New carbohydrate derivatives as tools to bind and metabolically label strains of the Mycobacterium tuberculosis complex

Tuberculosis (Tb), caused by Mycobacterium tuberculosis (Mtb), is the single leading bacterial cause of death from infectious diseases worldwide. Tb therapy is complicated by an increase in antibiotic resistant Mtb strains and the lack of fast and specific diagnostic tools. The cell envelope of Mtb bacteria represents a major virulence factor and contributes to the intrinsic difficulties of eradicating the pathogen. However, the uniqueness of the Mtb cell envelope can also be seen as an advantage and could bear the chance to develop novel tuberculosis-specific diagnostic and therapeutic tools. In this thesis the unique features of the Mtb cell envelope were utilized to enrich mycobacteria from solution and to metabolically label different Mtb clinical isolates.Tuberkulose (Tb), hervorgerufen durch das Mycobacterium tuberculosis (Mtb), ist weltweit die am häufigsten zum Tode führende bakterielle Infektionskrankheit. Erschwert wird die Therapie dieser Erkrankung durch das zunehmende Auftreten antibiotikaresistenter Mtb Stämme und dem Mangel an Testsystemen, die eine schnelle und spezifische Diagnose der Tb ermöglichen. Die Zellhülle der Mtb-Bakterien trägt dabei wesentlich zur Virulenz des Erregers bei und stellt bislang ein besonderes Hindernis bei der Tb-Bekämpfung dar. Die Einzigartigkeit dieser mykobakteriellen Zellhülle kann jedoch auch als Vorteil verstanden werden und dazu beitragen, neue Methoden für die spezifische Diagnostik und gezielte Therapie von Tb zu entwickeln. In der hier vorliegenden Arbeit wurden die besonderen Eigenschaften der Mtb Zellhülle gezielt ausgenutzt, um Mykobakterien aus einer Lösung anzureichern und verschiedene klinische Mtb Isolate metabolisch zu markieren

Berichte aus dem Ökolandbau 2021 - Dauerversuch zur organischen und mineralischen Grunddüngung: Einfluss steigender organischer sowie mineralischer P- und K-Düngung auf Merkmale der Bodenfruchtbarkeit, Ertrag und Qualität der Fruchtarten in einem ökologischen Dauerversuch auf Lehmboden

Als Ziel der Veröffentlichung wird eine Gesamtauswertung der Ergebnisse eines Dauerversuches zur Grunddüngung vorgenommen. Eine steigende organische Düngung von 0 – 160 kg N/ha führte zu einer Zunahme der GE-Erträge der Fruchtfolge auf 107 – 141 %. Trotz sehr niedriger P-Versorgung des Bodens wurde je nach Fruchtart nur eine geringe Ertragswirkung zwischen 101 – 104 % in Folge steigender Rohphosphatdüngung mit Dolophos (0 – 60 kg P/ha) nachgewiesen. Durch eine mineralische K-Düngung von 0 – 160 kg K/ha kam es zu einer Erhöhung der GE-Erträge der Fruchtarten um bis zu 125 % im Vergleich zu keiner Düngung (= 100 %). Im Mittelpunkt der Auswertungen standen sowohl die erzielten Erträge und Qualitäten der Haupt- und Nebenprodukte der Fruchtarten als auch die zeitlichen Veränderungen der verschiedenen Bodenparameter in der Ackerkrume und im Untergrund in Abhängigkeit von den jeweiligen Prüffaktoren. Darüber hinaus wurde eine Gewichtung der erhaltenen Ergebnisse zur P- und K-Versorgung des Bodens an einem überregionalen Verbund an Versuchen aus 8 Bundesländern vorgenommen. Zur allgemeinen Bewertung der Wirkung der organischen Düngung auf die Erträge der Fruchtarten kam erstmals ein System auf Basis der N-Bilanzierung zum Einsatz. Im ersten Teil des Heftes wird der Einfluss von Stroh- und Gründüngung auf den Ertrag und die Qualität von Öko-Hafer untersucht. 3-jährige Feldversuche zeigen, dass Strohdüngung einen leichten, nicht signifikanten Ertragsrückgang bei Hafer zur Folge hat. Gründüngung dagegen führt sowohl mit Leguminosen als auch mit Gras zu deutlichen Mehrerträgen. In einem weiteren Beitrag werden 10 Jahre Ökolandbau im Lehr- und Versuchsgut Köllitsch ausgewertet. Dabei zeigt sich, dass trotz negativer Nährstoffbilanzen für N, P, K und Mg lediglich bei K eine Unterversorgung messbar wird. Die Erträge der angebauten Kulturen spiegeln bislang noch keinen K-Mangel wider. Die Humusbilanz der Fruchtfolge ist trotz unterbliebener organischer Düngung positiv. Identification and characterization of the BRI2 interactome in the brain

BRI family proteins are ubiquitous type II transmembrane proteins but BRI2 is highly expressed in some neuronal tissues. Possible BRI2 functions include neuronal maturation and differentiation. Protein complexes appear to be important in mediating its functions. Previously described BRI2 interactors include the Alzheimer's amyloid precursor protein and protein phosphatase 1, but clearly the identification of novel interactors provides an important tool to understand the role and function of BRI2. To this end three rat brain regions (cerebellum, hippocampus, and cerebral cortex) were processed by BRI2 immunoprecipitation;co-precipitating proteins were identified by Nano-HPLC-MS/MS. The pool of the brain regions resulted in 511 BRI2 interacting proteins (BRI2 brain interactome) of which 120 were brain specific and 49 involved in neuronal differentiation. Brain region-specific analyses were also carried out for cerebellum, hippocampus, and cerebral cortex. Several novel BRI2 interactors were identified among them DLG4/PSD-95, which is singularly important as it places BRI2 in the postsynaptic compartment. This interaction was validated as well as the interaction with GAP-43 and synaptophysin. In essence, the resulting BRI2 brain interactome, associates this protein with neurite outgrowth and neuronal differentiation, as well as synaptic signalling and plasticity. It follows that further studies should address BRI2 particularly given its relevance to neuropathological conditions

TIRANA. Architecture as Political Actor

This book on the architecture of Tirana contains findings of the interdisciplinary seminar “TIRANA. Architecture as Political Actor” at Bauhaus-Universität Weimar. In photographs, texts and diagrams it shows the approach to an unknown city that was explored following the ideas of the Actor Network Theory (ANT). Thus, the book gives an insight into scientific as well as artistic works, both mirroring the attempt to grasp the role of architecture within political processes in the 20th century and today. In this compilation of the architectural-political networks, an image of the city of Tirana emerges that gives an idea of specific built structures as well as of the architecture as political actor on a meta-level. In doing so, the book itself becomes an actor in the discussion of the relationship of architecture and politics in Albania and an example for the use of ANT as scientific-artistic tool for the research on architectural “things” in the context of a city

Der jährlich erscheinende Newsletter berichtet über Aktivitäten rund um Service User Involvement an Hochschulen im deutschsprachigen Raum. Sie finden in dieser Ausgabe folgende Beiträge: Zu Beginn berichten Bärbel Haag, Thomas Heidenreich und Bettina Müller von der Hochschule Esslingen über die neue Zusammenarbeit mit Bildungsfachkräften, die z. B. Input zu Inklusions- und Exklusionserfahrungen von Menschen mit Behinderungen teilgeben. Daran anschließen kann David Dörrer, der uns über das Annelie-Wellensiek-Zentrum für Inklusive Bildung an der Pädagogischen Hochschule Heidelberginformiert, an der Menschen mit und ohne Behinderungserfahrungen gemein-sam zu Querschnittsaufgaben der Inklusion forschen und lehren. Es folgt ein Bericht und Plädoyer aus Berlin von Reinhard Burtscher und Anne Schöbel über Lerner-Expert:innen, die durch ihre Erfahrungsberichte dem Thema „Alphabetisierung und Grundbildung“ ein Gesicht geben. Katharina Lutz und Benjamin Benz machen uns auf den Sozial-Wissenschaftsladen aufmerksam, der das gemeinsame Forschen für zivilgesellschaftliche Vertreter_innen, Praktiker_innen und Hochschulangehörige in Lehrforschungsprojekten und im Rahmen von Bachelor- und Masterarbeiten ermöglicht. Forschend geht es auch weiter mit Katharina Scholz, die sich mit Lehr-Evaluation für Seminare mit Beteiligung von Adressat*innen befasst und von neuen Vorhaben aus der HAW Hamburg berichtet. Kornelia Birkemeyer stellt das Projekt EX-IN vor, berichtet als Genesungsbegleiterin über die Kooperation mit der Hochschule Esslingen und liefert Antworten auf die Frage: „Warum brauchen Studierende von sozialer Arbeit die Hilfe von Service Usern?“. Um Gesungsbegleiter*innen als Teil eines trialogischen Konzeptes geht es auch bei der interaktiven Online-Plattform TriNetz, die von Christel Baatz-Kolbe und Christoph Walther vorgestellt wird. Die Plattform hat eine Lotsenfunktion und vermittelt zwischen Interessierten, Ausbildungsstätten und potentiellen Arbeitgebern. Bei Birgit Mayrhofer und Laura Ressler geht es gleichermaßen um die Peer-Arbeit von Genesungsbegleiter*innen. Sie stellen ihr Masterforschungsprojekt vor, das sich mit der Rollenkonstruktion der Peer-Arbeit befasst. Die Peer-Begleiterin Barbara Kohlmann vom Projekt „Upsides“ verabschiedet uns mit ihren Gedanken zu Peer-Arbeit in Zeiten der Covid-19-Pandemie. Qualitätsmanagement in Zahnarztpraxen - Eine qualitative Interviewstudie mit Hamburger Zahnärzten zur kritischen Reflexion der Qualitätsmanagement-Richtlinie

Mit einer zunehmenden Qualitätsorientierung wird den Herausforderungen des Gesundheitswesens und den sich verändernden Erwartungen der Patienten begegnet. Zahnarztpraxen haben seit dem 1. Januar 2011 verpflichtend ein einrichtungsinternes Qualitätsmanagement umzusetzen. Die Mindestanforderungen sind in der Qualitätsmanagement-Richtlinie des Gemeinsamen Bundesausschusses festgeschrieben. Das Ziel dieser Untersuchung ist es, herauszufinden, welche Qualitätsvor-stellungen bei den befragten Zahnärzten vorliegen und welche Anforderungen der Richtlinie als angemessen und praktikabel angesehen werden. Mittels qualitativer Leitfadeninterviews wurden die interessierenden Fragestellungen thematisiert. Die Leitfadenentwicklung basiert auf einem Phasensystem nach Schirmer. Die Stichprobenauswahl erfolgte durch telefonischen Erstkontakt, die Interviews wurden mittels Tonband aufgezeichnet und anschließend transkribiert. Das Auswertungsverfahren erfolgte durch eine qualitative Inhaltsanalyse nach Mayring. Es wurde eine Response-Rate von ca. acht Prozent erzielt, sodass fünf Interviews mit Hamburger Zahnärzten durchgeführt wurden. Unter Qualitätsmanagement wird ein Strukturieren der Arbeitsabläufe verstanden. Als weitere Qualitätskriterien wurden Patientenzufriedenheit, Schmerzfreiheit des Patienten, das Einführen eines Fehlermanagements und die Kontrolle zahntechnischer Produkte genannt. Die Zahnärzte zeigten sich insgesamt mit der Richtlinie gut vertraut. Der Konkretisierungsgrad der QM-Richtlinie wurde unter-schiedlich eingeschätzt. Die Richtlinie strebt eine Verbesserung der Patientenversorgung sowie der Praxisorganisation an. Seitens der befragten Zahnärzte wurden die angedachte Patientenorientierung und eine stete Qualitätsverbesserung nicht als zentrales Ziel erkannt. Die Richtlinie bezieht sich auf die Arbeitsabläufe, jedoch nicht primär auf eine Erhöhung der Behand-lungsqualität. Der Rollenkonflikt eines Zahnarztes, zwischen medizinischer Indikation und monetären Aspekten entscheiden zu müssen, wird in der Richtlinie nicht thematisiert. Es bedarf eines erweiterten Qualitätsbegriffes, um den Kostenaspekt in der Zahnmedizin zu berücksichtigen. Weiterhin sollten Qualitätsindikatoren wie die Eigenverantwortung des Patienten oder auch die partizipative Entscheidungsfindung mit dem Patienten in den Qualitätsbegriff integriert werden.The challenges in the health care sector are counteracted with an increasing orientation and focus on quality. Dental surgeries need to have implemented an in-house quality management since January 1st, 2011. The minimum requirements are specified in the quality-directive of the Federal Joint Committee. The aim of this investigation is to find out, which associations to quality do exist for the interviewed dentists and which requirements of the quality-directive are seen as appropriate and practicable. Therefore, qualitative guided interviews will be employed as the instrument of investiga-tion for making interested aspects the subjects of discussion. The development of guided interview is based on a system of phases by Schirmer. By telephone dental surgeries got contacted in order to get a probability sampling. The interviews were digitally recorded and transliterated afterwards. As a method of evaluation, a qualitative content analysis by Mayring was used in this investigation. A response-rate by eight percent was achieved. Hence five interviews with dentists from Hamburg were conducted. Quality management is seen to be the structuring of working processes. Concerning further quality criteria, the dentists cited patient satisfaction, pa-tients being free from pain, implementing a control system for nonconforming treatment and a control of dental technology products. The dentists were well-informed about the quality-directive, whereas the concretion was assessed differently. The quality-directive aims at an improvement in patient care and in the organization of dental surgeries. On the part of the dentists, the considered patient orientation and con-tinuous quality improvement were not named as the central aims of the directive. Fur-thermore, the quality-directive applies to working processes and not primarily to an im-provement of quality in treatment. The dentists’ role conflict, to differentiate in his decision between indication and monetary aspects, is not addressed at all. One can recognize the need for an expansion of the quality definition in dental medicine in order to consider the relatedness to costs of dental products. Still quality indicators, by way of example the em-powerment of patients or a shared decision-making, need to get integrated in definitions of quality
