    Studies on the Restriction of Murine Leukemia Viruses by Mouse APOBEC3

    APOBEC3 proteins function to restrict the replication of retroviruses. One mechanism of this restriction is deamination of cytidines to uridines in (−) strand DNA, resulting in hypermutation of guanosines to adenosines in viral (+) strands. However, Moloney murine leukemia virus (MoMLV) is partially resistant to restriction by mouse APOBEC3 (mA3) and virtually completely resistant to mA3-induced hypermutation. In contrast, the sequences of MLV genomes that are in mouse DNA suggest that they were susceptible to mA3-induced deamination when they infected the mouse germline. We tested the possibility that sensitivity to mA3 restriction and to deamination resides in the viral gag gene. We generated a chimeric MLV in which the gag gene was from an endogenous MLV in the mouse germline, while the remainder of the viral genome was from MoMLV. This chimera was fully infectious but its response to mA3 was indistinguishable from that of MoMLV. Thus, the Gag protein does not seem to control the sensitivity of MLVs to mA3. We also found that MLVs inactivated by mA3 do not synthesize viral DNA upon infection; thus mA3 restriction of MLV occurs before or at reverse transcription. In contrast, HIV-1 restricted by mA3 and MLVs restricted by human APOBEC3G do synthesize DNA; these DNAs exhibit APOBEC3-induced hypermutation

    Cross-cutting principles for planetary health education

    Since the 2015 launch of the Rockefeller Foundation Lancet Commission on planetary health,1 an enormous groundswell of interest in planetary health education has emerged across many disciplines, institutions, and geographical regions. Advancing these global efforts in planetary health education will equip the next generation of scholars to address crucial questions in this emerging field and support the development of a community of practice. To provide a foundation for the growing interest and efforts in this field, the Planetary Health Alliance has facilitated the first attempt to create a set of principles for planetary health education that intersect education at all levels, across all scales, and in all regions of the world—ie, a set of cross-cutting principles

    WHO global research priorities for antimicrobial resistance in human health

    The WHO research agenda for antimicrobial resistance (AMR) in human health has identified 40 research priorities to be addressed by the year 2030. These priorities focus on bacterial and fungal pathogens of crucial importance in addressing AMR, including drug-resistant pathogens causing tuberculosis. These research priorities encompass the entire people-centred journey, covering prevention, diagnosis, and treatment of antimicrobial-resistant infections, in addition to addressing the overarching knowledge gaps in AMR epidemiology, burden and drivers, policies and regulations, and awareness and education. The research priorities were identified through a multistage process, starting with a comprehensive scoping review of knowledge gaps, with expert inputs gathered through a survey and open call. The priority setting involved a rigorous modified Child Health and Nutrition Research Initiative approach, ensuring global representation and applicability of the findings. The ultimate goal of this research agenda is to encourage research and investment in the generation of evidence to better understand AMR dynamics and facilitate policy translation for reducing the burden and consequences of AMR

    IntroductionThe use of controlled substances like cocaine increases the risk of cardiovascular disease (CVD) and myocardial infarction (MI). However, outside of alcohol and tobacco, substance use is not included in CVD risk assessment tools. We identified the effects of using multiple substances (nicotine/cotinine, cannabis, alcohol, cocaine, methamphetamine, heroin and other opioids) on cardiac injury measured by high-sensitivity troponin (hsTnI) in homeless and unstably housed women.MethodsWe recruited 245 homeless and unstably housed women from shelters, free meal programs and street encampments. Participants completed six monthly study visits. Adjusting for traditional CVD risk factors, we examined longitudinal associations between substance use and hsTnI.ResultsMedian participant age was 53 years and 74 % were ethnic minority women. At baseline, 76 % of participants had hypertension, 31 % were HIV-positive, 8% had a history of a prior MI and 12 % of prior stroke. The most commonly used substances were cotinine/nicotine (80 %), cannabis (68 %) and cocaine (66 %). HsTnI exceeding the 99th percentile (14.7 ng/L) - a level high enough to signal possible MI - was observed in 14 participants during >1 study visit (6%). In adjusted analysis, cocaethylene and fentanyl were significantly associated with higher hsTnI levels.ConclusionsFentanyl use and the co-use of cocaine and alcohol are associated with myocardial injury, suggesting that the use of these substances may act as long-term cardiac insults. Whether risk counseling on these specific substances and/or including their use in CVD risk stratification would improve CVD outcomes in populations where substance use is high merits further investigation

    Resume - Le Leg 120 du programme international de forages oceaniques (Ocean Drilling Project) a permis de determiner la nature et l\u27age du plateau de Kerguelen et de decrire son evolution depuis Ie Cretace. Le socle, fore aux sites 747, 749 ct 750, est compose de coulees basaltiqucs qui correspondent a des tholeiites transitionnelles saturees en silice. Ces basaltes se sont mis en place dans des conditions aeriennes ou sub-aeriennes avant Ie Cenomanien sur une surface quasiment horizontalc. Les premiers sediments, d\u27age Cenomanien, se sont deposes en milieu fluviatile. Au Campanien et a la fin du Maestrichtien Ie plateau a subside rapidement et a ete affecte par une phase tectonique extensive majeure entre 75 et 68 M.a. Ces deux evenernents marquent sans doute la separation du domaine sud du plateau de Kerguelen et de I\u27ensemble Broken Ridge-Diamantina Zone. Abstract - Leg 120 of the Ocean Drilling Program was devoted to studying the nature and evolution of the Central and Southern Kerguelen Plateau. The basement drilled at sites 747, 749 and 750 is composed of basaltic flows which are silica-saturated transitional tholeiites. These basalts were erupted prior to the Cenomanian under subaerial or shallow marine conditions and were emplaced on a near horizontal surface. The early sedimentation during the Cenomanian denotes fluvial conditions. During the Campanian and late Maestrichtian the plateau subsided rapidly and a major tectonic episode occurred between 75 and 68 M.a. These events are possibly related to the breakup between the Southern Kerguelen Plateau and Broken Ridge-Diamantina Zone
