    Lyophilisation of influenza, rabies and Marburg lentiviral pseudotype viruses for the development and distribution of a neutralisation-assay based diagnostic kit

    Pseudotype viruses (PVs) are chimeric, replication-deficient virions that mimic wild-type virus entry mechanisms and can be safely employed in neutralisation assays, bypassing the need for high biosafety requirements and performing comparably to established serological assays. However, PV supernatant necessitates -80°C long-term storage and cold-chain maintenance during transport, which limits the scope of dissemination and application throughout resource-limited laboratories. We therefore investigated the effects of lyophilisation on influenza, rabies and Marburg PV stability, with a view to developing a pseudotype virus neutralisation assay (PVNA) based kit suitable for affordable global distribution. Infectivity of each PV was calculated after lyophilisation and immediate reconstitution, as well as subsequent to incubation of freeze-dried pellets at varying temperatures, humidities and timepoints. Integrity of glycoprotein structure following treatment was also assessed by employing lyophilised PVs in downstream PVNAs. In the presence of 0.5M sucrose-PBS cryoprotectant, each freeze-dried pseudotype was stably stored for 4 weeks at up to 37°C and could be neutralised to the same potency as unlyophilised PVs when employed in PVNAs. These results confirm the viability of a freeze-dried PVNA-based kit, which could significantly facilitate low-cost serology for a wide portfolio of emerging infectious viruses

    Assessing current genetic status of the Hainan gibbon using historical and demographic baselines: implications for conservation management of species of extreme rarity

    Evidence-based conservation planning is crucial for informing management decisions for species of extreme rarity, but collection of robust data on genetic status or other parameters can be extremely challenging for such species. The Hainan gibbon, possibly the world's rarest mammal, consists of a single population of c.25 individuals restricted to one protected area on Hainan Island, China, and has persisted for over 30 years at exceptionally low population size. Analysis of genotypes at 11 microsatellite loci from faecal samples for 36% of the current global population and tissue samples from 62% of existing historical museum specimens demonstrates limited current genetic diversity (Na=2.27, Ar=2.24, He =0.43); diversity has declined since the 19th century and even further within the last 30 years, representing declines of c.30% from historical levels (Na=3.36, Ar=3.29, He =0.63). Significant differentiation is seen between current and historical samples (FST =0.156, P=0.0315), and the current population exhibits extremely small Ne (current Ne =2.16). There is evidence for both a recent population bottleneck and an earlier bottleneck, with population size already reasonably low by the late 19th century (historical Ne =1162.96). Individuals in the current population are related at the level of half- to full-siblings between social groups, and full-siblings or parent-offspring within a social group, suggesting that inbreeding is likely to increase in the future. The species' current reduced genetic diversity must be considered during conservation planning, particularly for expectations of likely population recovery, indicating that intensive, carefully planned management is essential

    Gender and depression in Anglo-Jewry

    This study found similar prevalence of case depression among men as among women in a sample of 339 Jews affiliated to orthodox synagogues (157 men and 182 women). There were significant gender differences in several social-situational factors and symptoms, mostly in the direction that would suggest that case depression would be higher among women than among men. That this was not so is suggested to be the result of the cultural milieu: social factors that have been found to be associated with depression in other groups of people did not function as risk or vulnerability factors among the Jews studied. In particular, the evidence indicates the importance of specific cultural-religious values in contributing towards the prevalences that were observed. These values included the esteem attached to women's central role in family management, and the low use of alcohol and suicide as escape routes from depression

    The social circumstances of anxiety and its symptoms among Anglo-Jews

    In this community study of orthodox-affiliated Jews in London the social circumstances of anxiety were examined. By contrast with previous work on women in London, danger and early adversity bore only a weak relationship with anxiety in this sample. Eventfulness had the strongest relationship with anxiety of all the circumstances examined. Women were more likely to suffer from borderline anxiety than were men, but there were no gender differences in case anxiety. Women had more eventful lives than men but this could not solely account for gender differences in anxiety. Findings suggest the importance of cultural factors in aetiology

    Characteristics of vertebrate-dispersed fruits in Hong Kong

    Hong Kong has a native angiosperm flora of approximately 1800 species, of which 27% (482 spp) bear fleshy, presumably vertebrate-dispersed fruits, including 76% of the 337 tree and shrub species and 70% of the 103 climber species. Morphological characteristics were determined for 255 species and nutritional characteristics of the fruit pulp for 153 species. Most fruit species were black (45 1%) or red (24.3%) and 85.9% had a mean diameter <13 mm Nutritional characteristics varied widely between species with ranges and median values as follows: pulp percentage (range 10.0-99 2%, median 69 2%), water content of pulp (11 1-94 0%, 78%), lipid (0-84 0%, 2 0%), soluble carbohydrate (4-88%, 53%), nitrogen (0.2-3.4%, 0.86%), neutral detergent fibre (1-44%, 14.3%) Fruit development time (50-360 d, 156 d) showed a negative correlation with lipid content, but no significant correlation with fruit or seed size. Principal components analysis of fruit characteristics was dominated by a trend from single-seeded fruits with a thin, lipid-rich pulp layer to multiple-seeded fruits with much, watery, carbohydrate-rich pulp. Bird-dispersed species cover the full range of fruit characteristics except those too large to swallow and too hard to peck bits from. Mammals (bats, civets and/or macaques) are known or suspected to consume most of the fruits too large for birds as well as many bird fruits but none with high-lipid content Summer fruits (May-September) were significantly larger and had significantly higher seed size and carbohydrate content than winter fruits (November-March) Winter fruits took more than twice as long to develop as summer fruits.published_or_final_versio

    RemoveDebris Mission, In Orbit Operations

    International audienceThe RemoveDebris mission has been the first Active Debris Removal (ADR) mission to give in orbit demonstrations of cost effective technologies that can be used to obser ve, capture and dispose of space debris. The craft was launched to the ISS on the 2nd of April 2018, on board a Dragon capsule. From here the satellite was deployed via the NanoRacks Kaber system into an orbit at 405km altitude and has performed key technology demonstrations including the use of a net, a harpoon, vision-based navigation (VBN) and a dragsail in a realistic space operational environment. Two CubeSats have been released by the main platform and used as targets for the net demonstration and for the VBN, whereas the harpoon demonstration has used a target mounted at the end of a boom deployed from the platform. These have been the first ever in-orbit successful demonstrations of technologies for large space debris capture. The dragsail demonstration presented some anomalies, however the lessons learned have already been implemented in new successful dragsails already deployed in space missions. This paper briefly outlines the development of the mission, discussing some of its challenges, and focusses on the various in orbit experiments, describing the operations and overall outcomes

    Local Competition Between Foraging Relatives: Growth and Survival of Bruchid Beetle Larvae

    Kin selection theory states that when resources are limited and all else is equal, individuals will direct competition away from kin. However, when competition between relatives is completely local, as is the case in granivorous insects whose larval stages spend their lives within a single seed, this can reduce or even negate the kin-selected benefits. Instead, an increase in competition may have the same detrimental effects on individuals that forage with kin as those that forage with non-kin. In a factorial experiment we assessed the effects of relatedness and competition over food on the survival and on fitness-related traits of the bruchid beetle Callosobruchus maculatus. Relatedness of competitors did not affect the survival of larvae. Larval survival substantially decreased with increasing larval density, and we found evidence that beetles maturing at a larger size were more adversely affected by competition, resulting in lower survival rates. Furthermore, females showed a reduction in their growth rate with increasing larval density, emerging smaller after the same development time. Males increased their growth rate, emerging earlier but at a similar size when food was more limited. Our results add to the growing number of studies that fail to show a relationship between relatedness and a reduction in competition between relatives in closed systems, and emphasize the importance of the scale at which competition between relatives occurs
