    A preocupa??o com sa?de p?blica ? um fator relevante na hist?ria da humanidade, epidemias de doen?as causadas por v?rus e bact?rias podem provocar impactos socioecon?micos, nas ?reas atingidas por elas. Essa preocupa??o tem impulsionado a pesquisa e o desenvolvimento de dispositivos que sejam capazes de prevenir e detectar patologias auxiliando no tratamento. Ao longo dos anos t?m sido propostas e desenvolvidas diversas plataformas de sensoriamento visando a detec??o e an?lise de subst?ncias qu?micas e biol?gicas que podem caracterizar algumas patologias espec?ficas na ?rea de sa?de. A constru??o dessas plataformas sensoras resultam de estudos multidisciplinares que agregam diferentes ?reas de estudo, como engenharia e medicina resultando em diversas configura??es de sensores e biossensores. A obten??o de sensoriamento pode ser realizada a partir de algumas abordagens, dentre as quais destaca-se o efeito de resson?ncia de pl?smons de superf?cie (Surface Plasmon Resonance - SPR) que, aliado ? tecnologia das fibras ?pticas, tem possibilitado a constru??o de biochips ?pticos que integram o sistema de sensoriamento. As fibras ?pticas apresentam algumas vantagens de utiliza??o em compara??o com os biochips tradicionais baseados em prisma, tais como, dimens?es reduzidas e imunidade ?s interfer?ncias eletromagn?ticas, uma caracter?stica inerente ?s fibras ?pticas. Dentre as fibras, um tipo especial tem sido utilizado na constru??o de biochips SPR para an?lise de intera??es biomoleculares, as chamadas fibras ?pticas microestruturadas (Microstructured Optical Fibers ? MOFs). Estas fibras possuem caracter?sticas geom?tricas que permitem o controle e/ou a manipula??o da propaga??o luz. Altera??es na geometria destas fibras podem causar varia??es na propaga??o da luz, tornandoas interessantes a aplica??es que exploram a intera??o entre elas, biomol?culas e filmes finos met?licos. Este trabalho apresenta um modelo te?rico de biochip ?ptico constru?do a partir de fibras ?pticas microestruturadas em que a resson?ncia de pl?smons de superf?cie ? utilizada como forma de detec??o.     Increase in Electrical Parameters Using Sucrose in Tomato Waste

    "The use of organic waste as fuel for energy generation will reduce the great environmental problems currently caused by the consumption of fossil sources, giving agribusiness companies a profitable way to use their waste. In this research, tomato waste with different percentages of sucrose (0-target, 5, 10, and 20%) was used in microbial fuel cells manufactured on a laboratory scale with zinc and copper electrodes, managing to generate maximum peaks of voltage and a current of 1.08 V and 6.67 mA in the cell with 20% sucrose, in which it was observed that the optimum operating pH was 5.29, while the MFC with 0% (target) sucrose generated 0.91 V and 3.12 A on day 13 with a similar pH, even though all the cells worked in an acidic pH. Likewise, the cell with 20% sucrose had the lowest internal resistance (0.148541 ± 0.012361 KΩ) and the highest power density (224.77 mW/cm2 ) at a current density of 4.43 mA/cm2 , while the MFC with 0% sucrose generated 160.52 mW/cm2 and 4.38 mA/cm2 of power density and current density, respectively, with an internal resistance of 0.34116 ± 0.2914 KΩ. In this sense, the FTIR (Fourier-transform infrared spectroscopy) of all the substrates used showed a high content of phenolic compounds and carboxylate acids. Finally, the MFCs were connected in a series and managed to generate a voltage of 3.43 V, enough to light an LED (green). These results give great hope to companies and society that, in the near future, this technology can be taken to a larger scale.

    Aromatic Compound-Dependent Staphylococcus aureus

    Staphylococcus aureus nasal carriage is a risk factor for individuals suffering from trauma, surgical procedures, invasive devices, and/or decreased immunity. Recently, we demonstrated that artificial nasal colonization with an attenuated S. aureus mutant reduced by bacterial interference with the colonization of pathogenic strains of S. aureus. This could be an optional tool to diminish the rate of S. aureus infections in hospitalized patients. The aim of this study was to construct a safe ΔaroA mutant of S. aureus and to discriminate it from nasal colonizing and osteomyelitis S. aureus isolates by SmaI pulsed-field gel electrophoresis (PFGE) typing. The ΔaroA mutant, named RD17, exhibited an LD50 (3.2 × 106 colony-forming unit (CFU)) significantly higher than that of the parental strain (2.2 × 103 CFU). The colony number of the RD17 mutants recovered from nares of leukopenic mice was similar to that observed in the animals of the control group. Therefore, the ΔaroA mutant was demonstrated to be safe due to maintaining low growth levels in the nares regardless of immune status of the animals. PFGE typing allowed the unequivocal identification of the S. aureus and differentiation of aroA mutants in nasal colonizing and osteomyelitis isolates. This information could be important to discriminate endogenous infections from laboratory strains of S. aureus

    <i>orsai</i>, the Drosophila homolog of human ETFRF1, links lipid catabolism to growth control

    BACKGROUND: Lipid homeostasis is an evolutionarily conserved process that is crucial for energy production, storage and consumption. Drosophila larvae feed continuously to achieve the roughly 200-fold increase in size and accumulate sufficient reserves to provide all energy and nutrients necessary for the development of the adult fly. The mechanisms controlling this metabolic program are poorly understood. RESULTS: Herein we identified a highly conserved gene, orsai (osi), as a key player in lipid metabolism in Drosophila. Lack of osi function in the larval fat body, the regulatory hub of lipid homeostasis, reduces lipid reserves and energy output, evidenced by decreased ATP production and increased ROS levels. Metabolic defects due to reduced Orsai (Osi) in time trigger defective food-seeking behavior and lethality. Further, we demonstrate that downregulation of Lipase 3, a fat body-specific lipase involved in lipid catabolism in response to starvation, rescues the reduced lipid droplet size associated with defective orsai. Finally, we show that osi-related phenotypes are rescued through the expression of its human ortholog ETFRF1/LYRm5, known to modulate the entry of β-oxidation products into the electron transport chain; moreover, knocking down electron transport flavoproteins EtfQ0 and walrus/ETFA rescues osi-related phenotypes, further supporting this mode of action. CONCLUSIONS: These findings suggest that Osi may act in concert with the ETF complex to coordinate lipid homeostasis in the fat body in response to stage-specific demands, supporting cellular functions that in turn result in an adaptive behavioral response.     Ligand diversity contributes to the full activation of the jasmonate pathway in Marchantia polymorpha

    In plants, jasmonate signaling regulates a wide range of processes from growth and development to defense responses and thermotolerance. Jasmonates, such as jasmonic acid (JA), (+)-7-iso-jasmonoyl-l-isoleucine (JA-Ile), 12-oxo-10,15(Z)-phytodienoic acid (OPDA), and dinor-12-oxo-10,15(Z)-phytodienoic acid (dn-OPDA), are derived from C18 (18 Carbon atoms) and C16 polyunsaturated fatty acids (PUFAs), which are found ubiquitously in the plant kingdom. Bryophytes are also rich in C20 and C22 long-chain polyunsaturated fatty acids (LCPUFAs), which are found only at low levels in some vascular plants but are abundant in organisms of other kingdoms, including animals. The existence of bioactive jasmonates derived from LCPUFAs is currently unknown. Here, we describe the identification of an OPDA-like molecule derived from a C20 fatty acid (FA) in the liverwort Marchantia polymorpha (Mp), which we term (5Z,8Z)-10-(4-oxo-5-((Z)-pent-2-en-1-yl)cyclopent-2-en-1-yl)deca-5,8-dienoic acid (C20-OPDA). This molecule accumulates upon wounding and, when applied exogenously, can activate known Coronatine Insensitive 1 (COI1) -dependent and -independent jasmonate responses. Furthermore, we identify a dn-OPDA-like molecule (Δ4-dn-OPDA) deriving from C20-OPDA and demonstrate it to be a ligand of the jasmonate coreceptor (MpCOI1-Mp Jasmonate-Zinc finger inflorescence meristem domain [MpJAZ]) in Marchantia. By analyzing mutants impaired in the production of LCPUFAs, we elucidate the major biosynthetic pathway of C20-OPDA and Δ4-dn-OPDA. Moreover, using a double mutant compromised in the production of both Δ4-dn-OPDA and dn-OPDA, we demonstrate the additive nature of these molecules in the activation of jasmonate responses. Taken together, our data identify a ligand of MpCOI1 and demonstrate LCPUFAs as a source of bioactive jasmonates that are essential to the immune response of M. polymorpha.Peer reviewe

    Genomes of Fasciola hepatica from the Americas reveal colonization with Neorickettsia Endobacteria related to the agents of Potomac Horse and Human Sennetsu Fevers

    Food borne trematodes (FBTs) are an assemblage of platyhelminth parasites transmitted through the food chain, four of which are recognized as neglected tropical diseases (NTDs). Fascioliasis stands out among the other NTDs due to its broad and significant impact on both human and animal health, as Fasciola sp., are also considered major pathogens of domesticated ruminants. Here we present a reference genome sequence of the common liver fluke, Fasciola hepatica isolated from sheep, complementing previously reported isolate from cattle. A total of 14,642 genes were predicted from the 1.14 GB genome of the liver fluke. Comparative genomics indicated that F. hepatica Oregon and related food-borne trematodes are metabolically less constrained than schistosomes and cestodes, taking advantage of the richer millieux offered by the hepatobiliary organs. Protease families differentially expanded between diverse trematodes may facilitate migration and survival within the heterogeneous environments and niches within the mammalian host. Surprisingly, the sequencing of Oregon and Uruguay F. hepatica isolates led to the first discovery of an endobacteria in this species. Two contigs from the F. hepatica Oregon assembly were joined to complete the 859,205 bp genome of a novel Neorickettsia endobacterium (nFh) closely related to the etiological agents of human Sennetsu and Potomac horse fevers. Immunohistochemical studies targeting a Neorickettsia surface protein found nFh in specific organs and tissues of the adult trematode including the female reproductive tract, eggs, the Mehlis' gland, seminal vesicle, and oral suckers, suggesting putative routes for fluke-to-fluke and fluke-to-host transmission. The genomes of F. hepatica and nFh will serve as a resource for further exploration of the biology of F. hepatica, and specifically its newly discovered transkingdom interaction with nFh and the impact of both species on disease in ruminants and humans

    R534C mutation in hERG causes a trafficking defect in iPSC-derived cardiomyocytes from patients with type 2 long QT syndrome

    Patient-specific cardiomyocytes obtained from induced pluripotent stem cells (CM-iPSC) offer unprecedented mechanistic insights in the study of inherited cardiac diseases. The objective of this work was to study a type 2 long QT syndrome (LQTS2)-associated mutation (c.1600C > T in KCNH2, p.R534C in hERG) in CM-iPSC. Peripheral blood mononuclear cells were isolated from two patients with the R534C mutation and iPSCs were generated. In addition, the same mutation was inserted in a control iPSC line by genome editing using CRISPR/Cas9. Cells expressed pluripotency markers and showed spontaneous differentiation into the three embryonic germ layers. Electrophysiology demonstrated that action potential duration (APD) of LQTS2 CM-iPSC was significantly longer than that of the control line, as well as the triangulation of the action potentials (AP), implying a longer duration of phase 3. Treatment with the IKr inhibitor E4031 only caused APD prolongation in the control line. Patch clamp showed a reduction of IKr on LQTS2 CM-iPSC compared to control, but channel activation was not significantly affected. Immunofluorescence for hERG demonstrated perinuclear staining in LQTS2 CM-iPSC. In conclusion, CM-iPSC recapitulated the LQTS2 phenotype and our findings suggest that the R534C mutation in KCNH2 leads to a channel trafficking defect to the plasma membrane.Fil: Mesquita, Fernanda C.     An empirical evaluation of camera trap study design: How many, how long and when?

    Abstract Camera traps deployed in grids or stratified random designs are a well‐established survey tool for wildlife but there has been little evaluation of study design parameters. We used an empirical subsampling approach involving 2,225 camera deployments run at 41 study areas around the world to evaluate three aspects of camera trap study design (number of sites, duration and season of sampling) and their influence on the estimation of three ecological metrics (species richness, occupancy and detection rate) for mammals. We found that 25–35 camera sites were needed for precise estimates of species richness, depending on scale of the study. The precision of species‐level estimates of occupancy (ψ) was highly sensitive to occupancy level, with 0.75) species, but more than 150 camera sites likely needed for rare (ψ < 0.25) species. Species detection rates were more difficult to estimate precisely at the grid level due to spatial heterogeneity, presumably driven by unaccounted habitat variability factors within the study area. Running a camera at a site for 2 weeks was most efficient for detecting new species, but 3–4 weeks were needed for precise estimates of local detection rate, with no gains in precision observed after 1 month. Metrics for all mammal communities were sensitive to seasonality, with 37%–50% of the species at the sites we examined fluctuating significantly in their occupancy or detection rates over the year. This effect was more pronounced in temperate sites, where seasonally sensitive species varied in relative abundance by an average factor of 4–5, and some species were completely absent in one season due to hibernation or migration. We recommend the following guidelines to efficiently obtain precise estimates of species richness, occupancy and detection rates with camera trap arrays: run each camera for 3–5 weeks across 40–60 sites per array. We recommend comparisons of detection rates be model based and include local covariates to help account for small‐scale variation. Furthermore, comparisons across study areas or times must account for seasonality, which could have strong impacts on mammal communities in both tropical and temperate sites

    THE PROCESS OF WORKING IN NURSING SUPERVISION

    Objetivos: Descrever o processo de trabalho dos enfermeiros supervisores das Equipes Saúde da Família; identificar os desafios vivenciados pelos supervisores no processo de supervisão das Equipes Saúde da Família e discutir as possibilidades de novas práticas de trabalho que contribuam para efetivação do modelo centrado em prevenção e promoção da saúde. Método: Trata-se de uma pesquisa de campo, descritiva com abordagem qualitativa. Resultados: As supervisoras ressaltaram a importância do planejamento com as equipes, porém enfatizaram a sobrecarga de trabalho relativa à resolução de problemas administrativos da Unidade Básica de Saúde como um desafio a ser superado. Conclusão: As supervisoras demonstraram um desejo de realizar o acompanhamento das equipes apenas com um aspecto de “convisão” com a finalidade de apoiar, orientar e ajudar, sem a responsabilização por problemas administrativos da UBS. A supervisão deve superar o enfoque restrito do controle de tarefas, avançando para o caráter educativo que essa ação implica
