    Is Retention Enough? Learning and Satisfaction of First-Generation College Seniors

    Progress has been made in retaining and graduating traditional-age first-generation college students at four-year institutions. First-generation students, however, often experience college differently because of external factors, which can negatively influence their learning experience and overall satisfaction. This study explored learning and satisfaction measures of seniors at a small private university in the Midwest. Using Astin’s I-E-O model (1970), the following variables were considered: precollege student characteristics (input); academic engagement, co-curricular activities, campus relationships (environment); and satisfaction, learning (outcome). The sample consisted of seniors participating in NSSE and/or ETS MAPP (n=342). Findings confirmed that first-generation students typically worked more for pay, commuted, and had greater financial need. Results of the analysis of learning and satisfaction however, indicated no difference between the two groups (

    Crohn\u27s disease-associated ATG16L1 T300A genotype is associated with improved survival in gastric cancer

    BACKGROUND: A non-synonymous single nucleotide polymorphism of the ATG16L1 gene, T300A, is a major Crohn\u27s disease (CD) susceptibility allele, and is known to be associated with increased apoptosis induction in the small intestinal crypt base in CD subjects and mouse models. We hypothesized that ATG16L1 T300A genotype also correlates with increased tumor apoptosis and therefore could lead to superior clinical outcome in cancer subjects. METHODS: T300A genotyping by Taqman assay was performed for gastric carcinoma subjects who underwent resection from two academic medical centers. Transcriptomic analysis was performed by RNA-seq on formalin-fixed paraffin-embedded cancerous tissue. Tumor apoptosis and autophagy were determined by cleaved caspase-3 and p62 immunohistochemistry, respectively. The subjects\u27 genotypes were correlated with demographics, various histopathologic features, transcriptome, and clinical outcome. FINDINGS: Of the 220 genotyped subjects, 163 (74%) subjects carried the T300A allele(s), including 55 (25%) homozygous and 108 (49%) heterozygous subjects. The T300A/T300A subjects had superior overall survival than the other groups. Their tumors were associated with increased CD-like lymphoid aggregates and increased tumor apoptosis without concurrent increase in tumor mitosis or defective autophagy. Transcriptomic analysis showed upregulation of WNT/β-catenin signaling and downregulation of PPAR, EGFR, and inflammatory chemokine pathways in tumors of T300A/T300A subjects. INTERPRETATION: Gastric carcinoma of subjects with the T300A/T300A genotype is associated with repressed EGFR and PPAR pathways, increased tumor apoptosis, and improved overall survival. Genotyping gastric cancer subjects may provide additional insight for clinical stratification

    Ship in a bottle: confinement-promoted self-assembly

    Understanding self-assembly in confined spaces is essential to fully understand molecular processes in confined cell compartments and will offer clues on the behaviour of simple confined systems, such as protocells and lipid-vesicle based devices. Using a model system composed of lipid vesicles, a membrane impermeable receptor and a membrane-permeable ligand, we have studied in detail how compartmentalization modulates the interaction between the confined receptor and its ligand. We demonstrate that confinement of one of the building blocks stabilizes complex self-assembled structures to the extent that dilution leads, counterintuitively, to the formation of long range assemblies. The behaviour of the system can be explained by considering a confinement factor that is analogous, although not identical, to the effective molarity for intramolecular binding events. The confinement effect renders complex self-assembled species robust and persistent under conditions where they do not form in bulk solution. Moreover, we show that the formation of stable complex assemblies in systems compartmentalized by semi-permeable membranes does not require the prior confinement of all components, but only that of key membrane impermeable building blocks. To use a macroscopic analogy, lipid vesicles are like ship-in-a bottle constructs that are capable of directing the assembly of the confined ship following the confinement of a few key wooden planks. Therefore, we believe that the confinement effect described here would have played an important role in shaping the increase of chemical complexity within protocells during the first stages of abiogenesis. Additionally, we argue that this effect can be exploited to design increasingly efficient functional devices based on comparatively simple vesicles for applications in biosensing, nanoreactors and drug delivery vehicles

    Reconsidering the Alternatives: The Relationship Between Suspension, Disciplinary Alternative School Placement, Subsequent Juvenile Detention, and the Salience of Race

    Background: Alternative school settings for students who are identified as “disruptive or dangerous” are playing an increasingly prominent role in the world of public education, yet significant gaps in our understanding of their efficacy remain. Objective: This longitudinal investigation within a large school district serving 100,000 students examines multiple factors to determine how the risk of placement in the disciplinary alternative schools is systematically related to predictors and the risk of juvenile subsequent juvenile detention between 3rd and 12th grade. Methods: Four groups of discrete-time hazard models were run separately to determine the most significant predictors in each variable category (i.e., student demographics, behavior-related variables, non-cognitive variables, and academic-related variables); the most significant predictor from each of the four groups was used in the final full model. A second data set was constructed for the analysis of subsequent juvenile detention that included only the subset of cohort students who experienced an alternative placement event using logistic regression. Results: Cumulatively, nearly 1 in 10 children entering 3rd grade experienced placement in a disciplinary alternative school by 12th grade. The racial gaps were pronounced as 13% of all African-American students in the cohort experienced placement compared to 4% of the White students. Race, school mobility, grade retention, special education status, attendance, and out of school suspension were systematically related to the risk of placement in alternative school. The risk of placement was greatest in 7th grade. African-American students were disproportionately represented among those suspended, placed in alternative school, and detained as juveniles. Conclusions: There is a strong relationships between out of school suspensions, disciplinary alternative school placement, and subsequent juvenile detention that are most salient for African-American children, particularly those receiving free/reduced lunch
