    Hover performance tests of baseline metal and Advanced Technology Blade (ATB) rotor systems for the XV-15 tilt rotor aircraft

    Rotor hover performance data were obtained for two full-scale rotor systems designed for the XV-15 Tilt Rotor Research Aircraft. One rotor employed the rectangular planform metal blades (rotor solidity = 0.089) which were used on the initial flight configuration of the XV-15. The second rotor configuration examined the nonlinear taper, composite-construction, Advanced Technology Blade (ATB), (rotor solidity = 0.10) designed to replace the metal blades on the XV-15. Variations of the baseline ATB tip and cuff shapes were also tested. A new six-component rotor force and moment balance designed to obtain highly accurate data over a broad range of thrust and torque conditions is described. The test data are presented in nondimensional coefficient form for the performance results, and in dimensional form for the steady and alternating loads. Some wake and acoustic data are also shown

    'Atypical' bacteria are a common cause of community-acquired pneumonia in hospitalised adults

    Objectives. To assess the proportion of cases of community· acquired pneumonia caused by 'atypical' bacteria, inclUding the recently discovered Chlamydia pneumoniae, and to compare the clinical, radiographic and laboratory features of patients with and without 'atypical' bacteria.Methods. A prospective serological study was carried out on consecutive adult pneumonia patients from July 1987 to July 1988. Acute and convalescent sera were tested in batches for antibodies against Legionella pneumophila serogroup 1, C. pneumoniae, Chlamydia psittaci, Coxiella burnetii (phase-2 antigen) and Mycoplasma pneumoniae (lgG and IgM). Records and chest radiographs were examined retrospectively.Results. Acute and convalescent sera were available from 113 patients. The records of 4 patients could not be traced and 17 patients did not fulfil the inclusion criteria. Thirty-two of these 92 patients (35,9%) were found to be infected with 'atypical' bacteria. The two most common organisms were C. pneumoniae (20,7%) and L. pneumophila (8,7%). There. were no differences in the clinical and radiographic features of patients with and without 'atypical' bacteria. Clinicians prescribed erythromycin or tetracyclines with equal frequency in the two groups.Conclusions. 'Atypical' bacteria, especially C. pneumoniae, are a common cause of community-acquired pneumonia in adults in South Africa. This is the first demonstration of an aetiological role of C. pneumoniae in this country. We confirmed the finding of other studies that there are no clinical, radiographic or laboratory features characteristic of 'atypical' bacterial infection in hospitalised patients. This has major implications for therapy, as these organisms respond to erythromycin and tetracyclines, but· not to β-lactam antibiotics

    The promise of whole genome pathogen sequencing for the molecular epidemiology of emerging aquaculture pathogens

    Aquaculture is the fastest growing food-producing sector, and the sustainability of this industry is critical both for global food security and economic welfare. The management of infectious disease represents a key challenge. Here, we discuss the opportunities afforded by whole genome sequencing of bacterial and viral pathogens of aquaculture to mitigate disease emergence and spread. We outline, by way of comparison, how sequencing technology is transforming the molecular epidemiology of pathogens of public health importance, emphasizing the importance of community-oriented databases and analysis tools

    Improving the Sampling Strategy for Point-to-Point Line-Of-Sight Modelling in Urban Environments

    Visibility modelling calculates what an observer could theoretically see in the surrounding region based on a digital model of the landscape. In some cases it is not necessary, nor desirable, to compute the visibility of an entire region (i.e. a viewshed), but instead it is sufficient and more efficient to calculate the visibility from point-to-point, or from a point to a small set of points, such as computing the intervisibility of predators and prey in an agent based simulation. This paper explores how different line-of-sight (LoS) sample ordering strategies increases the number of early target rejections, where the target is considered to be obscured from view, thereby improving the computational efficiency of the LoS algorithm. This is of particular importance in dynamic environments where the locations of the observers, targets and other surface objects are being frequently updated. Trials were conducted in three UK cities, demonstrating a robust five-fold increase in performance for two strategies (hop, divide and conquer). The paper concludes that sample ordering methods do impact overall efficiency, and that approaches which disperse samples along the LoS perform better in urban regions than incremental scan methods. The divide and conquer method minimises elevation interception queries, making it suitable when elevation models are held on disk rather than in memory, while the hopping strategy was equally fast, algorithmically simpler, with minimal overhead for visible target cases

    Towards a System Level Understanding of Non-Model Organisms Sampled from the Environment: A Network Biology Approach

    The acquisition and analysis of datasets including multi-level omics and physiology from non-model species, sampled from field populations, is a formidable challenge, which so far has prevented the application of systems biology approaches. If successful, these could contribute enormously to improving our understanding of how populations of living organisms adapt to environmental stressors relating to, for example, pollution and climate. Here we describe the first application of a network inference approach integrating transcriptional, metabolic and phenotypic information representative of wild populations of the European flounder fish, sampled at seven estuarine locations in northern Europe with different degrees and profiles of chemical contaminants. We identified network modules, whose activity was predictive of environmental exposure and represented a link between molecular and morphometric indices. These sub-networks represented both known and candidate novel adverse outcome pathways representative of several aspects of human liver pathophysiology such as liver hyperplasia, fibrosis, and hepatocellular carcinoma. At the molecular level these pathways were linked to TNF alpha, TGF beta, PDGF, AGT and VEGF signalling. More generally, this pioneering study has important implications as it can be applied to model molecular mechanisms of compensatory adaptation to a wide range of scenarios in wild populations

    A high quality assembly of the Nile Tilapia (Oreochromis niloticus) genome reveals the structure of two sex determination regions

    Background  Tilapias are the second most farmed fishes in the world and a sustainable source of food. Like many other fish, tilapias are sexually dimorphic and sex is a commercially important trait in these fish. In this study, we developed a significantly improved assembly of the tilapia genome using the latest genome sequencing methods and show how it improves the characterization of two sex determination regions in two tilapia species.  Results  A homozygous clonal XX female Nile tilapia (Oreochromis niloticus) was sequenced to 44X coverage using Pacific Biosciences (PacBio) SMRT sequencing. Dozens of candidate de novo assemblies were generated and an optimal assembly (contig NG50 of 3.3Mbp) was selected using principal component analysis of likelihood scores calculated from several paired-end sequencing libraries. Comparison of the new assembly to the previous O. niloticus genome assembly reveals that recently duplicated portions of the genome are now well represented. The overall number of genes in the new assembly increased by 27.3%, including a 67% increase in pseudogenes. The new tilapia genome assembly correctly represents two recentvasagene duplication events that have been verified with BAC sequencing. At total of 146Mbp of additional transposable element sequence are now assembled, a large proportion of which are recent insertions. Large centromeric satellite repeats are assembled and annotated in cichlid fish for the first time. Finally, the new assembly identifies the long-range structure of both a ~9Mbp XY sex determination region on LG1 in O. niloticus, and a ~50Mbp WZ sex determination region on LG3 in the related species O. aureus.  Conclusions  This study highlights the use of long read sequencing to correctly assemble recent duplications and to characterize repeat-filled regions of the genome. The study serves as an example of the need for high quality genome assemblies and provides a framework for identifying sex determining genes in tilapia and related fish species
