    Communities in university mathematics

    This paper concerns communities of learners and teachers that are formed, develop and interact in university mathematics environments through the theoretical lens of Communities of Practice. From this perspective, learning is described as a process of participation and reification in a community in which individuals belong and form their identity through engagement, imagination and alignment. In addition, when inquiry is considered as a fundamental mode of participation, through critical alignment, the community becomes a Community of Inquiry. We discuss these theoretical underpinnings with examples of their application in research in university mathematics education and, in more detail, in two Research Cases which focus on mathematics students' and teachers' perspectives on proof and on engineering students' conceptual understanding of mathematics. The paper concludes with a critical reflection on the theorising of the role of communities in university level teaching and learning and a consideration of ways forward for future research

    Properties of low-alloy high-speed steel at elevated temperature

    This paper presents the results of research on the determination of the coefficient of thermal conductivity and hot hardness of cutters made of selected grades of low-alloy high-speed steels, HS 6-5-2 and HS3-1-2. The investigations of hot hardness and yield stress values of HS6-5-2 steel at elevated temperatures have shown that the hot hardness value decreased to 650 – 700 HV (59 – 60 HRC) in the temperature range of 500 – 550 °C. However, the hardness of the samples preheated to the temperature of 500 – 550 °C and measured at room temperature does not change. A decrease of the hot hardness of the steel is correlated with decreasing yield stress at elevated temperature

    Family studies of somatic and functional characteristics in the polish rural population

    In the present investigation we were trying to determine the genetic and environmental conditioning of the chosen somatic and functional traits in Polish rural population during ontogenesis. In order to find out interactions between environmental and genetic conditions of the studied traits, classical methods of quantitative features were applied: correlation coefficients corrected by assortative mating in the chosen types of heritability were evaluated on their base, heritability coefficients of analyzed features were assessed. The biggest stability of the correlation coefficients was observed for the length-parameters. We did not noticed stronger genetic control of functional features in men. Mean-strong genetic control among analyzed traits was observed in: reaction time, space orientation and static strength expressed as relative and absolute strength

    Appropriately differentiated ARPE-19 cells regain phenotype and gene expression profiles similar to those of native RPE cells.

    PurposeThe RPE cell line ARPE-19 provides a dependable and widely used alternative to native RPE. However, replication of the native RPE phenotype becomes more difficult because these cells lose their specialized phenotype after multiple passages. Compounding this problem is the widespread use of ARPE-19 cells in an undifferentiated state to attempt to model RPE functions. We wished to determine whether suitable culture conditions and differentiation could restore the RPE-appropriate expression of genes and proteins to ARPE-19, along with a functional and morphological phenotype resembling native RPE. We compared the transcriptome of ARPE-19 cells kept in long-term culture with those of primary and other human RPE cells to assess the former's inherent plasticity relative to the latter.MethodsARPE-19 cells at passages 9 to 12 grown in DMEM containing high glucose and pyruvate with 1% fetal bovine serum were differentiated for up to 4 months. Immunocytochemistry was performed on ARPE-19 cells grown on filters. Total RNA extracted from ARPE-19 cells cultured for either 4 days or 4 months was used for RNA sequencing (RNA-Seq) analysis using a 2 × 50 bp paired end protocol. The RNA-Seq data were analyzed to identify the affected pathways and recognize shared ontological classification among differentially expressed genes. RPE-specific mRNAs and miRNAs were assessed with quantitative real-time (RT)-PCR, and proteins with western blotting.ResultsARPE-19 cells grown for 4 months developed the classic native RPE phenotype with heavy pigmentation. RPE-expressed genes, including RPE65, RDH5, and RDH10, as well as miR-204/211, were greatly increased in the ARPE-19 cells maintained at confluence for 4 months. The RNA-Seq analysis provided a comprehensive view of the relative abundance and differential expression of the genes in the differentiated ARPE-19 cells. Of the 16,757 genes with detectable signals, nearly 1,681 genes were upregulated, and 1,629 genes were downregulated with a fold change of 2.5 or more differences between 4 months and 4 days of culture. Gene Ontology analysis showed that the upregulated genes were associated with visual cycle, phagocytosis, pigment synthesis, cell differentiation, and RPE-related transcription factors. The majority of the downregulated genes play a role in cell cycle and proliferation.ConclusionsThe ARPE-19 cells cultured for 4 months developed a phenotype characteristic of native RPE and expressed proteins, mRNAs, and miRNAs characteristic of the RPE. Comparison of the ARPE-19 RNA-Seq data set with that of primary human fetal RPE, embryonic stem cell-derived RPE, and native RPE revealed an important overall similar expression ratio among all the models and native tissue. However, none of the cultured models reached the absolute values in the native tissue. The results of this study demonstrate that low-passage ARPE-19 cells can express genes specific to native human RPE cells when appropriately cultured and differentiated

    This paper presents the software for comprehensive processing and visualization of 2D and 3D electrical tomography data. The system name as TomoKIS Studio has been developed in the frame of DENIDIA international research project and has been improved in the frame of Polish Ministry of Science and Higher Education Project no 4664/B/T02/2010/38. This software is worldwide unique because it simultaneously integrates the process of tomographic data acquisition, numerical FEM modeling and tomographic images reconstruction. The software can be adapted to specific industrial applications, particularly to monitoring and diagnosis of two-phase flows. The software architecture is composed of independent modules. Their combination offers calibration, configuration and full-duplex communication with any tomographic acquisition system with known and open communication protocol. The other major features are: online data acquisition and processing, online and offline 2D/3D images linear and nonlinear reconstruction and visualization as well as raw data and tomograms processing. Another important ability is 2D/3D ECT sensor construction using FEM modeling. The presented software is supported with the multi-core GPU technology and parallel computing using Nvidia CUDA technology.W artykule autorzy przedstawiają środowisko komputerowe do kompleksowego przetwarzania i wizualizacji tomograficznych danych pomiarowych. Oprogramowanie  TomoKIS Studio powstało w Instytucie Informatyki Stosowanej PŁ w ramach projektu DENIDIA i zostało rozwinięte w ramach projektu MNiSW nr 4664/B/T02/2010/38. Zbudowane oprogramowanie jest unikalne w skali światowej, gdyż integruje w sobie proces pozyskiwania danych pomiarowych, modelowanie numeryczne oraz proces konstruowania obrazów tomograficznych, z możliwością adaptacji dla różnych aplikacji przemysłowych, w szczególności dla potrzeb monitorowania i diagnostyki przepływów dwufazowych gaz-ciecz. Architektura aplikacji oparta jest na zestawie niezależnych modułów, które pozwalają na w pełni dwukierunkową komunikacją, konfigurację oraz kalibrację dowolnego urządzenia tomografii elektrycznej z otwartym protokołem pomiarowym, akwizycję i przetwarzanie danych pomiarowych on-line, liniową oraz nieliniową rekonstrukcję obrazów 2D i 3D w czasie rzeczywistym, a także wizualizację surowych danych pomiarowych i tomogramów. Istotnym elementem systemu jest moduł numerycznego modelowania czujników pojemnościowych wykorzystujący metodę elementów skończonych, oparty na autorskich algorytmach generowania siatek MES komputerowych modeli czujników pojemnościowych. Architektura prezentowanego systemu została zaprojektowana przy użyciu obliczeń równoległych na procesorach graficznych, z wykorzystaniem technologii Nvidia CUDA

    Assembly of Helicobacter pylori initiation complex is determined by sequence-specific and topology-sensitive DnaA-oriC interactions

    In bacteria, chromosome replication is initiated by binding of the DnaA initiator protein to DnaA boxes located in the origin of chromosomal replication (oriC). This leads to DNA helix opening within the DNA-unwinding element. Helicobacter pylori oriC, the first bipartite origin identified in Gram-negative bacteria, contains two subregions, oriC1 and oriC2, flanking the dnaA gene. The DNA-unwinding element region is localized in the oriC2 subregion downstream of dnaA. Surprisingly, oriC2–DnaA interactions were shown to depend on DNA topology, which is unusual in bacteria but is similar to initiator–origin interactions observed in higher organisms. In this work, we identified three DnaA boxes in the oriC2 subregion, two of which were bound only as supercoiled DNA. We found that all three DnaA boxes play important roles in orisome assembly and subsequent DNA unwinding, but different functions can be assigned to individual boxes. This suggests that the H. pylori oriC may be functionally divided, similar to what was described recently for Escherichia coli oriC. On the basis of these results, we propose a model of initiation complex formation in H. pylori

    Long Josephson junctions with spatially inhomogeneous driving

    The phase dynamics of a long Josephson junction with spatially inhomogeneously distributed bias current is considered for the case of a dense soliton chain (regime of the Flux Flow oscillator). To derive the analytical solution of the corresponding sine-Gordon equation the Poincare method has been used. In the range of the validity of the theory good coincidence between analytically derived and numerically computed current-voltage characteristics have been demonstrated for the simplest example of unitstep function distribution of bias current (unbiased tail). It is shown, that for the considered example of bias current distribution, there is an optimal length of unbiased tail that maximizes the amplitude of the main harmonic and minimizes the dynamical resistance (thus leading to reduction of a linewidth).Comment: 7 pages, 5 figure

    Effect of Grinding Parameters on the Surface Quality of Cutting Tools Made of High-Speed Low-Alloy Steels

    The occurrence of defects caused by sharpening leads to the considerable variation of lifetime of cutting tools under identical process conditions. The loss of cutting ability of cutting tools or the change of the blade original geometry influences the quality of the surface finish as well as both dimensional and shape accuracy of workpieces. The effect of the grinding parameters on the surface finish of selected high-speed steels has been investigated. The influence of the grinding parameters has been defined especially for surface roughness, grinding forces and grinding ratio with a wide range of grinding parameters. It is found that the value of average roughness (Ra ) parameter decreases together with an increase in the grinding depth and the feed length. Furthermore, the value of Ra increases proportionally to the grinding depth. To increase the efficiency of sharpening, it is required to increase the feed length other than the grinding depth
