    Tropical coastal habitats as surrogates of fish community structure, grazing, and fisheries value

    Habitat maps are frequently invoked as surrogates of biodiversity to aid the design of networks of marine reserves. Maps are used to maximize habitat heterogeneity in reserves because this is likely to maximize the number of species protected. However, the technique's efficacy is limited by intra-habitat variability in the species present and their abundances. Although communities are expected to vary among patches of the same habitat, this variability is poorly documented and rarely incorporated into reserve planning. To examine intra-habitat variability in coral-reef fishes, we generated a data set from eight tropical coastal habitats and six islands in the Bahamian archipelago using underwater visual censuses. Firstly, we provide further support for habitat heterogeneity as a surrogate of biodiversity as each predefined habitat type supported a distinct assemblage of fishes. Intrahabitat variability in fish community structure at scales of hundreds of kilometers (among islands) was significant in at least 75% of the habitats studied, depending on whether presence/absence, density, or biomass data were used. Intra-habitat variability was positively correlated with the mean number of species in that habitat when density and biomass data were used. Such relationships provide a proxy for the assessment of intra-habitat variability when detailed quantitative data are scarce. Intra-habitat variability was examined in more detail for one habitat (forereefs visually dominated by Montastraea corals). Variability in community structure among islands was driven by small, demersal families (e. g., territorial pomacentrid and labrid fishes). Finally, we examined the ecological and economic significance of intra-habitat variability in fish assemblages on Montastraea reefs by identifying how this variability affects the composition and abundances of fishes in different functional groups, the key ecosystem process of parrotfish grazing, and the ecosystem service of value of commercially important finfish. There were significant differences in a range of functional groups and grazing, but not fisheries value. Variability at the scale of tens of kilometers (among reefs around an island) was less than that among islands. Caribbean marine reserves should be replicated at scales of hundreds of kilometers, particularly for species-rich habitats, to capture important intra-habitat variability in community structure, function, and an ecosystem process

    SMG-1 and mTORC1 Act Antagonistically to Regulate Response to Injury and Growth in Planarians

    Planarian flatworms are able to both regenerate their whole bodies and continuously adapt their size to nutrient status. Tight control of stem cell proliferation and differentiation during these processes is the key feature of planarian biology. Here we show that the planarian homolog of the phosphoinositide 3-kinase-related kinase (PIKK) family member SMG-1 and mTOR complex 1 components are required for this tight control. Loss of smg-1 results in a hyper-responsiveness to injury and growth and the formation of regenerative blastemas that remain undifferentiated and that lead to lethal ectopic outgrowths. Invasive stem cell hyper-proliferation, hyperplasia, hypertrophy, and differentiation defects are hallmarks of this uncontrolled growth. These data imply a previously unappreciated and novel physiological function for this PIKK family member. In contrast we found that planarian members of the mTOR complex 1, tor and raptor, are required for the initial response to injury and blastema formation. Double smg-1 RNAi experiments with tor or raptor show that abnormal growth requires mTOR signalling. We also found that the macrolide rapamycin, a natural compound inhibitor of mTORC1, is able to increase the survival rate of smg-1 RNAi animals by decreasing cell proliferation. Our findings support a model where Smg-1 acts as a novel regulator of both the response to injury and growth control mechanisms. Our data suggest the possibility that this may be by suppressing mTOR signalling. Characterisation of both the planarian mTORC1 signalling components and another PIKK family member as key regulators of regeneration and growth will influence future work on regeneration, growth control, and the development of anti-cancer therapies that target mTOR signalling

    Many Labs 5:Testing pre-data collection peer review as an intervention to increase replicability

    Replication studies in psychological science sometimes fail to reproduce prior findings. If these studies use methods that are unfaithful to the original study or ineffective in eliciting the phenomenon of interest, then a failure to replicate may be a failure of the protocol rather than a challenge to the original finding. Formal pre-data-collection peer review by experts may address shortcomings and increase replicability rates. We selected 10 replication studies from the Reproducibility Project: Psychology (RP:P; Open Science Collaboration, 2015) for which the original authors had expressed concerns about the replication designs before data collection; only one of these studies had yielded a statistically significant effect (p < .05). Commenters suggested that lack of adherence to expert review and low-powered tests were the reasons that most of these RP:P studies failed to replicate the original effects. We revised the replication protocols and received formal peer review prior to conducting new replication studies. We administered the RP:P and revised protocols in multiple laboratories (median number of laboratories per original study = 6.5, range = 3?9; median total sample = 1,279.5, range = 276?3,512) for high-powered tests of each original finding with both protocols. Overall, following the preregistered analysis plan, we found that the revised protocols produced effect sizes similar to those of the RP:P protocols (?r = .002 or .014, depending on analytic approach). The median effect size for the revised protocols (r = .05) was similar to that of the RP:P protocols (r = .04) and the original RP:P replications (r = .11), and smaller than that of the original studies (r = .37). Analysis of the cumulative evidence across the original studies and the corresponding three replication attempts provided very precise estimates of the 10 tested effects and indicated that their effect sizes (median r = .07, range = .00?.15) were 78% smaller, on average, than the original effect sizes (median r = .37, range = .19?.50)

    Continuous, on-line DNA sequencing using oligodeoxynucleotide primers with multiple fluorophores

    A method for sequencing DNA by using a difluoresceinated primer and laser excitation is described. Dideoxy protocols have been determined that provide sequences for 600 bases starting with base 1 with \u3c1% error in a single load. Electrophoresis is at 20 W and the bands are detected 24 cm from the bottom of the loading well with a scanning fluorescence detector. Bands are imaged on a TV screen in two dimensions. The sequences can be read from the TV screen manually or semiautomatically by using a simple software program. The system allows more bases to be read with a lower error rate than any other reported automated sequencing method

    Sustainable Finance Analysis for the South Berry Islands Marine Reserve

    Because of interest in assisting the implementation of active management in the South Berry Islands Marine Reserve (SBIMR), we conducted a review of the sustainable finance literature for protected areas to highlight general guidelines and options for sustainable financing of the reserve. We then focused in on sustainable finance mechanisms that are of particular relevance to marine protected areas and the Caribbean region, and evaluated which of these offer the most potential for application in The Bahamas and especially the SBIMR. According to estimates derived from the Draft Management Plan for the SBIMR, active management of the reserve will require $1.8 million per year. Many potential options or mechanisms meeting this funding requirement are discussed in this report. In this summary, we focus on key options and mechanisms analyzed in the final section of this report

    We conducted a research study of the economic benefits (and costs) of the South Berry Island Marine Reserve to inform ongoing management planning for the reserve. 'Our approach combined market, non-market, and non-use valuation methods, and utilized both primary data collection and value transfer from existing sources of information'. The economic results of the research can be broken into two main categories. First, there are the current benefits to stakeholders from the habitats and species of the marine environment of the South Berry Islands. Second, there are the specific projected benefits (and costs) to stakeholders because of changes from active management of the reserve in their use of these habitats and species

    Adult attachment and the perception of emotional expressions: Probing the hyperactivating strategies underlying anxious attachment

    ABSTRACT According to adult attachment theory, individual differences in attachment-related anxiety reflect variation in individuals&apos; vigilance to cues relevant to appraising and monitoring the availability and responsiveness of significant others. To investigate this assumption, the authors adopted a morph movie paradigm in which participants were shown movies of faces in which an emotional facial expression changed gradually to a neutral one (Study 1) or a neutral expression changed to an emotional one (Studies 2-4). Participants were asked to judge the point at which the emotional expression had disappeared or emerged, respectively. Individuals who were highly anxious with respect to attachment were more likely to perceive the offset (Study 1) as well as the onset (Studies 2 and 3) of the facial expressions of emotion earlier than other people.
