    Depositional History and Neotectonics in Great Salt Lake, Utah, from High-Resolution Seismic Stratigraphy

    High-resolution seismic-reflection data from Great Salt Lake show that the basinal sediment sequence is cut by numerous faults with N–S and NE–SW orientations. This faulting shows evidence of varied timing and relative offsets, but includes at least three events totaling about 12 m following the Bonneville phase of the lake (since about 13.5 ka). Several faults displace the uppermost sediments and the lake floor. Bioherm structures are present above some faults, which suggests that the faults served as conduits for sublacustrine discharge of fresh water. A shallow, fault-controlled ridge between Carrington Island and Promontory Point, underlain by a well-cemented pavement, separates the main lake into two basins. The pavement appears to be early Holocene in age and younger sediments lap onto it. Onlap–offlap relationships, reflection truncations, and morphology of the lake floor indicate a low lake, well below the present level, during the early Holocene, during which most of the basin was probably a playa. This low stand is represented by irregular reflections in seismic profiles from the deepest part of the basin. Other prominent reflectors in the profiles are correlated with lithologic changes in sediment cores related to the end of the Bonneville stage of the lake, a thick mirabilite layer in the northern basin, and the Mazama tephra. Reflections below those penetrated by sediment cores document earlier lacustrine cycles

    Report and presentation completed by students enrolled in OLPD 5204: Designing the Adult Education Program, taught by Catherine Twohig in spring 2018.This project was completed as part of the 2017-2018 Resilient Communities Project (rcp.umn.edu) partnership with the City of Ramsey. Septic systems are a safe and legal method of treating household water when maintained properly. However, when residents do not properly care for their systems, they are likely to malfunction and risk contaminating local ground water supplies, posing a public health threat to residents. Because more than 4,000 households in Ramsey use septic systems, the City recognized the importance of helping residents understand the need to properly maintain their systems. Students in OLPD 5204: Designing the Adult Education Program, taught by Catherine Twohig, worked with City staff to create an adult education program that enables Ramsey staff to more effectively communicate to residents the importance of proper septic system use and regular septic system maintenance. The students’ final report and presentation are available.This project was supported by the Resilient Communities Project (RCP), a program at the University of Minnesota whose mission is to connect communities in Minnesota with U of MN faculty and students to advance community resilience through collaborative, course-based projects.     [Resumen] El estudio sísmico de la Laguna Chungará (69° 30' O, 18° 15'S, 4520 m. s.n.m, Altiplano chileno septentrional) y el análisis sedimentológico de varios sondeos ha permitido reconstruir la evolución de la sedimentación lacustre en el lago durante el Holoceno Superior. Se han identificado dos asociaciones de facies: i) litoral, más somera, compuesta por facies de turbera y in plataforma lacustre, más profunda, compuesta por facies con abundantes fragmentos de Characeae. Estas facies alternan en tres ciclos sedimentarios provocados por fluctuaciones en el nivel del lago. Estas variaciones hidrológicas en la laguna han sido causadas por cambios en el balance hídrico que a su vez reflejan importantes fluctuaciones climáticas durante el Holoceno Superior.[Abstract] We reconstruct the Late Holocene sedimentary history of Laguna Chungará (69° 30' W, 18° 10'5, 4520 m.a.s.l., northern Chilean Altiplano) based on high resolution seismic profiling and sedimentologic analyses of cores. Two sedimentary facies associations have been defined and interpreted: n macrophytedominant littoral, composed of black muds with macrophyte remains and peaty muds, and in Characeae-dominant lacustrine self, composed of gray muds and sands with abundant Characeae remains. The two facies associations define three cycles caused by oscillations in the lake level from shallower (macrophyte) to deeper (Characeae) conditions. Changes in the hydrology of Laguna Chungará reflect variations in the effective moisture (precipitation - evaporation) in the Altiplano during the Late Holocene

    Precipitation of soluble uric acid is necessary for in vitro activation of the NLRP3 inflammasome in primary human monocytes

    Objective. To investigate the effects of soluble uric acid (UA) on expression and activation of the NOD-like receptor (NLR) pyrin domain containing protein 3 (NLRP3) inflammasome in human monocytes to elucidate the role of hyperuricemia in the pathogenesis of gout. Methods. Primary human monocytes and the THP-1 human monocyte cell line were used to determine the effects of short- and long-term exposure to UA on activation of the NLRP3 inflammasome and subsequent interleukin-1β (IL-1β) secretion by enzyme linked immunosorbent assay (ELISA) and cell-based assays. Expression of key NLRP3 components in monocytes from patients with a history of gout were analysed by quantitative PCR. Results. Precipitation of UA was required for the activation of the NLRP3 inflammasome and subsequent release of IL-1β in human monocytes. Neither monosodium urate (MSU) crystals nor soluble UA had any effect on activation of the transcription factor, NF-κB. Prolonged exposure of monocytes to soluble UA did not alter these responses. However, both MSU crystals and soluble UA did result in a 2-fold increase in reactive oxygen species (ROS). Gout patients (n=15) had significantly elevated serum UA concentrations compared to healthy individuals (n=16), yet secretion of IL-1β and expression of NLRP3 inflammasome components in monocytes isolated from these patients were not different from healthy controls. Conclusion. Despite recent reports indicating that soluble UA can prime and activate the NLRP3 inflammasome in human peripheral blood mononuclear cells (PBMCs), precipitation of soluble UA into MSU crystals is essential for in vitro NLRP3 signalling in primary human monocytes
