    Secondary metabolites in the interaction of Aspergillus fumigatus and Pseudomonas aeruginosa in cystic fibrosis sputum

    Aspergillus fumigatus and Pseudomonas aeruginosa are the most persistent fungal and bacterial pathogens, respectively, commonly co-isolated from the expectorated sputum of cystic fibrosis (CF) patients. Co-colonisation by both microbes has been associated with a worse clinical outcome and a poorer prognosis for CF patients. The recent years have seen multiple attempts at characterising the interactions between the bacterial species and the fungus. While several mechanisms of mutual inhibition have been proposed in published literature, the interaction models have been based on conventional cultivation strategies, and have thus fallen short of appropriately acknowledging the particular nutritional environment, in which the interaction takes place. In the present study, I have developed a model of the interaction that imitates the native conditions of a cystic fibrosis lung more closely. Using the model, which mimics to a certain extent the growth conditions inside the lung, and, at the same time, do not provide a selective advantage to one of the microorganisms over the other, I investigated the behaviour and the secondary metabolism of the microbes. The study led, among others, to the identification of a novel isonitrilated fungal dipeptide alkaloid, as well as a cyanide detoxication mechanism deployed by the fungus in response to bacterial cyanogenesis. Summarily, the results presented here indicate that rather than engaging in extreme antagonistic behaviour, Aspergillus fumigatus and Pseudomonas aeruginosa co-occurring in the human CF lung achieve a delicate balance, detoxifying each other’s secreted secondary metabolic potential

    Technologies for Sustainable Biomass Supply - Overview of Market Offering

    This article introduces a collection of Information and Communications Technologies (ICT), Internet of Things (IoT), and Industry 4.0 technologies utilized in (or applicable to) biomass supply chains that constitute the current state-of-the-art along with their brief descriptions. The scoping of technologies has been conducted by means of direct interactions with bioeconomy stakeholders and technology providers, analysis of the reports from bioeconomy-related projects, literature surveys, and internet searches. It is to highlight that technology mapping investigated here is focused on commercially available tools and services, which usually come with support, thus removing the necessity for expert knowledge or unusual technical proficiency. The list with over 100 items represents the current best knowledge of its creators and it is currently available through the ICT-BIOCHAIN project platform, serving as a database with technology descriptions and capability for updating the information. The ultimate objective of the database and the platform is to serve as a common point facilitating the cross-sectorial connection, where biomass stakeholders looking for new ICT, IoT, and Industry 4.0 solutions to make their work more efficient and sustainable can browse and filter out the records of their interest and obtain the contact information of the providers

    Host cell factors in ovarian cancer influencing efficacy of oncolytic aenovirus dl922-947

    PhDAdenoviral gene therapy holds great potential for cancer treatment, but is limited by a lack of efficient vectors. dl922-947, an E1A CR2-deleted adenovirus, replicates selectively within and lyses cancer cells. It is believed that its selectivity depends upon abnormalities in the cell cycle regulatory Rb pathway and subsequent G1/S checkpoint, observed in 90% of human cancers. The cytotoxic efficacy of dl922-947 is greater than that of wild-type adenovirus and dl1520 (Onyx-015). Nevertheless, sensitivity to dl922-947 varies widely among ovarian cancer cell lines, despite similar infectivity. My work aimed to identify host cell factors that influence cytotoxicity and which could be potential biomarkers in clinical trials. Surprisingly, comparison of ovarian cancer lines indicated that cytotoxicity correlated poorly with infectivity, replication and virion production. Immunoblotting suggested correlation between sensitivity to dl922-947 and overexpression of p21, p27, Cyclin D, cdk4 and p16. Subsequent experiments confirmed a role for p21 in dl922-947 cytotoxic activity. In vitro and in vivo, Hct116 p21+/+ cells were significantly more sensitive to dl922-947 than matched p21-/- cells. p21 knock-down by siRNA in TOV21G and IGROV-1 cells reduced dl922-947 cytotoxicity, whilst re-expression in ACP-WAF1 cells increased activity. p21 expression was also greater in sensitive transformed TOSE cells compared to resistant normal IOSE25 cells. Results suggest that p21 promotes dl922-947 activity by stabilising Cyclin D thus promoting cell cycle progression. Comparative microarray analysis in TOSE1, 4 and IOSE25 cells and in MRC5 and MRC5-VA cells suggested determinants of dl922-947 activity beyond the Rb pathway, which may also prove valuable biomarkers. Moreover, pathways and processes emerged, correlating with sensitivity, and meriting future investigation. Together, my results suggest that a cellular environment conducive for dl922-947 function includes mediators of proliferative capacity, amongst which p21 plays a role in enhancing activity of the viru
