    The Industrial Impact on Wetlands in Jinja and the Nexus of Industry, Wetlands, and Community (A Rights-Based Sustainable Development Approach)

    Uganda is a beautiful country filled with an extensive amount of natural resources, one of the most profound being wetlands. It is said that these wetlands are a crucial source of food and water for almost 3 million people (NEMA, 2012 and Oguttu et.al, 2008). They also provide many more invaluable socioeconomic benefits for the country. As Uganda experiences increasing development as well as a rapidly increasing population, the wetlands become increasingly necessary, at the same time they become increasingly threatened. It is written in the 1995 Ugandan constitution that Ugandan policy is aimed at ensuring sustainable development for the protection of the environment as well as the prosperity of the people, however this has been debatable. This study looks at the nexus between industry, wetlands, and local villages in Jinja Uganda, analyzing the dynamics and views between various stakeholders. The overall goal was to evaluate how this nexus applies to rights-based sustainable development with a greater hope of gaining insight into how development can be done without compromising the environment as well as the rights of the people. Guided interviews with various stakeholders such as, village leaders(LC), and ecologist, industrial personnel, and environmental officers were conducted to gain a range of point of views on a number of dynamics with in the study. In addition, informal interviews as well as a focus group was done to gain similar results. The other people consulted were a health care worker, development studies student, as well as other community members. Three villages, Masese, Loco, and Buhumbuli were visited for the study, as well as three factories, Steel Rolling Mills, Skyfat, and Nile Agro. The study found that the relationship between the village and neighboring industries is complex. Both parties rely on the wetlands for various activities and everyone recognizes that these activities have detrimental effect on the wetlands as well as the future of Uganda. The respondents realize their county has various policies to prevent this degradation, but feel as though the implementation is faulty due to corruption, lack of communication, and underfunding. As a result the village members feel they are neglected and not considered when it comes to wetland protection and development. The study concluded that rights-based sustainable development is the necessary approach for Ugandan development, but that due to poor implementation of policy, this is not currently occurring. With better communication, lack of political influence, and increased respect for the local villages, Uganda can find the balance between development and environmental protection

    Dorsal hindbrain ablation results in rerouting of neural crest migration and changes in gene expression, but normal hyoid development

    Our previous studies have shown that hindbrain neural tube cells can regulate to form neural crest cells for a limited time after neural fold removal (Scherson, T., Serbedzija, G., Fraser, S. E. and Bronner-Fraser, M. (1993). Development 188, 1049-1061; Sechrist, J., Nieto, M. A., Zamanian, R. T. and Bronner-Fraser, M. (1995). Development 121, 4103-4115). In the present study, we ablated the dorsal hindbrain at later stages to examine possible alterations in migratory behavior and/or gene expression in neural crest populations rostral and caudal to the operated region. The results were compared with those obtained by misdirecting neural crest cells via rhombomere rotation. Following surgical ablation of dorsal r5 and r6 prior to the 10 somite stage, r4 neural crest cells migrate along normal pathways toward the second branchial arch. Similarly, r7 neural crest cells migrate primarily to the fourth branchial arch. When analogous ablations are performed at the 10- 12 somite stage, however, a marked increase in the numbers of DiI/Hoxa-3-positive cells from r7 are observed within the third branchial arch. In addition, some DiI-labeled r4 cells migrate into the depleted hindbrain region and the third branchial arch. During their migration, a subset of these r4 cells up-regulate Hoxa-3, a transcript they do not normally express. Krox20 transcript levels were augmented after ablation in a population of neural crest cells migrating from r4, caudal r3 and rostral r3. Long-term survivors of bilateral ablations possess normal neural crest-derived cartilage of the hyoid complex, suggesting that misrouted r4 and r7 cells contribute to cranial derivatives appropriate for their new location. In contrast, misdirecting of the neural crest by rostrocaudal rotation of r4 through r6 results in a reduction of Hoxa-3 expression in the third branchial arch and corresponding deficits in third arch-derived structures of the hyoid apparatus. These results demonstrate that neural crest/tube progenitors in the hindbrain can compensate by altering migratory trajectories and patterns of gene expression when the adjacent neural crest is removed, but fail to compensate appropriately when the existing neural crest is misrouted by neural tube rotation

    Digital Three-Dimensional Atlas of Quail Development Using High-Resolution MRI

    We present an archetypal set of three-dimensional digital atlases of the quail embryo based on microscopic magnetic resonance imaging (µMRI). The atlases are composed of three modules: (1) images of fixed ex ovo quail, ranging in age from embryonic day 5 to 10 (e05 to e10); (2) a coarsely delineated anatomical atlas of the µMRI data; and (3) an organ system–based hierarchical graph linked to the anatomical delineations. The atlas is designed to be accessed using SHIVA, a free Java application. The atlas is extensible and can contain other types of information including anatomical, physiological, and functional descriptors. It can also be linked to online resources and references. This digital atlas provides a framework to place various data types, such as gene expression and cell migration data, within the normal three-dimensional anatomy of the developing quail embryo. This provides a method for the analysis and examination of the spatial relationships among the different types of information within the context of the entire embryo

    "The Answer Really Lies in The Community": Exploring Inequity in Resilience Planning Through Community Voices - A Study of Post-Florence New Bern, North Carolina

    The most recent National Climate Assessment states that low-income and marginalized groups with “lower capacity to prepare for and cope with extreme weather and climate-related events” will continue to be most affected, and that “adaptation actions for the most vulnerable populations” should be prioritized. However, while equity is receiving more attention in planning discourse, the uneven impacts of hazards on socially vulnerable populations are often ignored by traditional planning efforts. In 2018 Hurricane Florence devastated New Bern, NC, and in its aftermath revealed communities that were disproportionately at risk from the impacts of the hurricane. Through interviews with New Bern residents, this study aims to provide valuable insight regarding challenges and barriers facing equitable resilience planning in New Bern. The participants’ stories, experiences, and insight speak to some of the factors contributing to uneven resilience across the city. This study found that the avoidance and lack of confronting racism in New Bern, coupled with issues of mistrust and poor community engagement practices, are contributing to patterns of inequitable resilience in New Bern. This paper aims to provide a further understanding of these complex challenges and offer insight that can inspire approaches to resilience planning that best serve all of New Bern’s residents.Master of City and Regional Plannin

    Visualizing and analyzing intracellular transport of organelles and other cargos in astrocytes

    Astrocytes are among the most abundant cell types in the adult brain, where they play key roles in a multiplicity of functions. As a central player in brain homeostasis, astrocytes supply neurons with vital metabolites and buffer extracellular water, ions, and glutamate. An integral component of the “tri-partite” synapse, astrocytes are also critical in the formation, pruning, maintenance, and modulation of synapses. To enable these highly interactive functions, astrocytes communicate among themselves and with other glial cells, neurons, the brain vasculature, and the extracellular environment through a multitude of specialized membrane proteins that include cell adhesion molecules, aquaporins, ion channels, neurotransmitter transporters, and gap junction molecules. To support this dynamic flux, astrocytes, like neurons, rely on tightly coordinated and efficient intracellular transport. Unlike neurons, where intracellular trafficking has been extensively delineated, microtubule-based transport in astrocytes has been less studied. Nonetheless, exo-and endocytic trafficking of cell membrane proteins and intracellular organelle transport orchestrates astrocytes’ normal biology, and these processes are often affected in disease or in response to injury. Here we present a straightforward protocol to culture high quality murine astrocytes, to fluorescently label astrocytic proteins and organelles of interest, and to record their intracellular transport dynamics using time-lapse confocal microscopy. We also demonstrate how to extract and quantify relevant transport parameters from the acquired movies using available image analysis software (i.e., ImageJ/FIJI) plugins

    The INCF Digital Atlasing Program: Report on Digital Atlasing Standards in the Rodent Brain

    The goal of the INCF Digital Atlasing Program is to provide the vision and direction necessary to make the rapidly growing collection of multidimensional data of the rodent brain (images, gene expression, etc.) widely accessible and usable to the international research community. This Digital Brain Atlasing Standards Task Force was formed in May 2008 to investigate the state of rodent brain digital atlasing, and formulate standards, guidelines, and policy recommendations.

Our first objective has been the preparation of a detailed document that includes the vision and specific description of an infrastructure, systems and methods capable of serving the scientific goals of the community, as well as practical issues for achieving
the goals. This report builds on the 1st INCF Workshop on Mouse and Rat Brain Digital Atlasing Systems (Boline et al., 2007, _Nature Preceedings_, doi:10.1038/npre.2007.1046.1) and includes a more detailed analysis of both the current state and desired state of digital atlasing along with specific recommendations for achieving these goals

    MBAT: A scalable informatics system for unifying digital atlasing workflows

    Abstract Background Digital atlases provide a common semantic and spatial coordinate system that can be leveraged to compare, contrast, and correlate data from disparate sources. As the quality and amount of biological data continues to advance and grow, searching, referencing, and comparing this data with a researcher's own data is essential. However, the integration process is cumbersome and time-consuming due to misaligned data, implicitly defined associations, and incompatible data sources. This work addressing these challenges by providing a unified and adaptable environment to accelerate the workflow to gather, align, and analyze the data. Results The MouseBIRN Atlasing Toolkit (MBAT) project was developed as a cross-platform, free open-source application that unifies and accelerates the digital atlas workflow. A tiered, plug-in architecture was designed for the neuroinformatics and genomics goals of the project to provide a modular and extensible design. MBAT provides the ability to use a single query to search and retrieve data from multiple data sources, align image data using the user's preferred registration method, composite data from multiple sources in a common space, and link relevant informatics information to the current view of the data or atlas. The workspaces leverage tool plug-ins to extend and allow future extensions of the basic workspace functionality. A wide variety of tool plug-ins were developed that integrate pre-existing as well as newly created technology into each workspace. Novel atlasing features were also developed, such as supporting multiple label sets, dynamic selection and grouping of labels, and synchronized, context-driven display of ontological data. Conclusions MBAT empowers researchers to discover correlations among disparate data by providing a unified environment for bringing together distributed reference resources, a user's image data, and biological atlases into the same spatial or semantic context. Through its extensible tiered plug-in architecture, MBAT allows researchers to customize all platform components to quickly achieve personalized workflows

    A Presumptive Developmental Role for a Sea Urchin Cyclin B Splice Variant

    We show that a splice variant–derived cyclin B is produced in sea urchin oocytes and embryos. This splice variant protein lacks highly conserved sequences in the COOH terminus of the protein. It is found strikingly abundant in growing oocytes and cells committed to differentiation during embryogenesis. Cyclin B splice variant (CBsv) protein associates weakly in the cell with Xenopus cdc2 and with budding yeast CDC28p. In contrast to classical cyclin B, CBsv very poorly complements a triple CLN deletion in budding yeast, and its microinjection prevents an initial step in MPF activation, leading to an important delay in oocyte meiosis reinitiation. CBsv microinjection in fertilized eggs induces cell cycle delay and abnormal development. We assume that CBsv is produced in growing oocytes to keep them in prophase, and during embryogenesis to slow down cell cycle in cells that will be committed to differentiation

    Interlinkages and gaps: a review of the literature on intergovernmental relations for flood management in the face of climate change

    Current approaches to flood management are increasingly insufficient to deal with intensifying flood trends. In this paper, we define and map out the responsibilities and relationships of local, state, and federal governing entities at various levels. We use these relationships to identify gaps in governance needed to address the high financial, human, and infrastructure costs of flooding. This paper offers a description of current flood policies and provides recommendations for innovations in policy solutions to improve governance gaps. We identify three themes from the literature on intergovernmental relations and flood governance: (1) intergovernmental relations (interlinkages and gaps) for flood governance; (2) risks inherent to flood governance (financial, physical, social and individual, and perception of risk); (3) data adequacy and interoperability
