A Twins Study of Communicative Adaptability: Heritability of Individual Differences
Recently, a model of communication theory and research has appeared in the literature within which stable individual differences in communication behavior represent individual differences in activation thresholds of neurobiological systems. The neurobiological systems thought to underly communication traits and behavior are assumed to be primarily due to genetic inheritance. As such, the model assigns a limited role to adaptability in social situations, instead positing communication adaptability as an inherited trait. In the present study, heritability estimates for the dimensions of communicative adaptability were derived from correlations based on identical and fraternal twins\u27 responses to a multidimensional communicative adaptability measure. Results indicated that social composure was 88% heritable, wit was 90% heritable, social confirmation was 37% heritable, articulation ability, and appropriate disclosure were 0% heritable. Theoretical implications are discussed
Heat Shock Protein-90 Inhibitors Enhance Antigen Expression on Melanomas and Increase T Cell Recognition of Tumor Cells
In an effort to enhance antigen-specific T cell recognition of cancer cells, we have examined numerous modulators of antigen-expression. In this report we demonstrate that twelve different Hsp90 inhibitors (iHsp90) share the ability to increase the expression of differentiation antigens and MHC Class I antigens. These iHsp90 are active in several molecular and cellular assays on a series of tumor cell lines, including eleven human melanomas, a murine B16 melanoma, and two human glioma-derived cell lines. Intra-cytoplasmic antibody staining showed that all of the tested iHsp90 increased expression of the melanocyte differentiation antigens Melan-A/MART-1, gp100, and TRP-2, as well as MHC Class I. The gliomas showed enhanced gp100 and MHC staining. Quantitative analysis of mRNA levels showed a parallel increase in message transcription, and a reporter assay shows induction of promoter activity for Melan-A/MART-1 gene. In addition, iHsp90 increased recognition of tumor cells by T cells specific for Melan-A/MART-1. In contrast to direct Hsp90 client proteins, the increased levels of full-length differentiation antigens that result from iHsp90 treatment are most likely the result of transcriptional activation of their encoding genes. In combination, these results suggest that iHsp90 improve recognition of tumor cells by T cells specific for a melanoma-associated antigen as a result of increasing the expressed intracellular antigen pool available for processing and presentation by MHC Class I, along with increased levels of MHC Class I itself. As these Hsp90 inhibitors do not interfere with T cell function, they could have potential for use in immunotherapy of cancer
Socio-cognitive constraints and opportunities for sustainable intensification in South Asia:Insights from fuzzy cognitive mapping in coastal Bangladesh
Appreciating and dealing with the plurality of farmers’ perceptions and their contextual knowledge and perspectives of the functioning and performance of their agroecosystems—in other words, their ‘mental models’—is central for appropriate and sustainable agricultural development. In this respect, the sustainable development goals (SDGs) aim to eradicate poverty and food insecurity by 2030 by envisioning social inclusivity that incorporates the preferences and knowledge of key stakeholders, including farmers. Agricultural development interventions and policies directed at sustainable intensification (SI), however, do not sufficiently account for farmers’ perceptions, beliefs, priorities, or interests. Considering two contrasting agroecological systems in coastal Bangladesh, we used a fuzzy cognitive mapping (FCM)-based simulation and sensitivity analysis of mental models of respondents of different farm types from 240 farm households. The employed FCM mental models were able to (1) capture farmers’ perception of farming system concepts and relationships for each farm type and (2) assess the impact of external interventions (drivers) on cropping intensification and food security. We decomposed the FCM models’ variance into the first-order sensitivity index (SVI) and total sensitivity index (TSI) using a winding stairs algorithm. Both within and outside polder areas, the highest TSIs (35–68%) were observed for effects of agricultural extension on changes in other concepts in the map, particularly food security and income (SI indicators), indicating the importance of extension programs for SI. Outside polders, drainage and micro-credit were also influential; within polders, the availability of micro-credit appears to affect farmer perceptions of SI indicators more than drainage. This study demonstrated the importance of reflection on the differing perspectives of farmers both within and outside polders to identify entry points for development interventions. In addition, the study underscores the need for micro-farming systems-level research to assess the context-based feasibility of introduced interventions as perceived by farmers of different farm types.</p

Preanalytical stability of plasma biomarkers for Alzheimer's disease pathology
Introduction
Plasma tests have demonstrated high diagnostic accuracy for identifying Alzheimer's disease pathology. To facilitate the transition to clinical utility, we assessed whether plasma storage duration and temperature affect the biomarker concentrations.
Methods
Plasma samples from 13 participants were stored at +4°C and +18°C. Concentrations of six biomarkers were measured after 2, 4, 6, 8, 10, and 24 h by single molecule array assays.
Results
Phosphorylated tau 181 (p-tau181), phosphorylated tau 231 (p-tau231), neurofilament light (NfL), and glial fibrillary acidic protein (GFAP) concentrations were unchanged both when stored at +4°C and +18°C. Amyloid-β 40 (Aβ40) and amyloid-β 42 (Aβ42) concentrations were stable for 24 h at +4°C but declined when stored at +18°C for longer than 6 h. This decline did not affect the Aβ42/Aβ40 ratio.
Discussion
Plasma samples can be stored for 24 h at +4°C or +18°C and result in valid assay results for p-tau181, p-tau231, Aβ42/Aβ40 ratio, GFAP, and NfL.publishedVersio

Genome-wide copy number alterations in subtypes of invasive breast cancers in young white and African American women.
Genomic copy number alterations (CNA) are common in breast cancer. Identifying characteristic CNAs associated with specific breast cancer subtypes is a critical step in defining potential mechanisms of disease initiation and progression. We used genome-wide array comparative genomic hybridization to identify distinctive CNAs in breast cancer subtypes from 259 young (diagnosed with breast cancer at 40%) for TN breast tumors at 10q, 11p, 11q, 16q, 20p, and 20q. In addition, we report CNAs that differ in frequency between TN breast tumors of AA and CA women. This is of particular relevance because TN breast cancer is associated with higher mortality and young AA women have higher rates of TN breast tumors compared to CA women. These data support the possibility that higher overall frequency of genomic alteration events as well as specific focal CNAs in TN breast tumors might contribute in part to the poor breast cancer prognosis for young AA women
Bone turnover markers are correlated with total skeletal uptake of 99mTc-methylene diphosphonate (99mTc-MDP)
ABSTRACT: BACKGROUND: Skeletal uptake of 99mTc labelled methylene diphosphonate (99mTc-MDP) is used for producing images of pathological bone uptake due to its incorporation to the sites of active bone turnover. This study was done to validate bone turnover markers using total skeletal uptake (TSU) of 99mTc-MDP. METHODS: 22 postmenopausal women (52-80 years) volunteered to participate. Scintigraphy was performed by injecting 520 MBq of 99mTc-MDP and taking whole body images after 3 minutes, and 5 hours. TSU was calculated from these two images by taking into account the urinary loss and soft tissue uptake. Bone turnover markers used were bone specific alkaline phosphatase (S-Bone ALP), three different assays for serum osteocalcin (OC), tartrate resistant acid phosphatase 5b (S-TRACP5b), serum C-terminal cross-linked telopeptides of type I collagen (S-CTX-I) and three assays for urinary osteocalcin (U-OC). RESULTS: The median TSU of 99mTc-MDP was 23% of the administered activity. All bone turnover markers were significantly correlated with TSU with r-values from 0.52 (p = 0.013) to 0.90 (p < 0.001). The two resorption markers had numerically higher correlations (S-TRACP5b r = 0.90, S-CTX-I r = 0.80) than the formation markers (S-Total OC r = 0.72, S-Bone ALP r = 0.66), but the difference was not statistically significant. TSU did not correlate with age, weight, body mass index or bone mineral density. CONCLUSION: In conclusion, bone turnover markers are strongly correlated with total skeletal uptake of 99mTc-MDP. There were no significant differences in correlations for bone formation and resorption markers. This should be due to the coupling between formation and resorption
