    Radioimmunoimaging of 131 I Labeled Monoclonal Antibody JHⅢFab′ in Nude Mice Bearing Human Hepatocellular Carcinoma

    本文选用两种细胞株:人肝癌bEl-7402和肺腺癌l-342细胞,利用MTT法检测平阳霉素对两株细胞药物敏感性的探讨。将每毫升含10--6个细胞的培养液按(1/2)--n(n=0~7)梯度稀释,种于96孔板(100μl/孔),5%CO_2,37℃,培养34H,加MTT20μl(母液2Mg/Ml);再37%培养4H,加dMSO100μl溶解,测570nMOd值,结果贴壁生长的bEl-7402细胞的平均Od值为0.31(0.43~0.24),悬浮生长的l-342细胞的平均Od值为0.27(0.29~0.24),两者无显著性差别(P>0.05)。MTT甲月赞产物最大吸收波长为550~570nM,细胞浓度在一定范围内与甲月赞量近似成正比,MTT浓度以每孔4μg,孵育时间4小时为宜。将平阳霉素(母液1Mg/Ml)按(n=0~5)梯度稀释,加到每孔含有10000个细胞的培养板中,同上法测570nMOd值,结果显

    人红细胞凝集型单抗的制备与鉴定

    目的制备人红细胞凝集型单克隆抗体并进行鉴定。方法以3种方式处理人血红细胞,获得红细胞免疫原,分别免疫Balb/c小鼠。应用杂交瘤技术,取免疫小鼠的脾细胞与Sp2/0骨髓瘤细胞融合,以凝集实验筛选分泌凝集型抗体的杂交瘤细胞,继而用有限稀释法克隆3次,并对杂交瘤细胞的稳定性进行测定。通过腹水法制备抗体,亲和层析法纯化抗体,对抗体的特性进行研究。结果 3种方式获得的免疫原均能建立有效的免疫反应,低渗加超声破碎法所获得的小鼠血清抗体效价最高。通过融合获得8株可稳定分泌抗体的杂交瘤细胞,其中,4A1分泌的抗体凝集效价最高。抗体可使4种人血型的红细胞凝集,无种属交叉凝集反应。结论有效制备了针对人红细胞的凝集性单抗,为其之后的应用研究奠定了基础。福建省省属公益类项目(2015R1036-3,2016R1034-2,2017R1036-3);;福建省科技计划(2014Y0082,2014Y0083

    用dEXTrAnT40为中介体的方法交联抗人肺腺癌单抗3d3和平阳霉素(A5),每克分子抗体可携带68克分子的药物,交联物中抗体活性仍能达到10-4水平,能较好地与靶细胞l-342结合。体外细胞毒实验,显示交联物和游离A5对靶细胞50%杀伤浓度分别为0.9μMOl/l和4.6μMOl/l,杀伤效果比游离A5强5倍多。单抗对靶细胞几乎没有杀伤作用;无关抗体交联物的作用也很弱。交联物和游离A5对非靶细胞MgC-803的杀伤作用无明显差别。结果提示MCAb3d3具有良好的导向作用,能携带结合的A5特异地杀伤肺腺癌l-342细胞,可作为临床抗肿瘤药物导向治疗的一种载体。The conjugate of pingyangmycin (A5) and monclonal antibody3D3(MCAb3D3) against human lung cancer was prepared with dextran T40 as the mediator.The molar ratio of A5 McAb3D3 in the conjugate was 68:1 the titer of the conjugate was remained at 10-4.In vitro,the cytotoxicity of the conjugate and Free A5 yieded IC50 of 0.9μmol/L and 4.6μmol/L,respectivel.The conjugate was Fivetimes stronger than Free A5 The cytotoxicity of the conjugate on non-target cells MGc-803 was much weaker and similar to that of.Free A5.There is no doubt that MCAb3D3 had targeting activity and could kill or damage tumor cells speciFically and could be taken as a kind of vector in studies of clinical targeting therapy

    重组人RGD-FasL对胶质瘤细胞U138/U343/U373的体外抗肿瘤的活性分析

    目的研究重组人RGD-FasL对3株胶质瘤细胞U138/U343/U373的杀伤作用及机制。方法采用RT-PCR方法、MTT比色法、DNA倍体分析、流式细胞术检测融合蛋白对3株胶质瘤细胞的杀伤作用;利用Western blot法探讨其作用机制。结果FasmRNA在U138和U343细胞中有表达,在U373细胞中未有可见表达;DcR3mRNA在3株细胞中均有表达,在U373细胞中强表达。采用流式细胞术检测肿瘤细胞表面两种受体的表达情况,结果与RT-PCR相符。U138细胞株对RGD-FasL敏感并呈剂量依赖性;U343细胞株对RGD-FasL相对敏感;U373细胞对RGD-FasL不敏感。用PI检测细胞周期与凋亡表明RGD-FasL能使U138/U343细胞停留在G1期,推迟进入S期,抑制细胞生长并诱导细胞发生凋亡。Western blot实验表明RGD-FasL作用细胞后caspase-8/3/9的表达升高,Bcl-2的表达降低。结论重组人RGD-FasL可以不同程度的诱导胶质瘤细胞凋亡,其机制与其受体的表达和caspase-8/3/9、Bcl-2的表达有关

    姜黄素诱导人胃腺癌MGC80-3细胞凋亡研究

    以 5、10和 2 0 μmol/ L 姜黄素处理 MGC80 - 372 h,细胞存活率随药物浓度及处理时间依赖性下降 ,并呈现典型的凋亡细胞形态 ;以 0、10、2 0及 30 μmol/ L姜黄素处理细胞 2 4 h后 ,2 0 μmol/ L以上的处理组 DNA琼脂糖电泳显示均有典型的凋亡梯形条带 ,同样条件处理后分别对断裂及未断裂 DNA作定量测定 ,计算细胞凋亡率分别为 7.3%、39.2 %、50 .5%和 6 4 .3% ,表明姜黄素具有诱导MGC 80 - 3细胞凋亡的显著作用 .流式细胞术检测显示经 10及 30μmol/ L姜黄素处理的 MGC 80 - 3细胞周期主要阻滞于 G2 / M期 ,提示细胞的 G2 / M期阻滞与姜黄素诱导 MGC80 - 3细胞凋亡有关

    史氏鲟鱼硫酸软骨素的提取及抗肿瘤活性的研究

    以史氏鲟鱼软骨为原料,按碱提酶解醇沉的工艺流程,进行硫酸软骨素的碱提工艺优化研究。分别通过碱浓度、碱液温度、碱液量的单因素试验及该3个因素的正交试验,研究其对产品得率及其氨基己糖含量的影响,综合分析得出最佳的碱提工艺为2倍量6%的NaOH,于40℃的水浴中搅拌提取4h;该工艺条件制备鲟鱼硫酸软骨素的得率可高达40.45%,产品质量符合硫酸软骨素口服片(WS1-C3-0030-2000)标准的要求。鲟鱼硫酸软骨素精品和粗品对3种细胞乳腺癌细胞株(MCF-7细胞)、胃癌细胞株(MGc803细胞)、肝癌细胞株(SMMC7721)作用72h时,其IC50分别为7.28mg/mL、7.68mg/mL、4.25mg/mL和8.86mg/mL、9.27mg/mL、7.37mg/mL,与鲨鱼硫酸软骨素精品和粗品有着相似的抗肿瘤活性
