Anti-CD45RC antibody immunotherapy prevents and treats experimental autoimmune polyendocrinopathy-candidiasis- ectodermal dystrophy syndrome
Targeted monoclonal antibody (mAb) therapies show great promise for the treatment of transplant rejection and autoimmune diseases by inducing more specific immunomodulatory effects than broadly immunosuppressive drugs routinely used. We recently described the therapeutic advantage of targeting CD45RC, expressed at high levels by conventional T (Tconv) cells (CD45RC(hi)), their precursors, and terminally differentiated T (TEMRA) cells, but not by regulatory T cells (Tregs; CD45RC(lo/-)). We demonstrated efficacy of anti-CD45RC mAb treatment in transplantation, but its potential has not been examined in autoimmune diseases. Autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) is a rare genetic syndrome caused by loss-of-function mutations of autoimmune regulator (AIRE), a key central tolerance mediator, leading to abnormal autoreactive T cell responses and autoantibody production. Herein, we show that, in a rat model of APECED syndrome, anti-CD45RC mAb was effective for both prevention and treatment of autoimmune manifestations and inhibited autoantibody development. Anti-CD45RC mAb intervention depleted CD45RC(hi) T cells, inhibited CD45RC(hi) B cells, and restored the Treg/Tconv cell ratio and the altered Treg transcriptomic profile. In APECED patients, CD45RC was significantly increased in peripheral blood T cells, and lesioned organs from APECED patients were infiltrated by CD45RC(hi) cells. Our observations highlight the potential role for CD45RC(hi) cells in the pathogenesis of experimental and human APECED syndrome and the potential of anti-CD45RC antibody treatment.Peer reviewe
Viral to metazoan marine plankton nucleotide sequences from the Tara Oceans expedition
A unique collection of oceanic samples was gathered by the Tara Oceans expeditions (2009-2013), targeting plankton organisms ranging from viruses to metazoans, and providing rich environmental context measurements. Thanks to recent advances in the field of genomics, extensive sequencing has been performed for a deep genomic analysis of this huge collection of samples. A strategy based on different approaches, such as metabarcoding, metagenomics, single-cell genomics and metatranscriptomics, has been chosen for analysis of size-fractionated plankton communities. Here, we provide detailed procedures applied for genomic data generation, from nucleic acids extraction to sequence production, and we describe registries of genomics datasets available at the European Nucleotide Archive (ENA, www.ebi.ac.uk/ena). The association of these metadata to the experimental procedures applied for their generation will help the scientific community to access these data and facilitate their analysis. This paper complements other efforts to provide a full description of experiments and open science resources generated from the Tara Oceans project, further extending their value for the study of the world's planktonic ecosystems
Effet des ultrasons de puissance sur les réactions d'oxydation aérobie d'alcools et d'aldéhydes aliphatiques : application à la synthÚse des phéromones de la Pyrale du buis (Cydalima perspectalis)
In the current context of sustainable development, alternatives to synthetic pesticides must be developed because their uses are hazardous for the environment. An alternative to these synthetic pesticides is chemical ecology, which uses chemical relations between an organism and its environment to control insect-pest. In this context, sex pheromones have already proved their efficiency. However, few syntheses pathways are described in the literature. Indeed, when oxidation reactions occur in their syntheses, oxidants used are generally harmful for people and for the environment.This PhD thesis has aimed at developing eco-compatible synthesis pathway for synthesis of sex pheromones components of the box tree pyralid (Cydalima perspectalis). Based on studies published in the literature, Wittig reaction has been chosen for the formation of (Z)-hexadec-11-en-1-ol leading to (Z)-hexadec-11-enal by oxidation.Aerobic oxidation reactions of aliphatic alcohols and aldehydes have been then investigated and potential impact of power ultrasound as non-conventional technique has been studied. In this context, analysis of photographic negatives of reaction media under ultrasound or silent conditions allowed to obtain original information on the interface modification âgas bubble/reaction mediaâ under ultrasound and their impact on the reaction.Ultrasound does not impact the overall rate of aerobic oxidation reaction of alcohols, but information on the reaction mechanism have been confirmed.Aerobic oxidation of aldehydes, conducted under low frequency ultrasound (20 kHz), allows enhancing results published in literature. Comparatives studies under silent and ultrasonic conditions allow to confirmed information on the reaction mechanism.Dans le contexte actuel de dĂ©veloppement durable, des alternatives aux pesticides de synthĂšse doivent ĂȘtre dĂ©veloppĂ©es puisque leur utilisation est dangereuse pour lâenvironnement. Asymmetric Cu1-Catalyzed Insertion Reaction of 1-Aryl-2,2,2- trifluoro-1-diazoethanes into SiâH Bonds
An asymmetric copper(I)-catalyzed SiâH insertion reaction of 1-aryl-2,2,2-trifluoro-1-diazoethanes in dimethyl carbonate as a green solvent alternative was developed. A C2-symmetric copper(I) diimine complex enabled the asymmetric insertion reaction to give enantioenriched (1-aryl-2,2,2-trifluoroethyl)silanes with excellent stereoselectivities (up to 98:2 er). Successful conversion of the silanes into the corresponding alcohols was performed with retention of stereochemistry. Mechanistic studies and DFT calculations support the proposed model for the observed stereoselectivities
