, no previous studies have described the methylation pattern of CDH1 promoter in canine PC. Herein, we evaluated the E-cadherin protein and gene expression in canine PC compared to normal tissues. DNA methylation pattern was investigated as a regulatory mechanism of CDH1 silencing. Our cohort is composed of 20 normal prostates, 20 proliferative inflammatory atrophy (PIA) lesions, 20 PC, and 11 metastases from 60 dogs. The E-cadherin protein expression was assessed by immunohistochemistry and western blotting and gene expression by qPCR. Bisulfite- pyrosequencing assay was performed to investigate the CDH1 promoter methylation pattern. Membranous E-cadherin expression was observed in all prostatic tissues. A higher number of E-cadherin negative cells was detected more frequently in PC compared to normal and PIA samples. High-grade PC showed a diffuse membranous positive immunostaining. Furthermore, PC patients with a higher number of E-cadherin negative cells presented shorter survival time and higher Gleason scores. Western blotting and qPCR assays confirmed the immunohistochemical results, showing lower E-cadherin protein and gene expression levels in PC compared to normal samples. We identified CDH1 promoter hypermethylation in PIA and PC samples. An in vitro assay with two canine prostate cancer cells (PC1 and PC2 cell lines) was performed to confirm the methylation as a regulatory mechanism of E-cadherin expression. PC1 cell line presented CDH1 hypermethylation and after 5-Aza-dC treatment, a decreased CDH1 methylation and increased gene expression levels were observed. Positive E-cadherin cells were massively found in metastases (mean of 90.6%). In conclusion, low levels of E-cadherin protein, gene downregulation and CDH1 hypermethylation was detected in canine PC. However, in metastatic foci occur E-cadherin re-expression confirming its relevance in these processes

    Determination of Organochlorines in Soil of a Suburban Area of SĂŁo Paulo Brazil

    Technological advances have promoted improvements in several science fields, especially related to environmental and analytical areas with the improvement of detection and development of environmentally friendly extraction techniques. This study applied Quick, Easy, Cheap, Effective, Rugged and Safe method (QuEChERS) for soil extraction and assessed its performance through a validation study using samples from the soil of a contaminated area in Caieiras, SP, Brazil. Nine organochlorine pesticides, including the isomers alpha, beta, gamma and delta- hexachlorocyclohexane; cis- and trans-heptachlor epoxide; cis- and trans-chlordane and heptachlor were analyzed by gas chromatography coupled to electron capture detector. The method was validated according to ISO 5725-4 (2020), EURACHEM (2014) and DOQ-CGCRE-008 (2016). The limits of detection and quantification of the method for the nine organochlorines were α-HCH (1.2 and 12.6 µg kg−1), β-HCH (1.7 and 12.0 µg kg−1), γ-HCH (1.5 and 11.6 µg kg−1), δ-HCH (0.8 and 11.6 µg kg−1), heptachlor (1.0 and 10.8 µg kg−1), cis-heptachlor epoxide (0.9 and 11.5 µg kg−1), trans-heptachlor epoxide (0.9 and 11.5 µg kg−1), cis-chlordane (0.4 and 7.9 µg kg−1) and trans-chlordane (0.5 and 10.9 µg kg−1), respectively, and all of them were within the maximum limits recommended by the EPA for the compounds α-HCH (86.0 and 360.0 µg kg−1), β-HCH (300.0 and 1.3 × 103 µg kg−1), γ-HCH (570.0 and 2.5 × 103 µg kg−1), δ-HCH (not defined), heptachlor (130.0 and 630.0 µg kg−1), cis-/trans-heptachlor epoxide (7.0 and 330.0 µg kg−1), cis-/trans-chlordane (1.77 × 103 and 7.7 × 103 µg kg−1) in residential and industrial soil, respectively. Recovery results were between 65% and 105% for almost all compounds, which is an optimum result for multi-residue analytical methods, considering the complexity of the matrix used in the study. Caieiras presented contamination levels of α-HCH in the range of 2.0 to 66.0 µg g−1, which was higher than the limits established by EPA, corresponding to 0.077 µg g−1 for residential soil and 0.27 µg g−1 for industrial soil. According to the validation study, the analytical method proposed was reliable for organochlorine quantification, and the QuEChERS was considered efficient for organochlorine extraction from soil

    A utilização de fitoterapia no cotidiano de uma população rural [The phytoterapy utilization in the rural population routine]

    Esta pesquisa tem como objetivos estudar o uso da fitoterapia, em uma população da zona rural no interior de MG; identificar as plantas utilizadas com maior freqĂŒĂȘncia e formas de uso e comparar os seus efeitos com aqueles descritos na literatura. Foram entrevistadas 33 pessoas em 1997. Em relação Ă s plantas fitoterĂĄpicas conhecidas pelos entrevistados foram citadas 106, sendo 60 efetivamente utilizadas. A transmissĂŁo do conhecimento referente ao seu uso ocorreu basicamente dos avĂłs para os filhos e netos. A fitoterapia foi utilizada com fins semelhantes aos descritos na literatura, visando Ă  prevenção e tratamento das doenças.<br>El objetivo de esta pesquisa es estudiar lo empleo de la fitoterapia en una poblaciĂłn de la ĂĄrea rural en lo interior de Minas Gerais, identificar las plantas utilizadas con mayor frecuencia e comparar los resultados con lo descrito en la literatura. Fueran entrevistadas 33 personas en 1997. Fueran citadas 106 plantas conocidas por los entrevistados y 60 que son efectivamente utilizadas. La transmisiĂłn de los conocimientos referente a lo uso paso de los abuelos para los hijos y nietos. La fitoterapia fue usada para finalidad semejante a de la literatura, con la finalidad de prevenciĂłn y tratamiento de las enfermedades.<br>The aims of this study are to analyze the use of phytoterapy and the context of its utilization in a rural areapopulation - MG; to identify the herbs used with more frequency and their forms of use; to compare the results of medicinal herbs with that described in literature. Thirty three people were interviewed in 1997. About the phytoterapy herbs known by them 106 different species were quoted, and 60 of them were effectively used in their routine. The transmission of knowledge related to the use of the herbs occurred basically from grandparents to the children. The phytoterapy was used by the population studied with similar purposes described in the literature aiming the prevention and treatment of diseases

    Check-list of the Family Lamiaceae in Pernambuco, Brazil

    A survey of the family Lamiaceae in the state of Pernambuco revealed the presence of 20 genera encompassing 46 species. The genera with the highest number of exsiccatae were Hyptis and Ocimum; others had little representative material, and some taxa were collected only once. Regarding the phytogeographical distribution in the state, 9.1% of the registrations of species occurred in the coastal zone, 54.5% in the forest zone and 36.3% in the caatinga zone.<br>O levantamento da famĂ­lia Lamiaceae no estado de Pernambuco revelou a existĂȘncia de 20 gĂȘneros aos quais estĂŁo subordinadas 46 espĂ©cies. Os gĂȘneros com maior nĂșmero de exsicatas foram Hyptis e Ocimum, outros tiveram poucos materiais representativos, existindo aqueles coletados uma Ășnica vez. Com relação a distribuição fitogeogrĂĄfica no estado, 9,1% dos registros de espĂ©cies sĂŁo da Zona do Litoral, 54,5% da Zona da Mata e 36,3% da Zona da Caatinga
