Identification of genetic regions associated with Alzheimer ‘s disease blood-based biomarkers in Mexican American families

Background: Dementias are heterogeneous age-associated neurodegenerative disorders generally included in the broad term, Alzheimer’s disease (AD) and related dementias (ADRD). ADRD affects individuals of all ethnicities, but Hispanic individuals show a 1.5-fold higher risk when compared to non-Hispanic whites. Many AD risk biomarkers have been proposed but not much is known about how genetic elements control their expression. We used 2000 genome sequenced Mexican Americans from extended pedigrees to identify genetic regions associated with AD blood-based biomarkers. Method: A total of 70 Mexican American subjects were diagnosed as ADRD cases (h2= 0.75, p = 2.6×10-5). We quantified plasma concentrations, using a QuanterixÒ elisa assay, of four AD candidate biomarkers AB40, AB42, TAU and NFL. We identified genomic regions associated with these biomarkers using a linear mixed model as implemented in SOLAR on a set of 28 million SNPs. Our genetic variance decomposition model is tailored for relatively rare diseases where extended pedigrees are available. Candidate SNPs identified were extensively annotated and their individual eQTL contribution to the expression of flanking genes defined. Result: The heritability component of each AD biomarker was defined, and the most significant estimate observed for NFL (h2= 0.33, p=6.96×10-17). We tested the association between circulating tau and identified two genome-wide significant associations for SNPs rs242557 (3.11×10-14) and rs242562 (1.34×10-13)(Figure 1). Both SNPs are common and located in the first and second introns of the gene MAPT, respectively, and are in a conserved region with chromatin accessibility for transcription factors, suggesting regulatory functions (Figure 2). We tested the association between both SNPs and flanking gene expression, identifying strong evidence of cis-regulation with the expression of the antisense-RNA (KANSL1_AS1) of the gene KANSL1. Conclusion: The identification of genetic variants associated with AD biomarkers is key to providing a reliable and cost-effective genetic test for AD risk for the Hispanic population. The gene KANSL1 encodes a subunit of a histone acetylation enzyme and has been associated with Koolen-de Vries Syndrome and cognitive impairment. The genetic associations are promising, but demand an independent validation to confirm their impact for the AD risk estimation

O objetivo do presente artigo é analisar o papel do Ministério Público Brasileiro na defesa dos direitos dos usuários dos serviços públicos. Inicialmente, analisa a conceituação, titularidade, princípios e classificação dos serviços públicos. Após, apresenta os direitos básicos dos usuários dos serviços públicos no Brasil, conforme previsto na legislação consumerista e administrativista. Por fim, analisa os instrumentos jurídicos de atuação do Ministério Público Brasileiro na defesa desses direitos. A metodologia utilizada foi a pesquisa bibliográfica. Conclui que a atuação ministerial é fundamental na efetivação dos direitos dos usuários desses serviços, bem como na melhoria dos mesmos

INDISPONIBILIDADE CAUTELAR DE BENS EM AÇÕES CIVIS PÚBLICAS POR ATOS DE IMPROBIDADE ADMINISTRATIVA

O objetivo do presente artigo é fazer uma análise crítica acerca do instituto jurídico da indisponibilidade de bens prevista no art. 7º da Lei 8.429/92. A pesquisa é de natureza qualitativa, o método utilizado é o exploratório e a técnica de pesquisa é o levantamento bibliográfico de obras doutrinárias e jurisprudência. Conclui que a indisponibilidade de bens nas ações civis públicas de improbidade administrativa é um poderoso instrumento jurídico à disposição do Ministério Público para a defesa do patrimônio público e social, nos termos do art. 129, III, da CF/88

Hydroxyapatite crystallinity does not affect the repair of critical size bone defects

OBJECTIVE: The physicochemical properties of hydroxyapatite (HA) granules were observed to affect the biological behavior of graft materials. The aim of this work was to analyze the tissue response of two HA granules with different crystallinity and Ca/P ratio in vivo. MATERIAL AND METHODS: The HA granules were produced in the Biomaterials Laboratory (COPPE/UFRJ). The testing materials were HA granules presenting a Ca/P molar ratio of 1.60 and 28% crystallinity (HA-1), and a Ca/P molar ratio of 1.67 and 70% crystallinity (HA-2). Both HAs were implanted into a critical-size calvaria rat defects. RESULTS: To note, in the control group, the bone defects were filled with blood clot only. Descriptive and histomorphometric analyses after 1, 3, and 6 months postoperatively showed mild inflammatory infiltrate, mainly comprising macrophage-like and multinucleated giant cells, and an increase in the volume density of the fibrous tissues (

Caracterização físico-química de seis hidroxiapatitas comerciais para aplicações médico-odontológicas como material de enxerto

O objetivo desse trabalho foi caracterizar seis hidroxiapatitas granulares para aplicação em área médico-odontológica como material de enxerto. Estes materiais foram produzidos por quatro diferentes fabricantes e são todos facilmente disponíveis no mercado brasileiro. A caracterização físico-química foi realizada por meio de microscopia eletrônica de varredura, difração de raios-X, espectroscopia de infravermelho e área superficial específica (BET). Os resultados mostraram que apenas um dos produtos teve a caracterização perfeitamente de acordo com a especificação do fabricante. Os parâmetros de cristalinidade, área superficial e composição podem ser usados para estimar a biodegradabilidade da hidroxiapatita e como critério de controle de qualidade desses materiais.The aim of this work was to characterize six hydroxyapatites in granular form for applications in medicine and dentistry as bone graft and to compare with manufacturers' specification. These samples were produced by four different manufacturers, and all of them are easily available in the Brazilian market. Physicochemical characterization was carried out by using electron microscopy (SEM), X-ray diffraction (XRD), infrared spectroscopy (FTIR) and surface area (BET) measurements. The results show that just one hydroxyapatite sample exhibited characterisitics in perfect agreement with the manufacturers' specifications. The studied parameters, namely crystallinity, surface area and composition may be used to estimate hydroxyapatite's biodegradability and as a criterion for quality control
