    Experimental Study on the Basic Effect of Huatan Anshen Decoction and on Orexin Expression in Hypothalamus of Insomnia Rats

    目的:研究化痰安神方治疗失眠症的药效学基础及其对失眠大鼠下丘脑食欲素表达的影响,揭示本方部分药效学机制。方法:通过小鼠自发活动实验、戊巴比妥钠阈下及阈上睡眠协同实验探索化痰安神方治疗失眠症的药效学基础;通过对氯苯丙氨酸(PCPA)制造失眠大鼠模型,分为中药组、西药组、空白组、模型组,每组干预7天,使用免疫组化检测实验大鼠下丘脑食欲素(Orexin)。结果:(1)化痰安神方可抑制小鼠自发活动,与空白组比较,具有显著差异(P〈0.01);(2)对戊巴比妥钠阈下入睡率的影响:中药组、对照组、西药组实验小鼠入睡率分别为:65%、72%和100%,西药组入睡率明显高于中药组、对照组(P〈0.01);睡眠持续时间,中药组长于西药组(P〉0.05),西药组长于对照组(P〈0.05);(3)对戊巴比妥钠阈上睡眠情况的影响:中药组睡眠持续时间明显长于西药组(P〈0.01),西药组长于对照组(P〈0.01);(4)对氯苯丙氨酸(PCPA)致失眠大鼠各组下丘脑食欲素表达水平无差异(P〉0.05)。结论:化痰安神方具有一定的镇静催眠作用,可明显延长失眠患者的持续睡眠时间,其作用机理可能不是通过调节食欲素水平这一途径。Objective: To study the basic effect of Huatan Anshen decoction and the effect on orexin expressions in Hypothalamus of insomnia rats and reveal the partial pharmacodynamics mechanism. Methods: Spontaneous activity in mice and synergistic action test of pentobarbital sodium were observed to study the basic effect of Huatan Anshen decoction. The second part, through para chlorophenylalanine (PCPA) making rat model of insomnia, divided into Chinese medicine group and Western medicine group, blank group, model group, each group of rats were intervened for 7 days, Hypothalamus Orexin was detected by immunohistochemisty. Results: (1) Huatan Anshen Decoction can inhibit the spontaneous activity of mice, compared with the control group, with significant difference (P〈0.01). (2) Effect on the rate of sleep pentobarbital sodium: Chinese medicine group and control group, western medicine group mice sleep rates were: 65%, 72% and 100%. The rate of western medicine group was significantly higher than that of traditional Chinese medicine group and control group (P〈0.01). Sleep duration: Chinese medicine group was longer than the western medicine group (P〈0.05), the western medicine group was longer than the control group (P〈0.05). (3)Sleep duration of Chinese medicine group was significantly longer than the western medicine group (P〈0.01), and the western medicine group was significantly longer than the control group(P〈0.01). (4) There was no difference in the expression level of orexin (P〈0.05) in the hypothalamus of insomnia rats induced by phenylalanine (PCPA). Conclusion: Huatan Anshen decoction has sedative and hypnotic effects, can prolong the duration of sleep in patients withinsomnia.     Rapid determination of Fe,Ni,Mn,Cu,Zn,Pb in seawater by co-precipitation-centrifuge-X-ray fluorescence spectrometer

    基于共沉淀的富集作用,采用自制的离心管离心沉淀,以手持式X-射线荧光仪(Xrf)测定,由此建立了表层海水中fE,nI,Mn,Cu,zn,Pb的共沉淀-离心-Xrf快速分析方法。测定时间为5 MIn/样;线性范围125μg/l~1 000μg/l,可以满足近岸表层海水中铁和锰的分析要求;基底加标200μg/l回收率为94.5%~116%;连续7次测定6种金属浓度均为500μg/l的海水加标样品,相对标准偏差(rSd)为2.86%~5.85%。与ICP-MS法比较,测定结果无显著性差异。本方法具有化学试剂污染小、方便快捷、可现场快速测定方法等优点。该方法已成功应用于厦门西港和福建九龙江河口表层海水中可溶态铁锰的现场测定,并在现场以手持式Xrf对颗粒物中铁锰进行了测定,获得了该海域颗粒物中和海水中铁锰的分布。Based on the enrichment of co-precipitation with a self-made centrifugal tube and detection with a portable X-ray fluorescence spectrometer(XRF),a rapid method for determination of iron(Fe),nickel(Ni),manganese(Mn),copper(Cu),zinc(Zn) and lead(Pb) in surface seawater was established.The analysis time was 5 min per sample.The linear range was 125 μg/L--1 000 μg/L,and it fulfilled the requirements for the analysis of iron and manganese in coastal surface seawater.The recoveries were between 94.5%--116% with a sample spiked at 200 μg/L;and the relative standard deviations(RSD) were 2.86% -- 5.85%(n=7) at 500 μg/L.Compared with the ICP-MS method,there was no significant difference between the determination results.The method has the advantages of being less contaminated from the chemical reagents,convenient and fast,and practicable in the field.The method was successfully applied in Xiamen western harbor and Jiulongjiang estuary in Fujian Province to measure dissolvable iron and manganese in surface seawater on boat.With the portable XRF,particulate iron and manganese were determined in field as well.The distributions of iron,manganese in particles and seawater were obtained.近海海洋环境科学国家重点实验室自主创新研究课题(MELRI1001

    Preparation of Organo-Functionalized Mesoporous Zeolites Supported Phosphine-Rhodium Complex and Their Catalytic Performance for Hexene-1 Hydroformylation

    制备了表面有机官能团化的MCM 4 1和MCM 4 8介孔分子筛以及SiO2 等载体固载铑膦配合物催化剂 ,考察了所制备催化剂对液相己烯 1氢甲酰化的催化性能 ,并用X射线衍射、BET、红外光谱和原子吸收光谱等技术对载体和催化剂的组成和结构进行了表征 .结果表明 ,固载化铑膦配合物对烯烃氢甲酰化的催化性能与有机官能团的种类及载体的结构有关 ;固载于含胺基和腈基有机官能团化介孔分子筛载体的铑膦配合物表现出较高的催化活性和稳定性 .Organo-functionalized mesoporous zeolites MCM-41 and MCM-48, and amorphous SiO 2 were used as supports to prepare heterogenized phosphine-rhodium complex via anchoring for the hydrofomylation of hexene-1. The supports and the catalysts were characterized by means of XRD, BET, FT-IR and AAS. The MCM-41 and MCM-48 were functionalized with silanes containing (mono-, di-, tri-) amino-, mercapto- and nitrile- groups, respectively, without destruction of the mesoporous structure of the zeolites. The catalytic performance of the catalysts was related to the structure of organo-groups and also to the structural features of the supports. Higher catalytic activity and selectivity for n-C 6H 13CHO were obtained in the case of the catalysts using the mesoporous zeolites functionalized with amino- and nitrile-silanes as supports.
