    Involvement of heparanase in the pathogenesis of acute kidney injury: Nephroprotective effect of PG545

    Despite the high prevalence of acute kidney injury (AKI) and its association with increased morbidity and mortality, therapeutic approaches for AKI are disappointing. This is largely attributed to poor understanding of the pathogenesis of AKI. Heparanase, an endoglycosidase that cleaves heparan sulfate, is involved in extracellular matrix turnover, inflammation, kidney dysfunction, diabetes, fibrosis, angiogenesis and cancer progression. The current study examined the involvement of heparanase in the pathogenesis of ischemic reperfusion (I/R) AKI in a mouse model and the protective effect of PG545, a potent heparanase inhibitor. I/R induced tubular damage and elevation in serum creatinine and blood urea nitrogen to a higher extent in heparanase over-expressing transgenic mice vs. wild type mice. Moreover, TGF-\u3b2, vimentin, fibronectin and \u3b1-smooth muscle actin, biomarkers of fibrosis, and TNF\u3b1, IL6 and endothelin-1, biomarkers of inflammation, were upregulated in I/R induced AKI, primarily in heparanase transgenic mice, suggesting an adverse role of heparanase in the pathogenesis of AKI. Remarkably, pretreatment of mice with PG545 abolished kidney dysfunction and the up-regulation of heparanase, pro-inflammatory (i.e., IL-6) and pro-fibrotic (i.e., TGF-\u3b2) genes induced by I/R. The present study provides new insights into the involvement of heparanase in the pathogenesis of ischemic AKI.Our results demonstrate that heparanase plays a deleterious role in the development of renal injury and kidney dysfunction,attesting heparanase inhibition as a promising therapeutic approach for AKI

Yet, the extent to which cardiorespiratory fitness-related brain activation is associated with better cognitive performance is not well understood. In this cross-sectional study, we examined whether the association between cardiorespiratory fitness and executive function was mediated by greater prefrontal cortex activation in healthy older adults. Brain activation was measured during dual-task performance with functional magnetic resonance imaging in a sample of 128 healthy older adults (59-80 years). Higher cardiorespiratory fitness was associated with greater activation during dual-task processing in several brain areas including the anterior cingulate and supplementary motor cortex (ACC/SMA), thalamus and basal ganglia, right motor/somatosensory cortex and middle frontal gyrus, and left somatosensory cortex, controlling for age, sex, education, and gray matter volume. Of these regions, greater ACC/SMA activation mediated the association between cardiorespiratory fitness and dual-task performance. We provide novel evidence that cardiorespiratory fitness may support cognitive performance by facilitating brain activation in a core region critical for executive function

    Trade-offs between security and inspection capacity - Policy options for land border ports of entry

    Security 2006: Transportation Research Record, 1942: pp. 16-22.Observations of primary inspections at land border ports of entry between Ciudad Juarez, Mexico, and El Paso, Texas, indicate that the majority of inspections are of limited depth. 79% of primary inspections do not involve opening a vehicle compartment and 45% last 20 seconds or less. Slightly less than 2% of vehicles are referred to secondary inspections. Three policy options are considered for allowing more thorough primary inspections. The first would require all primary inspections to involve opening a vehicle compartment. This would increase average inspection times from 34 seconds to 70 seconds. The more thorough inspections would reduce the processing capacity of the ports of entry to roughly 50% of current peak hourly demand, creating congestion with the potential to propagate throughout the regional traffic network. The second option would limit the time in primary inspections to 63 seconds. Vehicles requiring more time to complete the inspection process would be referred to secondary inspections which would greatly increase the frequency of referrals to secondary inspections, but allow for the percentage of primary inspections that involve physical inspection of at least one vehicle compartment to be increased to 35%. The third option would increase the number of crossers in the SENTRI program, where pre-screened participants are subject to expedited inspections. Reducing the volume in the non-SENTRI lanes would allow more detailed inspections in these lanes. However, SENTRI participants currently constitute only a small portion of total border crossers. A doubling of the current SENTRI program would be required to raise the average non-SENTRI, primary inspection time from 34 to 40 seconds. This study concludes that none of these options, whether alone or in combination, have the potential to avoid conflicts between national security requirements that favor more detailed inspections and local traffic flow consideration that favor less detailed inspections. However, these strategies are amenable to incremental implementation, and such incremental implementation may increase the thoroughness of the inspection process without interfering with local traffic flows

    Pitfalls of Professionalism? Military Academies and Coup Risk

    Military academies tend to be strongly linked to the professionalization of the armed forces. This explains why many countries in the world have created such institutions. The following article studies a potential negative externality stemming from military schools: increased coup risk. We argue that military academies may create, inculcate, and strengthen cohesive views that could conflict with incumbent policies, and that these schools establish networks among military officers that may facilitate coordination necessary for plotting a putsch. We also contend and empirically demonstrate that these negative side effects of military academies are in particular pronounced in nondemocracies, that is, military academies have diverse effects across regime types. This work has significant implications for our understanding civil–military relations. Furthermore, we contribute to the literature on military education and professionalization, as we suggest that military academies are important vehicles through which coups can emerge predominantly in authoritarian states

    DNA-Free Recombinant SV40 Capsids Protect Mice from Acute Renal Failure by Inducing Stress Response, Survival Pathway and Apoptotic Arrest

    Viruses induce signaling and host defense during infection. Employing these natural trigger mechanisms to combat organ or tissue failure is hampered by harmful effects of most viruses. Here we demonstrate that SV40 empty capsids (Virus Like Particles-VLPs), with no DNA, induce host Hsp/c70 and Akt-1 survival pathways, key players in cellular survival mechanisms. We postulated that this signaling might protect against organ damage in vivo. Acute kidney injury (AKI) was chosen as target. AKI is critical, prevalent disorder in humans, caused by nephrotoxic agents, sepsis or ischemia, via apoptosis/necrosis of renal tubular cells, with high morbidity and mortality. Systemic administration of VLPs activated Akt-1 and upregulated Hsp/c70 in vivo. Experiments in mercury-induced AKI mouse model demonstrated that apoptosis, oxidative stress and toxic renal failure were significantly attenuated by pretreatment with capsids prior to the mercury insult. Survival rate increased from 12% to >60%, with wide dose response. This study demonstrates that SV40 VLPs, devoid of DNA, may potentially be used as prophylactic agent for AKI. We anticipate that these finding may be projected to a wide range of organ failure, using empty capsids of SV40 as well as other viruses
