Two-loop corrections to the parameter in Two-Higgs-Doublet Models
Models with two scalar doublets are among the simplest extensions of the
Standard Model which fulfill the relation at lowest order for the
parameter as favored by experimental data for electroweak observables
allowing only small deviations from unity. Such small deviations
originate exclusively from quantum effects with special sensitivity to mass
splittings between different isospin components of fermions and scalars. In
this paper the dominant two-loop electroweak corrections to are
calculated in the -conserving THDM, resulting from the top-Yukawa coupling
and the self-couplings of the Higgs bosons in the gauge-less limit. The
on-shell renormalization scheme is applied. With the assumption that one of the
-even neutral scalars represents the scalar boson observed by the LHC
experiments, with standard properties, the two-loop non-standard contributions
in can be separated from the standard ones. These contributions
are of particular interest since they increase with mass splittings between
non-standard Higgs bosons and can be additionally enhanced by and
, an additional free coefficient of the Higgs potential, and can
thus modify the one-loop result substantially. Numerical results are given for
the dependence on the various non-standard parameters, and the influence on the

Biographic narratives are shaped by many factors. The shape old people finally select for the retrospective view of their lives, is for example influenced by the current narrative situation, by the individual's perception of the past, or by the patterns of reconstruction. Using the example of 67 biographical narratives, the graduate thesis âTelling about life in the 20th Centuryâ demonstrates, how cultural influences shape the practice of memory, and how narrative traditions ultimately decide on the form of autobiographical representation.Lebensgeschichtliche ErzĂ€hlungen werden von vielen Faktoren geformt. Von der gegenwĂ€rtigen ErzĂ€hlsituation ĂŒber die individuelle Wahrnehmung des Vergangenen bis hin zur Rekonstruktion in Form von Erinnerung und ErzĂ€hlung prĂ€gen viele EinflĂŒsse die Form, die alte Menschen schlieĂlich fĂŒr die retrospektive Darstellung ihres Lebens wĂ€hlen. Die vorliegende Arbeit hat zum Ziel ein theoretisches und methodisches Fundament fĂŒr die Erforschung leiblichen Empfindens am Beispiel des leiblich-affektiven Spracherlebens in einem monolingual habitualisierten Feld zu bilden.
Ausgang wird zum einen von der in der PhĂ€nomenologie begrĂŒndeten Theorie der Leiblichkeit Maurice Merleau-Pontys genommen, welche den Leib als das Medium der Wahrnehmung und des Bezugs zur Welt erkennt. Zum anderen wird eine Mehrsprachigkeit von SprecherInnen im Sinne eines versatilen Einsatzes sprachlicher Mittel und somit ein lebensweltlicher Multilingualismus konstatiert. Zusammen mit Letzterem bildet ein habitualisierter Monolingualismus, welcher sich in und durch Sprachideologien einsprachiger Ausrichtung grĂŒndet, ein Spannungsfeld, in dem das leibliche Erfahren von Sprache zu betten ist. Das leibliche Erleben ist als ein sinnlich-sinnhafter Entwurf zur Welt zu verstehen, welcher ĂŒber Strukturen und Funktionen verfĂŒgt, und folglich ein aufschlussreiches Gebiet fĂŒr soziologische Forschung darstellt
Sprache & das Selbst
Die vorliegende Arbeit befasst sich mit der Frage, welche Rollen und Funktionen Sprache bei der Selbst-Konstruktion von russischen und ukrainischen MigrantInnen nach der Migration ĂŒbernimmt. Vom Umgang mit Sprache werden SchlĂŒsse auf die sozialen und gesellschaftlichen Strukturen der Lebenswelt von russischsprachigen MigrantInnen gezogen. Dazu sind von Juli bis Oktober 2009 zehn Interviews mit zehn Personen, die in Russland oder der Ukraine geboren sind und heute in Wien leben, gefĂŒhrt worden.
MigrantInnen aus Russland und der Ukraine stellen fĂŒr die Verortung ihres Selbst diskursiv ethnische, nationale und kulturelle Zugehörigkeiten her, wobei sowohl die russische, als auch die deutsche Sprache eine bedeutende Rolle spielen. Es konnten drei Funktionen von Sprache identifiziert werden: Erstens stellen Russisch-Erstsprachkenntnisse der Befragten ein fĂŒr den Arbeitsmarkt und die soziale Anerkennung relevantes Kapital dar. Zweitens ist die russische Sprache der Konnex zu dem persönlichen und familiĂ€ren Hintergrund, der die Vergangenheit mit der Zukunft einer Person verbindet und so eine sinnvolle Konstruktion eines Selbst ermöglicht. Drittens werden sowohl russisch, als auch deutsch fĂŒr ethnische Selbst- und Fremdaskriptionen herangezogen. Besonders russische Muttersprachen- und Erstsprachenkenntnisse werden als Beleg fĂŒr eine ethnische Zugehörigkeit funktionalisiert, wohingegen Fremdzuschreibungen auf Abweichungen von einem Standdarddeutsch beruhen. WĂ€hrend Selbstaskriptionen das Selbst in seinem Wert erhöhen, dienen Fremdzuschreibungen einer Wertminderung und einer Statusherabsetzung. Dieser Umstand verdeutlicht, dass Selbst-Konstruktionen einer Person in höchstem MaĂe von Machtstrukturen einer Gesellschaft beeinflusst werden. Eine Sprachideologie schafft eine Teilung einer Gesellschaft in Gruppen von âGleichenâ und âFremdenâ. Die Konstruktion des Selbst mittels Sprache steht bei MigrantInnen besonders deutlich im Kontext von Machtbeziehungen sowie vom Ringen um gesellschaftliche Vormachtstellung

Small RNA and genome interactions in Chlamydomonas reinhardtii recombinants
Examined by Phillip Wigge (Sainsbury) andWhen conspecific individuals are crossed, the ensuing hybridization creates a spectrum of phenotypes in the resulting offspring. Many of hybrid traits will be additive, similar to the parental phenotypes. In some cases however, transgressive phenotypes are formed, outside the range of that of the parental phenotypes.
Transgressive phenotypes can either be restricted to the F1 generation or be heritable throughout the hybrid lineage. While the mechanism behind heritable transgressive phenotyped is yet to be determined, transgressive gene expression is thought to be the root cause of their formation. Epigenetics modifications, heritable variation separate to the DNA code, can alter gene expression, persist through generations, and vary between individuals and over time. This makes them ideal candidates to be involved in the formation of transgressive phenotypes.
RNA silencing is an epigenetic mechanism of gene regulation relying on 20Q24nt single stranded small RNAs (sRNAs). Small RNAs, due to their ability to set up persistent epigenetic marks at a locus, have the potential to create heritable transgressive gene expression. For example, when genetic variation from one parental genome presents novel targets to the sRNAs of the other parental genome, new epigenetic marks such as DNA methylation or secondary sRNAs can be created at target sites.
In order to understand the potential of small RNAs to influence hybrid phenotype, I designed crossing experiments with Chlamydomonas reinhardtii, choosing this unicellular alga due to the genetic tools available and the haploid nature of its vegetative cells. The specific aim of the experiment was to identify transgressively expressed sRNA populations. Crossing two geographically distinct strains of C. reinhardtii, and sequencing both the genomes and sRNAomes of parents and recombinants, I was able catalogue both genetic and epigenetic variation in the parental strains providing unique insight into the inheritance of small RNAs in this alga.
In this thesis, I first compare the genomes of the parental strains, identifying polymorphisms and assessing genetic variation in RNA silencing pathway components. I then describe the sRNA profiles of the parental strains, identifying differentially expressed sRNA loci. I then describe my approach to identifying transgressively expressed sRNA loci in the hybrids. While many sRNA loci in the recombinants exhibit additive sRNA expression, I found multiple transgressively expressed sRNA loci.
Using the available bioinformatics tools, I identified potential miRNAs and phased secondary sRNAs within the list of transgressively expressed loci. Target analysis of one of the transgressively expressed miRNAs linked it with the transgressive expression of certain phased loci, suggesting a potential for sRNAs to be able to set up heritable epigenetic marks in recombinant C. reinhardtii cells.Funding provided by the Lewin-Fritsch Studentship (Downing College), Lander Studentship (Downing College) and the Gene Silencing Group in the Department of Plant Sciences

Die Arbeit gibt Einblick in das leiblich-affektive Spracherleben mehrsprachiger und in Wien lebender Menschen. Zu diesem Zwecke wurde eine qualitative Forschung durchgefĂŒhrt, im Zuge derer elf Interviews mit zehn InterviewpartnerInnen gefĂŒhrt, fein transkribiert und in Auswertungsteams einer Sequenzanalyse unterzogen wurden. Diskutiert werden insbesondere GefĂŒhle der Aggression, des Stolzes, der Scham und Peinlichkeit, der Freude und Traurigkeit sowie des Selbstwerts und des Minderwerts, welche die Befragten als wesentlich fĂŒr das Erleben von Sprache in einem Spannungsfeld von lebensweltlicher MultilingualitĂ€t und habitualisierter MonolingualitĂ€t artikulieren. Das theoretische Fundament bilden sowohl die in der PhĂ€nomenologie verankerte Theorie der Leiblichkeit Maurice Merleau-Pontys, als auch Sprachkonzepte der MultilingualitĂ€t, entsprechend derer Menschen zu einer umfassenden, verleiblichten und engagierten varilingualen Interaktion mit der Welt befĂ€higt sind
MORN-repeat proteins and TbMORN1 targeting in Trypanosoma brucei
Im Protist Trypanosoma brucei liegen die meisten seiner kanonischen Organellen- das Flagellum, das Mitochondrium, der Golgi-Apparat- als einfache Kopie vor und daher wird er gerne zur Beantwortung von elementaren zellbiologischen Fragen he-rangezogen. Bei Studien ĂŒber die Replikation des Golgi-Apparats wurde eine neue Organelle des Zytoskeletts, genannt Bilobe, entdeckt. KĂŒrzlich wurden zwei struktu-relle Komponenten der Bilobe TbMORN1 und TbLRRP1 identifiziert. TbMORN1 besteht aus einer höchst repetitiven PrimĂ€rstruktur von 15 MORN-repeats, daher muss sich die Zielinformation zur Bilobe innerhalb dieser Wiederholungen befinden. Im Genom von T. brucei gibt es 17 weitere Leserahmen, welche MORN-repeat-Proteine vorhersagen. Keines dieser Proteine ist experimentell charakterisiert. Unser primĂ€rer Ansatz war es daher, diese Proteine in prozyklischen Trypanosomen zu exprimieren und auf eine Lokalisierung auf der Bilobe hin zu untersuchen. Sieben Proteine lokalisierten zu dem Axoneme, eines zu dem PFR, zwei wurden am Basal body gefunden, drei waren im Zytosol vorhanden und eines möglicherweise auf dem Mitochondrium. 10 von 14 Proteinen wurden auf Strukturen des Zytoskeletts gefun-den, was auf eine strukturelle Rolle der MORN-repeat-Proteine hinweist. Jedoch wurde keines auf der Bilobe entdeckt. Da die Zielinformation in der TbMORN1-Sequenz vorhanden sein muss, fĂŒhrte ich parallel dazu VerkĂŒrzungsexperimente durch. Interessanterweise wurden die meisten verkĂŒrzten Proteine zusĂ€tzlich zur Bi-lobe am basal body gefunden. Des Weiteren beobachtete ich bei C-terminal verkĂŒrz-ten Proteinen TbMORN1(1-350) â TbMORN1(1-260), ein Muster von Lokalisierung und Misslokalisierung zur Bilobe. Meine Vermutung ist, dass diese ungewöhnliche Beobachtung durch hydrophobische Schlaufen, welche das Protein eventuell an die Plasmamembran binden können, zu erklĂ€ren ist. Nach Auswertung aller Daten erga-ben sich fĂŒnf wichtige DomĂ€nen. Die AminosĂ€uren 1-72 und 350-353 sind fĂŒr eine Lokalisierung ausschlieĂlich zur Bilobe notwendig, um eine starke, stabile Bindung herzustellen. FĂŒr die Bildung von sognannten âSpearsâ, parakistallinen Artefakten, wurden die AminosĂ€uren 343-350 als wichtig gefunden. FĂŒr das Bilobe-targeting sind die AminosĂ€uren 215-312 notwendig, wobei gleichzeitig ein targeting zum basal body stattfand. FĂŒr die Lokalisierung zum basal body war das letzte MORN-repeat ausreichend. Es ist möglich, dass das targeting durch eine Sequenz, welche nicht Teil der MORN-repeats ist, bewerkstelligt wurde. Aus meinen Daten ziehe ich den Schluss, dass das 3D-Motiv der entscheidende Faktor fĂŒr das targeting von TbMORN1 an die Bilobe ist.The protist Trypanosoma brucei posses most canonical eukaryotic organelles - flagel-lum, mitochondrion, Golgi - in single copy and is therefore widely used to address basic cell biological questions. Previously a novel cytoskeletal organelle called the bilobe was serendipitously discovered during studies on Golgi replication. Recently two of its structural components TbMORN1 and TbLRRP1 were identified. TbMORN1 consists of a highly repetitive primary structure of 15 MORN - repeats. Therefore the targeting information to the bilobe must be somewhere in these repeats. In the T.brucei genome there are 17 other open reading frames predicted to encode MORN-repeat proteins which are all experimentally uncharacterized. The candidate approach was to express these proteins in procyclic Trypanosomes and screen them for localization redolent of the bilobe. 7 proteins localized to the axoneme, 1 to the PFR, 2 were found at the basal body, 3 were present in the cytosol and one might be mitochondrial. 10/14 proteins where found on cytoskeletal structures which hints at a structural role of MORN-repeat proteins. However none were found at the bilobe. Since TbMORN1 must have the targeting information somewhere in its sequence I in parallel generated truncation constructs to gain information on which MORN-repeats are essential and which are dispensable for bilobe localization. Interestingly most of the truncated proteins were additionally found at the basal body in intact cells which suggests that tagged proteins might traffic to the bilobe through the basal body. I further observed a strange pattern of localization and mis-localization to the bilobe in C-terminal truncations, TbMORN1(1-350)- TbMORN1(1-260). I suggest that this unusual observation could be explained by hydrophobic loops which might bind the protein to the plasma membrane. Combining data from all truncations, I found that 5 domains are important. Amino acids 1-72 and 350-353 are required for targeting exclusively to the bilobe and forming a strong, stable association. The domain 343-350 was found important for the formation of spears, artefactual paracrystaline structures. Further, amino acids 215-312 are necessary for bilobe targeting with impaired targeting to the basal body. For targeting to the basal body the last MORN-repeat was sufficient. It is possible that the targeting is achieved through a sequence that is not part of the MORN-repeat consensus sequence. Concluding from our data I rather suggest that a 3D motif is the crucial factor for targeting TbMORN1
The mycotoxin deoxynivalenol predisposes for the development of Clostridium perfringens-induced necrotic enteritis in broiler chickens
Both mycotoxin contamination of feed and Clostridium perfringens-induced necrotic enteritis have an increasing global economic impact on poultry production. Especially the Fusarium mycotoxin deoxynivalenol (DON) is a common feed contaminant. This study aimed at examining the predisposing effect of DON on the development of necrotic enteritis in broiler chickens. An experimental Clostridium perfringens infection study revealed that DON, at a contamination level of 3,000 to 4,000 mg/kg feed, increased the percentage of birds with subclinical necrotic enteritis from 2062.6% to 4763.0% (P<0.001). DON significantly reduced the transepithelial electrical resistance in duodenal segments (P<0.001) and decreased duodenal villus height (P = 0.014) indicating intestinal barrier disruption and intestinal epithelial damage, respectively. This may lead to an increased permeability of the intestinal epithelium and decreased absorption of dietary proteins. Protein analysis of duodenal content indeed showed that DON contamination resulted in a significant increase in total protein concentration (P = 0.023). Furthermore, DON had no effect on in vitro growth, alpha toxin production and netB toxin transcription of Clostridium perfringens. In conclusion, feed contamination with DON at concentrations below the European maximum guidance level of 5,000 mg/kg feed, is a predisposing factor for the development of necrotic enteritis in broilers. These results are associated with a negative effect of DON on the intestinal barrier function and increased intestinal protein availability, which may stimulate growth and toxin production of Clostridium perfringens
