The effect of plant domestication on host control of the microbiota
Macroorganisms are colonized by microbial communities that exert important biological and ecological functions, the composition of which is subject to host control and has therefore been described as âan ecosystem on a leashâ. However, domesticated organisms such as crop plants are subject to both artificial selection and natural selection exerted by the agricultural ecosystem. Here, we propose a framework for understanding how host control of the microbiota is influenced by domestication, in which a double leash acts from domesticator to host and host to microbes. Folic acid-functionalized human serum albumin nanocapsules for targeted drug delivery to chronically activated macrophages
Activated synovial macrophages play a key role in Rheumatoid Arthritis (RA). Recent studies have shown that folate receptor beta (FRÎČ) is specifically expressed by activated macrophages. Therefore a folate-based nanodevice would provide the possibility of delivering therapeutic agents to activated macrophages without affecting normal cells and tissues.
This study shows for the first time the sonochemical preparation of HSA nanocapsules avoiding toxic cross linking chemicals and emulsifiers used in other methods.
Production of HSA nanocapsules was optimized leading to a diameter of 443.5 ± 9.0 nm and a narrow size distribution indicated by a polydispersity index (PDI) of 0.066 ± 0.080. Nanocapsules were surface modified with folic acid (FA) and the FA content was determined to be 0.38 and 6.42 molecules FA per molecule HSA, depending on the surplus of FA employed. Dynamic light scattering was used to determine size, PDI and zetapotential of the produced nanocapsules before and after surface modification. FA distribution on the surface of HSA nanocapsules was localized three-dimensionally after fluorescence labeling using confocal laser scanning microscopy (CLSM). Furthermore, specific binding and internalization of HSA nanocapsules by FRÎČ-positive and FRÎČ-negative macrophages, obtained from human peripheral blood mononuclear cells, was demonstrated by flow cytometry. Bamboo fibre processing: insights into hemicellulase and cellulase substrate accessibility
A biological approach for degumming bamboo substrates has been assessed. The ability of various commercially available
enzymes, including cellulase, xylanase, pectinase and mannanase, to hydrolyze bamboo powders was investigated. In addition, a commercial cellulase preparation was applied onto bamboo fibre bundles obtained by natural retting. It was found
that almost all enzymes applied can use bamboo material as a substrate. Mild autoclave pre-treatment can enhance reducing sugar yield from different enzyme treatments. A most pronounced effect was observed with cellulase treatment in which
the hydrolysis degree increased 1.7 fold as measured by reducing sugars for autoclave pre-treated bamboo powders versus
non-treated powders after only a short period of incubation. The combined treatment of hemicellulase preparations showed no effect on the hydrolysis of bamboo substrates. The effect of autoclave pre-treatment on cellulase-treated samples was confirmed by the increase of sugar yield, protein absorption as well as by the enhancement of surface modification and
enzyme penetration observed by CLSM (confocal laser scanning microscopy). This work establishes a base for future

Fishing for MSY: using âpretty good yieldâ ranges without impairing recruitment
Pretty good yield (PGY) is a sustainable fish yield corresponding to obtaining no less than a specified large percentage of the maximum sustainable yield (MSY). We investigated 19 European fish stocks to test the hypothesis that the 95% PGY yield range is inherently precautionary with respect to impairing recruitment. An FMSY range was calculated for each stock as the range of fishing mortalities (F) that lead to an average catch of at least 95% of MSY in long-term simulations. Further, a precautionary reference point for each stock (FP.05) was defined as the F resulting in a 5% probability of the spawning-stock biomass falling below an agreed biomass limit below which recruitment is impaired (Blim) in long-term simulations. For the majority of the stocks analysed, the upper bound of the FMSY range exceeded the estimated FP.05. However, larger fish species had higher precautionary limits to fishing mortality, and species with larger asymptotic length were less likely to have FMSY ranges impairing recruitment. Our study shows that fishing at FMSY generally is precautionary with respect to impairing recruitment for highly exploited teleost species in northern European waters, whereas the upper part of the range providing 95% of MSY is not necessarily precautionary for small- and medium-sized teleosts.</jats:p

Consistent effects of independent domestication events on the plant microbiota
The effect of plant domestication on plant-microbe interactions remains difficult to prove. In this study, we provide evidence of a domestication effect on the composition and abundance of the plant microbiota. We focused on the genus Phaseolus, which underwent four independent domestication events within two species (P. vulgaris and P. lunatus), providing multiple replicates of a process spanning thousands of years. We targeted Phaseolus seeds to identify a link between domesticated traits and bacterial community composition as Phaseolus seeds have been subject to large and consistent phenotypic changes during these independent domestication events. The seed bacterial communities of representative plant accessions from subpopulations descended from each domestication event were analyzed under controlled and field conditions. The results showed that independent domestication events led to similar seed bacterial community signatures in independently domesticated plant populations, which could be partially explained by selection for common domesticated plant phenotypes. Our results therefore provide evidence of a consistent effect of plant domestication on seed microbial community composition and abundance and offer avenues for applying knowledge of the impact of plant domestication on the plant microbiota to improve microbial applications in agriculture
âDrivin' with your eyes closedâ: Results from an international, blinded simulation experiment to evaluate spatial stock assessments
Spatial models enable understanding potential redistribution of marine resources associated with ecosystem drivers and climate change. Stock assessment platforms can incorporate spatial processes, but have not been widely implemented or simulation tested. To address this research gap, an international simulation experiment was organized. The study design was blinded to replicate uncertainty similar to a real-world stock assessment process, and a data-conditioned, high-resolution operating model (OM) was used to emulate the spatial dynamics and data for Indian Ocean yellowfin tuna (Thunnus albacares). Six analyst groups developed both single-region and spatial stock assessment models using an assessment platform of their choice, and then applied each model to the simulated data. Results indicated that across all spatial structures and platforms, assessments were able to adequately recreate the population trends from the OM. Additionally, spatial models were able to estimate regional population trends that generally reflected the true dynamics from the OM, particularly for the regions with higher biomass and fishing pressure. However, a consistent population biomass scaling pattern emerged, where spatial models estimated higher population scale than single-region models within a given assessment platform. Balancing parsimony and complexity trade-offs were difficult, but adequate complexity in spatial parametrizations (e.g., allowing time- and age-variation in movement and appropriate tag mixing periods) was critical to model performance. We recommend expanded use of high-resolution OMs and blinded studies, given their ability to portray realistic performance of assessment models. Moreover, increased support for international simulation experiments is warranted to facilitate dissemination of methodology across organizations.Peer reviewe
Assessing the impact of COVID-19 on liver cancer management (CERO-19).
BACKGROUND & AIMS: The coronavirus disease 2019 (COVID-19) pandemic has posed unprecedented challenges to healthcare systems and it may have heavily impacted patients with liver cancer (LC). Herein, we evaluated whether the schedule of LC screening or procedures has been interrupted or delayed because of the COVID-19 pandemic. METHODS: An international survey evaluated the impact of the COVID-19 pandemic on clinical practice and clinical trials from March 2020 to June 2020, as the first phase of a multicentre, international, and observational project. The focus was on patients with hepatocellular carcinoma or intrahepatic cholangiocarcinoma, cared for around the world during the first COVID-19 pandemic wave. RESULTS: Ninety-one centres expressed interest to participate and 76 were included in the analysis, from Europe, South America, North America, Asia, and Africa (73.7%, 17.1%, 5.3%, 2.6%, and 1.3% per continent, respectively). Eighty-seven percent of the centres modified their clinical practice: 40.8% the diagnostic procedures, 80.9% the screening programme, 50% cancelled curative and/or palliative treatments for LC, and 41.7% modified the liver transplantation programme. Forty-five out of 69 (65.2%) centres in which clinical trials were running modified their treatments in that setting, but 58.1% were able to recruit new patients. The phone call service was modified in 51.4% of centres which had this service before the COVID-19 pandemic (n = 19/37). CONCLUSIONS: The first wave of the COVID-19 pandemic had a tremendous impact on the routine care of patients with liver cancer. Modifications in screening, diagnostic, and treatment algorithms may have significantly impaired the outcome of patients. Ongoing data collection and future analyses will report the benefits and disadvantages of the strategies implemented, aiding future decision-making.
