    Land consumption processes in alluvial plains and abandonment of agricultural and breeding activities in mountain areas, both of which result in the loss of agricultural land, are today the major threats to biodiversity in Tuscany. These processes result in an increase in environmental fragmentation in the plain and in the homogenisation of mountain landscapes. The Project for a Tuscan ecological network, as an invariant element of the Regional landscape Plan, is a new tool to counteract such dynamics, highlighting the remarkable naturalistic interest of traditional agricultural landscapes. In the context of biodiversity conservation policies, the ecological network also highlights the need to overcome the 'protected islands' approach by using planning tools apt to improve the levels of quality and widespread permeability of the regional territorial ecosystem

    Interventions that can reduce the loss and the fragmentation of ecosystems are very important in the sphere of biodiversity conservation policies applied to areas of high human activity. Ecological networks, on a di erent scale, are important analytical and design tools, and they can highlight sources of biodiversity with the intention of ecological unification. The Tuscany Ecological Network project, based on habitat sustainability models and converted into a planning tool for the landscape plan of Tuscany, has underlined the need to pursue the objectives of biodiversity conservation within a landscape or extended area.     Status of global fits to neutrino oscillations

    We review the present status of global analyses of neutrino oscillations, taking into account the most recent neutrino data including the latest KamLAND and K2K updates presented at Neutrino2004, as well as state-of-the-art solar and atmospheric neutrino flux calculations. We give the two-neutrino solar + KamLAND results, as well as two-neutrino atmospheric + K2K oscillation regions, discussing in each case the robustness of the oscillation interpretation against departures from the Standard Solar Model and the possible existence of non-standard neutrino physics. Furthermore, we give the best fit values and allowed ranges of the three-flavour oscillation parameters from the current worlds' global neutrino data sample and discuss in detail the status of the small parameters \alpha \equiv \Dms/\Dma as well as sin⁥2Ξ13\sin^2\theta_{13}, which characterize the strength of CP violating effects in neutrino oscillations. We also update the degree of rejection of four-neutrino interpretations of the LSND anomaly in view of the most recent developments.Comment: v6: In the last Appendix we provide updated neutrino oscillation results which take into account the relevant oscillation data released by the MINOS and KamLAND collaboration

    Spinocerebellar Ataxia Tethering PCR: A Rapid Genetic Test for the Diagnosis of Spinocerebellar Ataxia Types 1, 2, 3, 6, and 7 by PCR and Capillary Electrophoresis

    Spinocerebellar ataxias (SCA) type 1, 2, 3, 6, and 7, associated with a (CAG)n repeat expansion in coding sequences, are the most prevalent autosomal dominant ataxias worldwide (approximately 60% of the cases). In addition, the phenotype of SCA2 expansions has been now extended to Parkinson's disease and amyotrophic lateral sclerosis. Their diagnosis is presently based on a PCR to identify small expanded alleles, followed by a second-level test whenever the suspect of false normal homozygous, or a CAT interruption in SCA1 needs to be verified. Next-generation sequencing still does not allow efficient detection of these repeats. Here, we show the efficacy of a novel, rapid, and cost-effective method to identify and size pathogenic expansions in SCA1-3, 6, and 7 and recognize large alleles or interruptions without a second-level test. Twenty-five healthy controls and 33 expansion carriers were analyzed: alleles migrated consistently in different PCRs/capillary runs, and homozygous subjects were always distinguishable from heterozygous carriers of both common and large (>100 repeats) pathogenic CAG expansions. Repeat number could be calculated counting the number of peaks, except for the largest SCA2 and SCA7 alleles. Interruptions in SCA1 were always visible. Overall, our method allows a simpler, cost-effective, and sensibly faster SCA diagnostic protocol compared to the standard technique and to the still unadapted next-generation sequencing

    MicroRNA-Restricted Transgene Expression in the Retina

    Background: Gene transfer using adeno-associated viral (AAV) vectors has been successfully applied in the retina for the treatment of inherited retinal dystrophies. Recently, microRNAs have been exploited to fine-tune transgene expression improving therapeutic outcomes. Here we evaluated the ability of retinal-expressed microRNAs to restrict AAV-mediated transgene expression to specific retinal cell types that represent the main targets of common inherited blinding conditions. Methodology/Principal Findings: To this end, we generated AAV2/5 vectors expressing EGFP and containing four tandem copies of miR-124 or miR-204 complementary sequences in the 39UTR of the transgene expression cassette. These vectors were administered subretinally to adult C57BL/6 mice and Large White pigs. Our results demonstrate that miR-124 and miR-204 target sequences can efficiently restrict AAV2/5-mediated transgene expression to retinal pigment epithelium and photoreceptors, respectively, in mice and pigs. Interestingly, transgene restriction was observed at low vector doses relevant to therapy.     Impacts of the Invasive Alien <i>Carpobrotus</i> spp. on Coastal Habitats on a Mediterranean Island (Giglio Island, Central Italy)

    Carpobrotus acinaciformis and C. edulis are well-known invasive alien plants native to South Africa, whose detrimental effects on native communities are widely documented in the Mediterranean basin and thus largely managed in coastal ecosystems. Most of the literature on these species focuses on their impacts on habitats of sandy coastal dunes, while the effects of Carpobrotus spp. invasion on other habitats such as rocky cliffs and coastal scrubs and garrigues are almost neglected. We present a study case conducted on a small Mediterranean island where Carpobrotus spp. invaded three different natural habitats listed within the Habitat Directive 92/43/CEE (Natura 2000 codes 1240, 1430, and 5320). We surveyed the presence and abundance of native species and Carpobrotus spp. on 44 permanent square plots of 4 m2 in invaded and uninvaded areas in each of the three habitats. We found impacts on plant alpha diversity (intended as the species diversity within each sampled plot) in all the habitats investigated in terms of a decrease in species richness, Shannon index, and abundance. Invaded communities also showed a severe change in species composition with a strong homogenization of the floras of the three habitats. Finally, the negative effect of invasion emerged even through the analyses of beta diversity (expressing the species diversity among sampled plots of the same habitat type), with Carpobrotus spp. replacing a large set of native species
