    Gene flow of common ash (<em>Fraxinus excelsior</em> L.) in a fragmented landscape

    Gene flow dynamics of common ash (Fraxinus excelsior L.) is affected by several human activities in Central Europe, including habitat fragmentation, agroforestry expansion, controlled and uncontrolled transfer of reproductive material, and a recently introduced emerging infectious disease, ash dieback, caused by Hymenoscyphus fraxineus. Habitat fragmentation may alter genetic connectivity and effective population size, leading to loss of genetic diversity and increased inbreeding in ash populations. Gene flow from cultivated trees in landscapes close to their native counterparts may also influence the adaptability of future generations. The devastating effects of ash dieback have already been observed in both natural and managed populations in continental Europe. However, potential long-term effects of genetic bottlenecks depend on gene flow across fragmented landscapes. For this reason, we studied the genetic connectivity of ash trees in an isolated forest patch of a fragmented landscape in Rösenbeck, Germany. We applied two approaches to parentage analysis to estimate gene flow patterns at the study site. We specifically investigated the presence of background pollination at the landscape level and the degree of genetic isolation between native and cultivated trees. Local meteorological data was utilized to understand the effect of wind on the pollen and seed dispersal patterns. Gender information of the adult trees was considered for calculating the dispersal distances. We found that the majority of the studied seeds (55-64%) and seedlings (75-98%) in the forest patch were fathered and mothered by the trees within the same patch. However, we determined a considerable amount of pollen flow (26-45%) from outside of the study site, representing background pollination at the landscape level. Limited pollen flow was observed from neighbouring cultivated trees (2%). Both pollen and seeds were dispersed in all directions in accordance with the local wind directions. Whereas there was no positive correlation between pollen dispersal distance and wind speed, the correlation between seed dispersal distance and wind speed was significant (0.71, p < 0.001), indicating that strong wind favours long-distance dispersal of ash seeds.     The Protective Effect of Kefir and Vitamin C on Azoxymethane Induced Toxicity and Induction of Metallothionein in Mice

    The present study was conducted to investigate whether vitamin C or kefir (a milk-based fermentation  product) protected Swiss albino mice from azoxymethane (AOM) toxicity. We also investigated the effect  of AOM administration on the induction of metallothionein (MT) expression in mice tissues. 40 12-weekold  male/female (20:20) Swiss albino mice with a mean weight of 31.4 g were allocated into four groups.  Animals in the first group were the control group. Animals in the other three groups were treated with  AOM (5mg/kg body weight) subcutaneously twice weekly for a total of 7 weeks. Animals in the second  group were treated only with AOM. Those in the third group were allowed access to kefir (50% wt/vol) ad  libitum. Those in the fourth group received vitamin C subcutaneously (500 mg/kg) per day for 7 weeks. Six  weeks after the final AOM treatment, all animals were sacrificed and necropsied. AOM administration  caused severe liver lesions including enlarged hepatocytes (megalocytes) and many contained enlarged  nuclei. Vitamin C and kefir administration clearly reduced the severity of AOM induced liver lesions.  Induction of MT expression was observed in the liver and kidneys, particularly in the centrilobular zones  and renal cortex, mainly in the distal renal tubules, collecting tubules, Henle's loop, and medulla, respectively.  In conclusion, vitamin C and kefir supplementation were found to be able to reduce the severity of  hepatotoxic lesions.,

    Muscle dysmorphia and associated psychological features of males in the Middle East: A systematic review

    Muscle dysmorphia (MD) is a mental health disorder characterized by a preoccupation with muscularity and is linked to excessive exercise and dieting, and using anabolic steroids. There is a global interest in the study of MD, however, the literature has yet to collect and synthesize the evidence base in the Middle East. The aim of this systematic review was therefore to understand the status of MD and its associated psychological features of males in the Middle East. A systematic search in PubMed, Web of Science, and Scopus was performed on February 4th, 2023 to identify the cross-sectional survey-based studies conducted in the ME using well-established assessments directly related to MD and its associated psychological features in males. The risk of bias was assessed using the Risk of Bias Instrument for Cross-Sectional Surveys of Attitudes and Practices. A total of ten eligible studies were included in this review. These studies were conducted in Turkey (n = 5), Iran (n = 2), Lebanon (n = 1), Kuwait (n = 1), and North Cyprus (n = 1). Participants in the included studies were mainly bodybuilders (n = 1958, 55.4%) and university students (n = 1510, 42.7%). Results suggest that disordered eating attitudes, obsession with healthy eating, perfectionism, vulnerable narcissistic disposition, low self-esteem, muscle dissatisfaction, need frustration, and low family income may contribute to or trigger MD symptoms in bodybuilders and male university students in the Middle East. Healthcare professionals should be mindful of the associated psychological correlates in treating MD symptoms in men living in the Middle East.publishedVersio

    Diabetology and rheumatology have much in common, including many biochemical features. Diabetic cases suffer excessively from common musculoskeletal conditions. The present study aimed to explore the association between the caffeic acid phenethyl ester (CAPE) treatment and the standard parameters of diabetic and rheumatologic controls. Serum anti-streptolysin-O (ASO), C-reactive protein (CRP), rheumatoid factor (RF) titers and glucose (GLU), cholesterol (CHOL), triglyceride (TG), high density lipoprotein (HDL), very low density lipoprotein (VLDL), alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), sodium (Na), potassium (K), chloride (Cl), calcium (Ca), phosphorus (P), uric acid (UA), iron (Fe), iron-binding capacity (IBC), total iron-binding capacity (TIBC) and superoxide dismutase (SOD) levels were measured in 22 female Wistar-Albino rats. The animals were divided into three groups: group 1: non-diabetic rats as control; group 2: streptozotocin (STZ)-induced, untreated diabetic rats; and group 3: STZ-induced, CAPE-treated diabetic rats. Levels of GLU were increased in group 2. ASO values were decreased in group 3. TG and VLDL levels were increased in group 3 compared with the group 1. Higher UA levels were detected in group 3 compared with group 2. The levels of Fe and TIBC were decreased in group 3. The results of the present study indicate the reducing effect of CAPE on the serum levels of diabetic and rheumatologic parameters in diabetic rats.Dijabetologija i reumatologija imaju mnoge sličnosti, uključujući biokemijske procese. Oboljeli od dijabetesa pate od teških stanja mišićno-koštanog sustava. U ovom radu istražena je povezanost između liječenja fenetil esterom kavene kiseline i standardnih pokazatelja koji se kontroliraju pri dijabetičnim i reumatoidnim bolestima. Kod 22 Wistar-Albino štakorice mjereni su: serumski anti-streptolizin-O (ASO), C-reaktivni protein (CRP), titar reumatoidnog faktora (RF), zatim razina glukoze (GLU), kolesterola (CHOL), triglicerida (TG), lipoproteina visoke gustoće (HDL), lipoproteina vrlo niske gustoće (VLDL), alkalne fosfataze (ALP), gamaglutamil transferaze (GGT), natrija (Na), kalija (K), klorida (Cl), kalcija (Ca), fosfora (P), mokraćne kiseline (UA), željeza (Fe), kapaciteta vezanja željeza (IBC), ukupnog kapaciteta vezanja željeza (TIBC) i superoksid dismutaze (SOD). Životinje su bile podijeljene u tri skupine: 1. štakorice bez dijabetesa (kontrolna skupina), 2. neliječene štakorice s dijabetesom izazvanim streptozotocinom (STZ) i 3. štakorice sa streptozotocin (STZ) - izazvanim dijabetesom liječenim fenetil esterom kavene kiseline. Razine GLU bile su povećane u skupini 2. ASO vrijednosti bile su snižene u skupini 3. Razine TG i VLDL bile su povišene u skupini 3 u usporedbi s razinama u skupini 1. Više razine UA utvrđene su u skupini 3 u usporedbi sa skupinom 2. Razine Fe i TIBC bile su snižene u skupini 3. Rezultati pokazuju reducirajuće učinke fenetil estera kavene kiseline na razinu dijabetičnih i reumatoloških pokazatelja u serumu štakorica sa šećernom bolešću izazvanom streptozotocinom

    Dizajn maske za lice s UVC-LED zraÄŤenjem i njezina djelotvornost protiv uobiÄŤajenih klica

    During the Covid-19 pandemic, one of the best means of personal protection was using face masks. In this context, the World Health Organization has declared the attempts to produce masks inactivating airborne virus species a welcome initiative. This preliminary study aimed to prove that airborne germs passing through a mask filter cartridge can be destroyed by the rays emitted from UVC LEDs placed in such cartridge. We therefore designed such a face mask and tested the efficiency of UVC LEDs placed in its cartridge against common contaminants, gram-positive Staphylococcus aureus, gram-negative Pseudomonas aeruginosa, and the influenza A/Puerto Rico/8/1934 virus because of its similarity with SARS CoV-2. Eight UVC LEDs with a total power of 75 mW provided sufficient germicidal effect for all three germs. In terms of safety, ozone production released during UVC LED emission was negligible. Our findings are promising, as they show that well-designed UVC-based face masks can be effective against airborne germs, but further research on a greater sample may help us learn more and optimise such face masks.Tijekom pandemije Covida-19 jedan od najboljih oblika osobne zaštite bilo je nošenje maski za lice. U tom je smislu Svjetska zdravstvena organizacija pozdravila pokušaje izrade maski koje ubijaju virusne vrste koje se prenose zrakom. Cilj je ovoga preliminarnog istraživanja bio dokazati da se zrakom nošene klice koje prolaze kroz filtarske uloške mogu uništiti zračenjem UVC ledica smještenih u takve uloške. Stoga smo osmislili masku za lice s tom namjenom i iskušali djelotvornost UVC ledica protiv uobičajenih izvora zaraza: gram-pozitivnoga Staphylococcus aureus, gram-negativnoga Pseudomonas aeruginosa i virusa influence A/Puerto Rico/8/1934 zbog njegove sličnosti s virusom SARS CoV-2. Osam UVC ledica ukupne snage 75 mW iskazale su dovoljan germicidni učinak protiv svih triju klica. U smislu sigurnosti primjene, ozon proizveden tijekom UVC-LED zračenja pokazao se zanemarivim. Naši su rezultati obećavajući jer pokazuju da dobro osmišljene maske za lice s UVC zračenjem mogu biti djelotvorne protiv zrakom nošenih klica, ali će tek daljnja istraživanja na većem uzorku pomoći da doznamo više i usavršimo takve maske za lice

    Formulation Development and Evaluation of Alginate Microspheres of Ibuprofen

    The present study was designed to investigate the effects of different variables on the release profile of ibuprofen microspheres formulated using modified emulsification method. Eight batches of microspheres (F1-F8) were prepared by applying 23 factorial design. The amount of sodium alginate, amount of calcium chloride, and amount of magnesium stearate were selected as formulation variables. All the batches were evaluated in terms of percentage yield, percentage encapsulation efficiency and in vitro release characteristics. The batch F7 was found to be optimum batch and was further characterized via scanning electron microscopy (SEM) and particle size analysis. Multiple linear regression was applied to confirm significant effect of each variable on release characteristics. The model developed in the present study can be effectively utilized to achieve the formulation with desired release characteristics

    A Cross-cultural Examination of Preferences for Work Attributes

    Purpose – The purpose of this paper is to examine the relationship between individual- and country-level values and preferences for job/organizational attributes. Design/methodology/approach – Survey data were collected from 475 full-time employees (average of nine years work experience, and three years in a managerial position) enrolled in part-time MBA programs in seven countries. Findings – Preference for a harmonious workplace is positively related to horizontal collectivism, whereas preference for remuneration/advancement is positively related to vertical individualism. The authors also find a positive relationship between preference for meaningful work and horizontal individualism, and between preference for employer prestige and social adjustment (SA) needs. Research limitations/implications – Although the sample comprised experienced, full-time professionals, using graduate business students may limit generalizability. Overall, the results provide initial support for the utility of incorporating the multi-dimensional individualism and collectivism measure, as well as SA needs, when assessing the relationships between values and employee preferences. Practical implications – For practitioners, the primary conclusion is that making assumptions about preferences based on nationality is risky. Findings may also prove useful for enhancing person-organization fit and the ability to attract and retain qualified workers. Originality/value – This study extends research on workers’ preferences by incorporating a new set of values and sampling experienced workers in a range of cultural contexts

    MIR376A is a regulator of starvation-induced autophagy

    Background: Autophagy is a vesicular trafficking process responsible for the degradation of long-lived, misfolded or abnormal proteins, as well as damaged or surplus organelles. Abnormalities of the autophagic activity may result in the accumulation of protein aggregates, organelle dysfunction, and autophagy disorders were associated with various diseases. Hence, mechanisms of autophagy regulation are under exploration. Methods: Over-expression of hsa-miR-376a1 (shortly MIR376A) was performed to evaluate its effects on autophagy. Autophagy-related targets of the miRNA were predicted using Microcosm Targets and MIRanda bioinformatics tools and experimentally validated. Endogenous miRNA was blocked using antagomirs and the effects on target expression and autophagy were analyzed. Luciferase tests were performed to confirm that 3’ UTR sequences in target genes were functional. Differential expression of MIR376A and the related MIR376B was compared using TaqMan quantitative PCR. Results: Here, we demonstrated that, a microRNA (miRNA) from the DlkI/Gtl2 gene cluster, MIR376A, played an important role in autophagy regulation. We showed that, amino acid and serum starvation-induced autophagy was blocked by MIR376A overexpression in MCF-7 and Huh-7 cells. MIR376A shared the same seed sequence and had overlapping targets with MIR376B, and similarly blocked the expression of key autophagy proteins ATG4C and BECN1 (Beclin 1). Indeed, 3’ UTR sequences in the mRNA of these autophagy proteins were responsive to MIR376A in luciferase assays. Antagomir tests showed that, endogenous MIR376A was participating to the control of ATG4C and BECN1 transcript and protein levels. Moreover, blockage of endogenous MIR376A accelerated starvation-induced autophagic activity. Interestingly, MIR376A and MIR376B levels were increased with different kinetics in response to starvation stress and tissue-specific level differences were also observed, pointing out to an overlapping but miRNA-specific biological role. Conclusions: Our findings underline the importance of miRNAs encoded by the DlkI/Gtl2 gene cluster in stress-response control mechanisms, and introduce MIR376A as a new regulator of autophagy
