    Zoo-FISH with human whole-chromosome paint probes delineated syntenic association of human homologous chromosome segments 3-21, 14-15, 16-19, 4-8, 7-16 and 12-22 (twice) in the European mole (Talpa europaea, Talpidae, Eulipotyphla, Mammalia). These segment associations represent shared ancestral Boreo-Eutherian traits, half of which were previously not described for Eulipotyphla.     RNA packaging motor: From structure to quantum mechanical modelling and sequential-stochastic mechanism

    The bacteriophages of the Cystoviridae family package their single stranded RNA genomic precursors into empty capsid (procapsids) using a hexameric packaging ATPase motor (P4). This molecular motor shares sequence and structural similarity with RecA-like hexameric helicases. A concerted structural, mutational and kinetic analysis helped to define the mechanical reaction coordinate, i.e. the conformational changes associated with RNA translocation. The results also allowed us to propose a possible scheme of coupling between ATP hydrolysis and translocation which requires the cooperative action of three consecutive subunits. Here, we first test this model by preparing hexamers with defined proportions of wild type and mutant subunits and measuring their activity. Then, we develop a stochastic kinetic model which accounts for the catalytic cooperativity of the P4 hexamer. Finally, we use the available structural information to construct a quantum-chemical model of the chemical reaction coordinate and obtain a detailed description of the electron density changes during ATP hydrolysis. The model explains the results of the mutational analyses and yields new insights into the role of several conserved residues within the ATP binding pocket. These hypotheses will guide future experimental work

    Sensemaking in Meetings - Collaborative Construction of Meaning and Decisions through Epistemic Authority

    Meetings are important in knowledge-intensive organizations where opportunities for sharing knowledge are the essence of daily work. Meetings are also the de facto event required in almost all organizations and businesses to make decisions. This study examines sensemaking in meetings of software engineers in an IT & telecommunications company. It provides an explanation of how sensemaking serves as a driver for decision making. The identities of the participant also come into play as they use their epistemic authority to influence the evolving meanings and decisions. By incorporating epistemic authority this study draws light on the real-time management of power in professional meetings. The study examines how language specifically is used as a resource for the construction of sensemaking. Conversation Analysis (CA) is used as a method to identify those interactions which characterize how sensemaking and epistemics are played out in organizations. The position taken in this study that sensemaking, decision making and epistemic authority are interactionally accomplished social activities, and the analysis demonstrates how they become consequential for the organizational activities. CA also makes it possible to show how these notions relate to decision making. Majority of existing research focuses on managerial practices or the chairperson’s role in meetings. This study adds to the existing literature on sensemaking and decision making by integrating the notion of epistemic authority as a factor in the accomplishment of these activities among professional peers. The data is comprised of video recordings of five authentic meetings among technical professionals in the area of system software development working in a large multinational company. The findings draw light on the collaborativeness of sensemaking. Firstly, the analysis shows that the practices through which the participants pursue their individual agendas tend to constrain the collaborativeness of sensemaking and they lead to long-winded discussion or argumentation, whereas the practices through which the participants pursue mutual sharing of knowledge lead to collaborative acts of sensemaking. Additionally, the closing phase of each topical discussion in the meetings formed a transition phase in which the past discussion is integrated with future actions, and this becomes labelled as decision. Sensemaking precedes as well as follows decision making at the point of discussion in which collaborative acknowledgement is expressed

    Application of Minisatellite DNA Probes to Linkage in MEN-2

    We describe the potential benefits and the limitations of the use of highly polymorphic minisatellite DNA probes for genetic linkage analysis of multiple endocrine neoplasia type 2A (MEN-2 A). The advantage of these probes is that up to 34 loci can be examined in a single experiment, and since the loci are highly polymorphic, almost every individual in every family is informative. The disadvantage is that the DNA fragment lengths of the alleles at any given locus differ from one family to another, so that families cannot be combined, and large single sibships are needed to obtain significant linkage data. A variable DNA fragment which appears to show linkage in an initial screen of a single sibship must therefore be purified and cloned before chromosomal assignment and extension to further families is possible. These features of the probes are illustrated by a tentative linkage obtained in a large sibship with MEN-2 A

    This paper describes an architecture for decoupling the development process from product implementation for a service in cloud domain. By using automation, a high enough level of abstraction can be achieved in order to effectively and easily create and deploy new instances of an application for customers without involving the developers of the underlying software. The goal of this is to free resources for enhancing the underlying code base, and to enable horizontal development for bringing in new customers. Työssä perehdytään esineiden internetiin ja sen rakenteeseen. Erityisesti työssä on tarkoituksena paneutua ohjelmistoympäristöön Node-RED. Työkalujen avulla luodaan sääaseman tiedonsiirtolinkki väliohjelmistolle. Väliohjelmistossa luodaan käyttöliittymä, josta siirrettyjä tietoja tarkastellaan.     Tradisjonelt har gravplassen vært et sted med primærfunksjonen å gravlegge de døde, og samfunnets felles sorg- og minnested. Befolkningsvekst og urbanisering øker presset på arealene, spesielt på grønne lunger som brukes til rekreasjonsformål. Dette har resultert i behov for å tenke sambruk; hvordan kan dette gjøres på en måte som tar hensyn til både funksjonalitet og estetikk? Os gravlund i Halden er i ferd med nå maks kapasitet, og i forbindelse med detaljreguleringen av utvidelsesområdet ønsker kommunen å gjenåpne Osbekken gjennom den utvidede gravplassen. Gravplassen er et av Halden sentrums største grøntområder, og dette danner bakteppe for denne masteroppgavens problemstilling: Hvordan kan en utvidelse av Os gravlund integreres med en gjenåpning av Osbekken for å skape et område som tilfredsstiller både rekreasjons- og gravlundsbehovene? Målet med oppgaven er å komme med et forslag til utforming av bekkeåpningen, og den utvidede gravplassen, og belyse muligheter og utfordringer rundt sambruk av gravplass, som et sted for både rekreasjon og kontemplasjon. Oppgaven presenterer et forslag til utforming av utvidelse av Os gravlund i Halden, som integrerer en gjenåpning av Osbekken gjennom gravlunden med nye gravfeltarealer, og som tilrettelegger for rekreasjon på gravplassen. Utformingsforslaget er basert på et verdi- og kunnskapsgrunnlag, samt registreringer og analyser av stedet. Den foreslåtte utformingen tar sikte på å skape et rom som gir et verdig og fredelig miljø for de sørgende på gravplassen, samtidig som det tilbyr et rom for rekreasjonsaktiviteter. Planforslaget i oppgaven viser en utforming som oppfyller gravplassens primærfunksjon, samtidig som det er planlagt for rekreasjonsbruk. Dette viser et potensial for gravplassen til å bli en plass som tjener flere funksjoner. Den foreslåtte utformingen av Os gravlund demonstrerer hvordan landskapsarkitekter må balansere ulike behov og interesser i planleggingsprosessen Gjennom oppgaven blir fellestrekk og eventuelle motsetninger mellom funksjonene kontemplasjon og rekreasjon diskutert, og avslutningsvis knyttes dette opp med valgene tatt i utformingen av planforslaget. Oppgaven konkluderer med at at kontemplasjon og rekreasjon i stor grad er forenlige, og sambruk av gravplasser til rekreasjon har mange positive sider, så lenge det ikke går på bekostning av gravplassens primærfunkjson.Traditionally, the cemetery has served as a place primarily dedicated to burying the dead and as a shared site for collective mourning and remembrance. Population growth and urbanization are increasing the need to make good use of the land, particularly on green spaces used for recreational purposes. This has resulted in the need to consider co-usage: How can this be done in a way that takes into account both functionality and aesthetics? Os Cemetery in Halden is reaching its maximum capacity, and as part of the detailed regulation of the expansion area, the municipality intends to reopen the Osbekken stream through the extended cemetery. The cemetery is one of the largest green areas in downtown Halden, providing the backdrop for the research question of this master’s thesis: How can an expansion of Os Cemetery be integrated with the reopening of Osbekken to create an area that meets both the needs for recreation and cemetery purposes? The aim of this thesis is to propose a design for the stream opening and the expanded cemetery and highlight opportunities and challenges related to the co-usage of the cemetery as a place for both recreation and contemplation. The thesis presents a design proposal for the expansion of Os Cemetery in Halden, integrating the reopening of Osbekken through the cemetery with new grave areas and facilitating recreation within the cemetery. The design proposal is based on a foundation of values and knowledge, as well as site surveys and analyses. The proposed design aims to create a space that provides a dignified and peaceful environment for mourners at the cemetery while offering space for recreational activities. The proposed plan in the thesis demonstrates a design that fulfills the primary function of the cemetery while also being planned for recreational use. This shows the potential for the cemetery to become a space that serves multiple functions. The proposed design of Os Cemetery illustrates how landscape architects must balance various needs and interests in the planning process. Throughout the thesis, commonalities and potential conflicts between the functions of contemplation and recreation are discussed, and these are connected to the choices made in the design proposal. The thesis concludes that contemplation and recreation are largely compatible, and the co-usage of cemeteries for recreation has many positive aspects as long as it does not compromise the primary function of the cemetery

    I need to be who I am : a qualitative interview study exploring the needs of people with dementia in Norway

    Objective Many people with dementia receive community services. These services are often based on the needs of informal caregivers and professional caregivers’ assessment. User involvement and participation are main objectives in the Norwegian National Dementia Plan 2020. To enhance user involvement and individual tailoring of healthcare services, more information is required about the needs of people with dementia. The aim of this study was to explore the experienced needs of people with dementia in order to facilitate user involvement in provided services. Methods An explorative cross-sectional study design was used. We performed semistructured interviews with people with dementia who were recruited from all regions of Norway. The sample comprised 35 participants diagnosed with dementia. The interviews were audio-recorded and transcribed, and the data material was analysed using a qualitative content analysis. Results Three main categories emerged from the interviews: (1) to stay connected; (2) to be active and participate; and (3) to live for the moment. The overarching theme was: the need to be who I am. Conclusions People with dementia participating in the study were heterogeneous regarding wants and requirements. Most of them expressed the need and wish to hold on to who they are. Close and robust relations with family and friends can give significant support to people with dementia. However, living with dementia might put considerable strain on relations. Services should provide support to enhance relationships, encourage existing networks to remain stable and facilitate participation in meaningful activities for people living with dementia.publishedVersio

    Quality of Life After Bilateral Adrenalectomy in MEN 2

    Pheochromocytoma is a major cause of morbidity and mortality in the multiple endocrine neoplasia type 2 (MEN 2) syndrome. For the physician, surgical treatment seems well justified even though bilateral adrenalectomy will induce iatrogenically complete loss of adrenocortical function. For the patient this treatment may be a cause of medical problems as well as worry. We have evaluated quality of life after bilateral adrenalectomy in 27 MEN 2 patients through a combined oral and written approach. Mortality was low (one of 27), as was serious morbidity. Most patients had adapted well to the postadrenalectomy state. However, fatigue, worry, and noncompliance with daily medication often caused problems
