Atypical pulmonary presentation of strongyloides stercoralis hyperinfection in a patient with Philadelphia chromosome-positive acute lympoblastic leukemia
Strongyloides stercoralis is a soil-transmitted helminth endemic to tropical and subtropical regions. Rhabditiform larvae in moist soil can develop into filariform larvae that can penetrate human skin and migrate through the circulation to the lungs.1,2 These larvae escape to the alveolar space and migrate to the pharynx. Larvae can settle in the small intestine and mature into adults afterbeing swallowed in pharyngeal secretion. In immunocompromised patients, the parasite can cause autoinfection with progression to hyperinfection syndrome. Effect of NDP-a-MSH on PPAR-c and –b Expression and Anti-Inflammatory Cytokine Release in Rat Astrocytes and Microglia
Brain inflammation plays a central role in numerous brain pathologies. Microglia and astrocytes are the main effector cells that become activated when an inflammatory process takes place within the central nervous system. a-melanocytestimulating hormone (a-MSH) is a neuropeptide with proven anti-inflammatory properties. It binds with highest affinity to the melanocortin receptor 4 (MC4R), which is present in astrocytes and upon activation triggers anti-inflammatory pathways. The aim of this research was to identify anti-inflammatory mediators that may participate in the
immunomodulatory effects of melanocortins in glial cells. Since peroxisome proliferator-activated receptors (PPARs) have
recently been implicated in the modulation of inflammation, we investigated the effect of an a-MSH analog, [Nle4
, D-Phe7]- a-MSH (NDP-a-MSH), on PPAR-b and PPAR-c gene and protein expression in rat primary astrocytes and microglia. We
initially demonstrated that rat primary microglia express MC4R and showed that treatment with NDP-a-MSH increases
PPAR-c protein levels and strongly decreases PPAR-b levels in both astrocytes and microglia. We also showed that
extracellular signal-regulated kinase 1/2 (ERK1/2)–mediated signaling is partially involved in these effects in a cell-specific fashion. Finally, we showed that NDP-a-MSH stimulates the release of the anti-inflammatory cytokines IL-10 and TGF-b from microglia and astrocytes, respectively. The presented data suggest a role for IL-10 and TGF-b in the protective action of melanocortins and a connection between MC4R pathway and that of the nuclear receptor PPAR-c. This is the first report providing evidence that MC4R is expressed in rat primary microglia and that melanocortins modulate PPAR levels in glial cells. Our findings provide new insights into the mechanisms underlying the activation of glial MC4R and open perspectives for new therapeutic strategies for the treatment of inflammation-mediated brain diseases.Fil: Carniglia, Lila. Efficient isolation on Vero.DogSLAMtag cells and full genome characterization of Dolphin Morbillivirus (DMV) by next generation sequencing
The Dolphin Morbillivirus (DMV) genome from the frst Mediterranean epidemic (1990-\u201992) is the
only cetacean Morbillivirus that has been completely sequenced. Here, we report the frst application
of next generation sequencing (NGS) to morbillivirus infection of aquatic mammals. A viral isolate,
representative of the 2006-\u201908 Mediterranean epidemic (DMV_IZSPLV_2008), efciently grew on
Vero.DogSLAMtag cells and was submitted to whole genome characterization by NGS. The fnal
genome length was 15,673 nucleotides, covering 99.82% of the DMV reference genome. Comparison
of DMV_IZSPLV_2008 and 1990-\u201992 DMV strain sequences revealed 157 nucleotide mutations and
47 amino acid changes. The sequence similarity was 98.7% at the full genome level. Whole-genome
phylogeny suggested that the DMV strain circulating during the 2006-\u201908 epidemics emerged from the
1990-\u201992 DMV strain. Viral isolation is considered the \u201cgold standard\u201d for morbillivirus diagnostics but
efcient propagation of infectious virus is difcult to achieve. The successful cell replication of this strain
allowed performing NGS directly from the viral RNA, without prior PCR amplifcation. We therefore
provide to the scientifc community a second DMV genome, representative of another major outbreak.
Interestingly, genome comparison revealed that the neglected L gene encompasses 74% of the genetic
diversity and might serve as \u201chypervariable\u201d target for strain characterization
Astrocytes: New Targets of Melanocortin 4 Receptor Actions
Astrocytes exert a wide variety of functions with paramount importance in brain physiology. After injury or infection, astrocytes become reactive and they respond by producing a variety of inflammatory mediators that help maintain brain homeostasis. Loss of astrocyte functions as well as their excessive activation can contribute to disease processes; thus, it is important to modulate reactive astrocyte response. Melanocortins are peptides with well-recognized anti-inflammatory and neuroprotective activity. Although melanocortin efficacy was shown in systemic models of inflammatory disease, mechanisms involved in their effects have not yet been fully elucidated. Central anti-inflammatory effects of melanocortins and their mechanisms are even less well known, and, in particular, the effects of melanocortins in glial cells are poorly understood. Of the five known melanocortin receptors (MCRs), only subtype 4 is present in astrocytes. MC4R has been shown to mediate melanocortin effects on energy homeostasis, reproduction, inflammation, and neuroprotection and, recently, to modulate astrocyte functions. In this review, we will describe MC4R involvement in anti-inflammatory, anorexigenic, and anti-apoptotic effects of melanocortins in the brain. We will highlight MC4R action in astrocytes and discuss their possible mechanisms of action. Melanocortin effects on astrocytes provide a new means of treating inflammation, obesity, and neurodegeneration, making them attractive targets for therapeutic interventions in the CNS.Fil: Caruso, Carla Mariana. A barnavirus sequence mined from a transcriptome of the Antarctic pearlwort Colobanthus quitensis.
Because so few viruses in the family Barnaviridae have been reported, we searched for more of them in public sequence databases. Here, we report the complete coding sequence of Colobanthus quitensis associated barnavirus 1, mined from a transcriptome of the Antarctic pearlwort Colobanthus quitensis. The 4.2-kb plus-strand sequence of this virus encompasses four main open reading frames (ORFs), as expected for barnaviruses, including ORFs for a protease-containing polyprotein, an RNA-dependent RNA polymerase whose translation appears to rely on - 1 ribosomal frameshifting, and a capsid protein that is likely to be translated from a subgenomic RNA. The possible derivation of this virus from a fungus associated with C. quitensis is discussed
Simulated global warming affects endophytic bacterial and fungal communities of Antarctic pearlwort leaves and some bacterial isolates support plant growth at low temperatures
Antarctica is one of the most stressful environments for plant life and the Antarctic pearlwort (Colobanthus quitensis) is adapted to the hostile conditions. Plant-associated microorganisms can contribute to plant survival in cold environments, but scarce information is available on the taxonomic structure and functional roles of C. quitensis-associated microbial communities. This study aimed at evaluating the possible impacts of climate warming on the taxonomic structure of C. quitensis endophytes and at investigating the contribution of culturable bacterial endophytes to plant growth at low temperatures. The culture-independent analysis revealed changes in the taxonomic structure of bacterial and fungal communities according to plant growth conditions, such as the collection site and the presence of open-top chambers (OTCs), which can simulate global warming. Plants grown inside OTCs showed lower microbial richness and higher relative abundances of biomarker bacterial genera (Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium, Aeromicrobium, Aureimonas, Hymenobacter, Novosphingobium, Pedobacter, Pseudomonas and Sphingomonas) and fungal genera (Alternaria, Cistella, and Vishniacozyma) compared to plants collected from open areas (OA), as a possible response to global warming simulated by OTCs. Culturable psychrotolerant bacteria of C. quitensis were able to endophytically colonize tomato seedlings and promote shoot growth at low temperatures, suggesting their potential contribution to plant tolerance to cold condition
