    Contrast-enhanced ultrasound: clinical applications in patients with atherosclerosis

    Contrast-enhanced ultrasound (CEUS) is increasingly being used to evaluate patients with known or suspected atherosclerosis. The administration of a microbubble contrast agent in conjunction with ultrasound results in an improved image quality and provides information that cannot be assessed with standard B-mode ultrasound. CEUS is a high-resolution, noninvasive imaging modality, which is safe and may benefit patients with coronary, carotid, or aortic atherosclerosis. CEUS allows a reliable assessment of endocardial borders, left ventricular function, intracardiac thrombus and myocardial perfusion. CEUS results in an improved detection of carotid atherosclerosis, and allows assessment of high-risk plaque characteristics including intraplaque vascularization, and ulceration. CEUS provides real-time bedside information in patients with a suspected or known abdominal aortic aneurysm or aortic dissection. The absence of ionizing radiation and safety of the contrast agent allow repetitive imaging which is particularly useful in the follow-up of patients after endovascular aneurysm repair. New developments in CEUS-based molecular imaging will improve the understanding of the pathophysiology of atherosclerosis and may in the future allow to image and directly treat cardiovascular diseases (theragnostic CEUS). Familiarity with the strengths and limitations of CEUS may have a major impact on the management of patients with atherosclerosis

    On the effect of prandtl number to subgrid-scale heat flux models

    Estimations of the grid size and computational cost for direct numerical simulation (DNS) and large-eddy simulation (LES) of Rayleigh-BĂ©nard convection (RBC) are presented in the {Ra, P r} phase space. Computational requirements to reach the so-called asymptotic Kraichnan or ultimate regime of turbulence using DNS are far too expensive. Therefore, we turn to LES to predict the large-scale behavior at very high Ra-numbers. However, a priori alignment studies clearly show why the modelization of the SGS heat flux is the main difficulty that (still) precludes reliable LES of buoyancy-driven flows at (very) high Ra-numbers. This inherent difficulty can be by-passed by carrying out simulations at low-P r numbers where no SGS heat flux activity is expected. This opens the possibility to reach the ultimate regime carrying out LES of RBC at low-P r using meshes of 1010 -1011 grid points.     Left-ventricular outflow tract acceleration time is associated with symptoms in patients with obstructive hypertrophic cardiomyopathy

    AIMS: Not all obstructive hypertrophic cardiomyopathy (HCM) patients are symptomatic. The relation between obstructive HCM and symptoms is not well understood. The hypothesis of this study is that left-ventricular outflow tract (LVOT) acceleration time (AT) is associated with symptoms. METHODS: We included 187 patients (61% men, mean age 55 ± 14 years) with obstructive HCM, defined as a maximal wall thickness ≥ 15 mm and a resting or provoked LVOT peak gradient ≥ 30 mmHg. Peak velocity (PV), left-ventricular (LV) ejection time (ET), and AT (the time between LVOT flow onset and the moment of PV) were measured on continuous-wave (CW) Doppler tracings. Logistic and Cox proportional hazard regression analyses were used to evaluate the relation between symptoms [New York Heart Association (NYHA) class ≥ II] and echocardiographic measurements, including AT. Reproducibility was assessed using the intraclass correlation coefficient (ICC). RESULTS: Symptomatic patients were more often female and had higher mean AT values. Logistic regression demonstrated a significant association between AT and symptomatic status (odds ratio 1.31 per 10 ms, p < 0.01) after adjustment for sex, negative inotropes, PV, LVOT diameter, and diastolic dysfunction. AT was independently associated with symptoms and septal reduction during follow-up (hazard ratio 1.09 per 10 ms, p < 0.05). The ICC was 0.98 with a mean difference of 0.28 ± 8.4 ms. CONCLUSION: In obstructive HCM patients, increased AT is significantly related to symptoms after adjustment for sex, negative inotropes, PV, LVOT diameter, and diastolic dysfunction, and is associated with the symptomatic status during follow-up. AT represents an easily measured echocardiographic variable with excellent inter-reader reproducibility.     Evaluation of residual stress and texture in isotope based (MgB2)-B-11 superconductor using neutron diffraction

    Magnesium diboride (MgB2) superconducting wires have demonstrated commercial potential to replace niobium-titanium (NbTi) in terms of comparable critical current density. Its higher critical temperature makes MgB2 wire suitable for liquid-helium-free operation. We recently reported boron-11 isotope-based low-activation Mg11B2 superconducting wire with decent critical current density appropriate for low-cost superconducting fusion magnets. In this study, we have mainly focused on the neutron diffraction technique to measure the residual stress in Mg11B2 superconducting wire for the first time. The residual stress state was given qualitative and quantitative interpretation in terms of micro- and macrostress generation mechanisms based on the isotropic model confirmed by neutron texture measurements. The relationship between the stress/strain state in the wire and the transport critical current density is also discussed. This investigation could pave the way to further enhancement of the critical current density of low-activation Mg11B2 superconducting wires suitable for next-generation fusion grade magnets

    Seasonal and interannual effects of hypoxia on fish habitat quality in central Lake Erie

    1. Hypoxia occurs seasonally in many stratified coastal marine and freshwater ecosystems when bottom dissolved oxygen (DO) concentrations are depleted below 2–3 mg O2 L-1. 2. We evaluated the effects of hypoxia on fish habitat quality in the central basin of Lake Erie from 1987 to 2005, using bioenergetic growth rate potential (GRP) as a proxy for habitat quality. We compared the effect of hypoxia on habitat quality of (i) rainbow smelt, Osmerus mordax mordax Mitchill (young-of-year, YOY, and adult), a cold-water planktivore, (ii) emerald shiner, Notropis atherinoides Rafinesque (adult), a warm-water planktivore, (iii) yellow perch, Perca flavescens Mitchill (YOY and adult), a cool-water benthopelagic omnivore and (iv) round goby Neogobius melanostomus Pallas (adult) a eurythermal benthivore. Annual thermal and DO profiles were generated from 1D thermal and DO hydrodynamics models developed for Lake Erie’s central basin. 3. Hypoxia occurred annually, typically from mid-July to mid-October, which spatially and temporally overlaps with otherwise high benthic habitat quality. Hypoxia reduced the habitat quality across fish species and life stages, but the magnitude of the reduction varied both among and within species because of the differences in tolerance to low DO levels and warm-water temperatures. 4. Across years, trends in habitat quality mirrored trends in phosphorus concentration and water column oxygen demand in central Lake Erie. The per cent reduction in habitat quality owing to hypoxia was greatest for adult rainbow smelt and round goby (mean: -35%), followed by adult emerald shiner (mean: -12%), YOY rainbow smelt (mean: -10%) and YOY and adult yellow perch (mean: -8.5%). 5. Our results highlight the importance of differential spatiotemporally interactive effects of DO and temperature on relative fish habitat quality and quantity. These effects have the potential to influence the performance of individual fish species as well as population dynamics, trophic interactions and fish community structure

    Analysis of Prototype Foamy Virus particle-host cell interaction with autofluorescent retroviral particles

    <p>Abstract</p> <p>Background</p> <p>The foamy virus (FV) replication cycle displays several unique features, which set them apart from orthoretroviruses. First, like other B/D type orthoretroviruses, FV capsids preassemble at the centrosome, but more similar to hepadnaviruses, FV budding is strictly dependent on cognate viral glycoprotein coexpression. Second, the unusually broad host range of FV is thought to be due to use of a very common entry receptor present on host cell plasma membranes, because all cell lines tested in vitro so far are permissive.</p> <p>Results</p> <p>In order to take advantage of modern fluorescent microscopy techniques to study FV replication, we have created FV Gag proteins bearing a variety of protein tags and evaluated these for their ability to support various steps of FV replication. Addition of even small N-terminal HA-tags to FV Gag severely impaired FV particle release. For example, release was completely abrogated by an N-terminal autofluorescent protein (AFP) fusion, despite apparently normal intracellular capsid assembly. In contrast, C-terminal Gag-tags had only minor effects on particle assembly, egress and particle morphogenesis. The infectivity of C-terminal capsid-tagged FV vector particles was reduced up to 100-fold in comparison to wild type; however, infectivity was rescued by coexpression of wild type Gag and assembly of mixed particles. Specific dose-dependent binding of fluorescent FV particles to target cells was demonstrated in an Env-dependent manner, but not binding to target cell-extracted- or synthetic- lipids. Screening of target cells of various origins resulted in the identification of two cell lines, a human erythroid precursor- and a zebrafish- cell line, resistant to FV Env-mediated FV- and HIV-vector transduction.</p> <p>Conclusions</p> <p>We have established functional, autofluorescent foamy viral particles as a valuable new tool to study FV - host cell interactions using modern fluorescent imaging techniques. Furthermore, we succeeded for the first time in identifying two cell lines resistant to Prototype Foamy Virus Env-mediated gene transfer. Interestingly, both cell lines still displayed FV Env-dependent attachment of fluorescent retroviral particles, implying a post-binding block potentially due to lack of putative FV entry cofactors. These cell lines might ultimately lead to the identification of the currently unknown ubiquitous cellular entry receptor(s) of FVs.</p

    A dinâmica e o construto territorial da região noroeste do estado do Rio Grande do Sul

    Este estudo busca contribuir com as discussões de diferentes perspectivas em relação à ótica do desenvolvimento regional com o enfoque territorial. Para isto, discute brevemente as definições de urbano, cidade, cidade-campo e territorialidades dentro da lógica global, bem como a própria definição de território. Apresenta-se uma breve contextualização sobre a Região Noroeste do Estado do Rio Grande do Sul e sua dinâmica de formação do território e de desenvolvimento regional dentro do contexto global do capital, baseada na monocultura da produção de soja, que contribuiu para o desenvolvimento da região através dos estímulos que proporcionou à modernização tecnológica da agropecuária regional na década de 1970 e os impulsos decorrentes para a expansão de atividades industriais e de serviços. No entanto, a dinâmica regional não pode estar subordinada a uma base produtiva monocultural, no caso, a produção de soja, pois nada garante que no futuro esta cultura continuará produzindo impactos relevantes na economia da Região
