    Accurate aeroacoustic measurements in closed-section hard-walled wind tunnels

    Noise emissions from aircraft are of major concern to aircraft manufacturers. There are various analytical, empirical and numerical tools to help in the design of quieter aircraft, however aeroacoustic measurements in wind tunnels are still required. There is a growing interest in simultaneous aerodynamic and aeroacoustic measurements in hard-walled closed-section wind tunnels. The research hypothesis of this work is whether accurate aeroacoustic measurements are possible in this type of wind tunnel. Two issues are of particular concern: the reverberation sound field and high background noise levels. De-reverberation, based on an Image Source Model (ISM), is proposed to tackle the first issue by incorporating the reflections in the focused beamformer. This technique is computationally fast and easy to implement. Source Power Integration and deconvolution techniques are shown to be still valid in de-reverberation. Measurements in a closed section wind tunnel have shown that an ISM gives a better estimate of the Green's functions, when compared to free-space Green's functions. Furthermore de-reverberation yielded more accurate source strength estimates from the beamformer. Qualitatively, de-convolved results were no different than when using free-space Green's functions. Simulations have shown that the ISM can become unstable at high frequencies if position errors are present. It is therefore recommended to limit the application of the ISM to frequencies below 10 kHz. At low frequencies the accuracy of beamforming levels is highly dependent on the level of noise contamination of the input data. Removing the diagonal of the cross spectral matrix might not be sufficient to eliminate this noise

    Global gene expression analyses of bystander and alpha particle irradiated normal human lung fibroblasts: Synchronous and differential responses

    <p>Abstract</p> <p>Background</p> <p>The existence of a radiation bystander effect, in which non-irradiated cells respond to signals from irradiated cells, is now well established. It raises concerns for the interpretation of risks arising from exposure to low doses of ionizing radiation. However, the regulatory mechanisms involved in the bystander response have not been well elucidated. To provide insight into the signaling pathways responding in bystanders, we have measured global gene expression four hours after bystander and direct alpha particle exposure of primary human lung fibroblasts.</p> <p>Results</p> <p>Although common p53-regulated radiation response genes like <it>CDKN1A </it>were expressed at elevated levels in the directly exposed cultures, they showed little or no change in the bystanders. In contrast, genes regulated by NFκB, such as <it>PTGS2 </it>(cyclooxygenase-2), <it>IL8 </it>and <it>BCL2A1</it>, responded nearly identically in bystander and irradiated cells. This trend was substantiated by gene ontology and pathway analyses of the microarray data, which suggest that bystander cells mount a full NFκB response, but a muted or partial p53 response. In time-course analyses, quantitative real-time PCR measurements of <it>CDKN1A </it>showed the expected 4-hour peak of expression in irradiated but not bystander cells. In contrast, <it>PTGS2, IL8 </it>and <it>BCL2A1 </it>responded with two waves of expression in both bystander and directly irradiated cells, one peaking at half an hour and the other between four and six hours after irradiation.</p> <p>Conclusion</p> <p>Two major transcriptional hubs that regulate the direct response to ionizing radiation are also implicated in regulation of the bystander response, but to dramatically different degrees. While activation of the p53 response pathway is minimal in bystander cells, the NFκB response is virtually identical in irradiated and bystander cells. This alteration in the balance of signaling is likely to lead to different outcomes in irradiated cells and their bystanders, perhaps leading to greater survival of bystanders and increased risk from any long-term damage they have sustained.</p

    Adjunctive rifampicin for Staphylococcus aureus bacteraemia (ARREST): a multicentre, randomised, double-blind, placebo-controlled trial.

    BACKGROUND: Staphylococcus aureus bacteraemia is a common cause of severe community-acquired and hospital-acquired infection worldwide. We tested the hypothesis that adjunctive rifampicin would reduce bacteriologically confirmed treatment failure or disease recurrence, or death, by enhancing early S aureus killing, sterilising infected foci and blood faster, and reducing risks of dissemination and metastatic infection. METHODS: In this multicentre, randomised, double-blind, placebo-controlled trial, adults (≥18 years) with S aureus bacteraemia who had received ≤96 h of active antibiotic therapy were recruited from 29 UK hospitals. Patients were randomly assigned (1:1) via a computer-generated sequential randomisation list to receive 2 weeks of adjunctive rifampicin (600 mg or 900 mg per day according to weight, oral or intravenous) versus identical placebo, together with standard antibiotic therapy. Randomisation was stratified by centre. Patients, investigators, and those caring for the patients were masked to group allocation. The primary outcome was time to bacteriologically confirmed treatment failure or disease recurrence, or death (all-cause), from randomisation to 12 weeks, adjudicated by an independent review committee masked to the treatment. Analysis was intention to treat. This trial was registered, number ISRCTN37666216, and is closed to new participants. FINDINGS: Between Dec 10, 2012, and Oct 25, 2016, 758 eligible participants were randomly assigned: 370 to rifampicin and 388 to placebo. 485 (64%) participants had community-acquired S aureus infections, and 132 (17%) had nosocomial S aureus infections. 47 (6%) had meticillin-resistant infections. 301 (40%) participants had an initial deep infection focus. Standard antibiotics were given for 29 (IQR 18-45) days; 619 (82%) participants received flucloxacillin. By week 12, 62 (17%) of participants who received rifampicin versus 71 (18%) who received placebo experienced treatment failure or disease recurrence, or died (absolute risk difference -1·4%, 95% CI -7·0 to 4·3; hazard ratio 0·96, 0·68-1·35, p=0·81). From randomisation to 12 weeks, no evidence of differences in serious (p=0·17) or grade 3-4 (p=0·36) adverse events were observed; however, 63 (17%) participants in the rifampicin group versus 39 (10%) in the placebo group had antibiotic or trial drug-modifying adverse events (p=0·004), and 24 (6%) versus six (2%) had drug interactions (p=0·0005). INTERPRETATION: Adjunctive rifampicin provided no overall benefit over standard antibiotic therapy in adults with S aureus bacteraemia.     KiDS-450: the tomographic weak lensing power spectrum and constraints on cosmological parameters

    We present measurements of the weak gravitational lensing shear power spectrum based on 450450 sq. deg. of imaging data from the Kilo Degree Survey. We employ a quadratic estimator in two and three redshift bins and extract band powers of redshift auto-correlation and cross-correlation spectra in the multipole range 76≤ℓ≤131076 \leq \ell \leq 1310. The cosmological interpretation of the measured shear power spectra is performed in a Bayesian framework assuming a Λ\LambdaCDM model with spatially flat geometry, while accounting for small residual uncertainties in the shear calibration and redshift distributions as well as marginalising over intrinsic alignments, baryon feedback and an excess-noise power model. Moreover, massive neutrinos are included in the modelling. The cosmological main result is expressed in terms of the parameter combination S8≡σ8Ωm/0.3S_8 \equiv \sigma_8 \sqrt{\Omega_{\rm m}/0.3} yielding $S_8 = \ 0.651 \pm 0.058(3z−bins),confirmingtherecentlyreportedtensioninthisparameterwithconstraintsfromPlanckat (3 z-bins), confirming the recently reported tension in this parameter with constraints from Planck at 3.2\sigma$ (3 z-bins). We cross-check the results of the 3 z-bin analysis with the weaker constraints from the 2 z-bin analysis and find them to be consistent.     SotonArray: Southampton University wind tunnel microphone array system guide

    This report shall serve as an introductory background to SotonArray, the microphone array system at the University of Southampton. It covers the practical aspects only; namely the instrumentation setup and the beamforming software. It aims to give a basic level of understanding of the system to the researcher who wants to use it, and especially to highlight both the capabilities and limitations of the system. Since the microphone array system is a new capability for the Aerodynamics and Flight Mechanics research group (AFM), the hardware and software is constantly being upgraded, and the details given here are correct only at the time of going to print

    Frequency response analysis of microphone preamplifiers in the audible and ultrasonic regime

    This report presents an investigation on the frequency response of two types of electret microphone preamplifiers currently in use with SotonArray, Southampton University's wind tunnel microphone array system. The investigation is presented as a comparison between the response of an electret microphone coupled to the preamplifiers and a reference instrumentation-grade microphone and preamplifier assembly. The results show that electret microphones can potentially be used above their rated frequency range of 20kHz; in fact no degradation in signal was noticed up to 48kHz. The results also show that the newly-built SES preamps have a much better high-frequency performance than the original ISVR preamps
