    Saturation Mutagenesis of Lysine 12 Leads to the Identification of Derivatives of Nisin A with Enhanced Antimicrobial Activity

    peer-reviewedIt is becoming increasingly apparent that innovations from the “golden age” of antibiotics are becoming ineffective, resulting in a pressing need for novel therapeutics. The bacteriocin family of antimicrobial peptides has attracted much attention in recent years as a source of potential alternatives. The most intensively studied bacteriocin is nisin, a broad spectrum lantibiotic that inhibits Gram-positive bacteria including important food pathogens and clinically relevant antibiotic resistant bacteria. Nisin is gene-encoded and, as such, is amenable to peptide bioengineering, facilitating the generation of novel derivatives that can be screened for desirable properties. It was to this end that we used a site-saturation mutagenesis approach to create a bank of producers of nisin A derivatives that differ with respect to the identity of residue 12 (normally lysine; K12). A number of these producers exhibited enhanced bioactivity and the nisin A K12A producer was deemed of greatest interest.     Atypical Listeria innocua strains possess an intact LIPI-3

    peer-reviewedBackground: Listeria monocytogenes is a food-borne pathogen which is the causative agent of listeriosis and can be divided into three evolutionary lineages I, II and III. While all strains possess the well established virulence factors associated with the Listeria pathogenicity island I (LIPI-1), lineage I strains also possess an additional pathogenicity island designated LIPI-3 which encodes listeriolysin S (LLS), a post-translationally modified cytolytic peptide. Up until now, this pathogenicity island has been identified exclusively in a subset of lineage I isolates of the pathogen Listeria monocytogenes. Results: In total 64 L. innocua strains were screened for the presence of LIPI-3. Here we report the identification of an intact LIPI-3 in 11 isolates of L. innocua and the remnants of the cluster in several others. Significantly, we can reveal that placing the L. innocua lls genes under the control of a constitutive promoter results in a haemolytic phenotype, confirming that the cluster is capable of encoding a functional haemolysin. Conclusions: Although the presence of the LIPI-3 gene cluster is confined to lineage I isolates of L. monocytogenes, a corresponding gene cluster or its remnants have been identified in many L. innocua strains.This work was funded by the Enterprise Ireland Commercialisation fund, a programme which is co-financed by the EU through the ERDF.     New Zealand Research on Skills Formation and the 'Learning Organisation': Critical Analysis of the Case Study Methodology

    In recent years there has been continuing debate about what skills are needed for New Zealand's competitive business environment. Many commentators talk of a skills crisis, while some academics argue that the skills crisis is a myth. Recent research on workplace reform in seven 'leading edge' New Zealand companies highlights six crucial limitations of the debate. These are a focus on: i) individual skills rather than organisational culture, work design and employment relationships; ii) formal training rather than work-based learning; iii) the skills of school leavers rather than those of managers, supervisors and workers; and iv) technical, rather than organisational and team, skills. The debate also pays inadequate attention to v) how people learn in teams; and vi) the complexity of the current business and policy environment. In November 1993, N7JSR &D commenced a FRST -funded, three-year project on 'Economic Restructuring and Skills Formation' to address these limitations. Based on jive case studies in a range of sectors, the project examines characteristics of enterprises seeking sustainable competitive advantage by operating as a 'learning organisation'; one where the organisational structure itself is an environment for continuing education. Case study analysis is also used to understand the relationship between the company's corporate strategy and skilling process; the core skills required by managers, supervisors and workers, and how these are formed. The purpose of this paper is to outline the project's case study design, explain why this methodology has been selected and examine debates about its validity and utility

    1. The cost, usability and power efficiency of available wildlife monitoring equipment currently inhibits full ground-level coverage of many natural systems. Developments over the last decade in technology, open science, and the sharing economy promise to bring global access to more versatile and more affordable monitoring tools, to improve coverage for conservation researchers and managers. 2. Here we describe the development and proof-of-concept of a low-cost, small-sized and low-energy acoustic detector: 'AudioMoth'. The device is open-source and programmable, with diverse applications for recording animal calls or human activity at sample rates of up to 384kHz. We briefly outline two ongoing real-world case studies of large-scale, long-term monitoring for biodiversity and exploitation of natural resources. These studies demonstrate the potential for AudioMoth to enable a substantial shift away from passive continuous recording by individual devices, towards smart detection by networks of devices flooding large and inaccessible ecosystems. 3. The case studies demonstrate one of the smart capabilities of AudioMoth, to trigger event logging on the basis of classification algorithms that identify specific acoustic events. An algorithm to trigger recordings of the New Forest cicada (Cicadetta montana) demonstrates the potential for AudioMoth to vastly improve the spatial and temporal coverage of surveys for the presence of cryptic animals. An algorithm for logging gunshot events has potential to identify a shotgun blast in tropical rainforest at distances of up to 500 m, extending to 1km with continuous recording. 4. AudioMoth is more energy efficient than currently available passive acoustic monitoring (PAM) devices, giving it considerably greater portability and longevity in the field with smaller batteries. At a build cost of ~US$43 per unit, AudioMoth has potential for varied applications in large-scale, long-term acoustic surveys. With continuing developments in smart, energy-efficient algorithms and diminishing component costs, we are approaching the milestone of local communities being able to afford to remotely monitor their own natural resources
