    Complete Assembly of the Genome of an Acidovorax citrulli Strain Reveals a Naturally Occurring Plasmid in This Species

    Acidovorax citrulli is the causal agent of bacterial fruit blotch (BFB), a serious threat to cucurbit crop production worldwide. Based on genetic and phenotypic properties, A. citrulli strains are divided into two major groups: group I strains have been generally isolated from melon and other non-watermelon cucurbits, while group II strains are closely associated with watermelon. In a previous study, we reported the genome of the group I model strain, M6. At that time, the M6 genome was sequenced by MiSeq Illumina technology, with reads assembled into 139 contigs. Here, we report the assembly of the M6 genome following sequencing with PacBio technology. This approach not only allowed full assembly of the M6 genome, but it also revealed the occurrence of a ∼53 kb plasmid. The M6 plasmid, named pACM6, was further confirmed by plasmid extraction, Southern-blot analysis of restricted fragments and obtention of M6-derivative cured strains. pACM6 occurs at low copy numbers (average of ∼4.1 ± 1.3 chromosome equivalents) in A. citrulli M6 and contains 63 open reading frames (ORFs), most of which (55.6%) encoding hypothetical proteins. The plasmid contains several genes encoding type IV secretion components, and typical plasmid-borne genes involved in plasmid maintenance, replication and transfer. The plasmid also carries an operon encoding homologs of a Fic-VbhA toxin-antitoxin (TA) module. Transcriptome data from A. citrulli M6 revealed that, under the tested conditions, the genes encoding the components of this TA system are among the highest expressed genes in pACM6. Whether this TA module plays a role in pACM6 maintenance is still to be determined. Leaf infiltration and seed transmission assays revealed that, under tested conditions, the loss of pACM6 did not affect the virulence of A. citrulli M6. We also show that pACM6 or similar plasmids are present in several group I strains, but absent in all tested group II strains of A. citrulli

    Using the bar-velocity to predict the one-repetition maximum in the power clean from the knee

    This study investigated the predictive validity and reliability of the load-velocity relationship with the one-repetition maximum test (1RM) in power clean from the knee exercise (PC). Initially, 12 healthy young males, with no PC experience, underwent eight sessions to learn the PC technique. After the learning period, the participants visited the laboratory on four more occasions with intervals between the visits from 72 to 96 h. The first two sessions were dedicated to the actual 1RM tests, while the last two sessions were performed to measure the barbell’s load-velocity relationship at 30%, 45%, 60%, 75%, and 90% of PC 1RM. The highest peak velocity recorded at each load was used to establish the linear regression equation and, consequently, to predict 1RM values. As a result, a low validity was observed between the highest actual 1RM value and the predicted 1RM in sessions 1 and 2 (typical errors = 3.6 to 5.0 kg, coefficients of variation = 6.03 to 8.21%, effect sizes = -1.23 to -1.00, and Bland-Altman bias = 8.5 to 11.7 kg). For reliability, higher measurement errors (intraclass correlation coefficient, typical error, coefficient of variation, and width of limits of agreement at 95%) were observed for the predicted 1RM compared to the actual 1RM test. In conclusion, the load-velocity relationship was not able to predict the 1RM value with high accuracy in the PC from the knee. Moreover, the predicted 1RM presented inferior reliability than the actual 1RM test

    Comparative analysis of adverse events between infliximab and adalimumab in Crohn's disease management: a Brazilian single-centre experience

    AbstractIntroductionData is scarce regarding adverse events (AE) of biological therapy used in the management of Crohn's Disease (CD) among Brazilian patients.ObjectivesTo analyse AE prevalence and profile in patients with CD treated with Infliximab (IFX) or Adalimumab (ADA) and to verify whether there are differences between the two drugs.MethodRetrospective observational single-centre study of CD patients on biological therapy. Variables analysed: Demographic data, Montreal classification, biological agent adminis- tered, treatment duration, presence and type of AE and the need for treatment interruption.ResultsForty-nine patients were analysed, 25 treated with ADA and 24 with IFX. The groups were homogeneous in relation to the variables studied. The average follow-up period for the group treated with ADA was 19.3 months and 21.8 months for the IFX group (p = 0.585). Overall, 40% (n = 10) of patients taking ADA had AE compared with 50% (n = 12) of IFX users (p = 0.571). There was a tendency towards higher incidence of cutaneous and infusion reac- tions in the IFX group and higher incidence of infections in the ADA treated group, although without significant difference.ConclusionsNo difference was found in the AE prevalence and profile between ADA and IFX CD patients in the population studied

    O presente artigo tem como objetivo apresentar detalhadamente as etapas do projeto e o desenvolvimento do protótipo de um robô móvel seguidor de linha de baixo custo construído com alumínio reciclado. A propósito, foi desenvolvido um veículo automatizado para percorrer trajetos e realizar tarefas a partir da identificação de padrões em seu caminho. O robô possui um sistema de direção e tomada de decisões operado por microcontrolador Arduino (programação em C++), a partir de informações coletadas por sensores periféricos. Seu sistema de acionamento de motores poderá acionar as rodas de tração independentemente, possibilitando a realização de curvas sem a necessidade de um eixo móvel. Ele é capaz de se deslocar em terrenos redutores de velocidade e em regiões de subida, atravessar regiões desconhecidas (em que a linha guia não  possa ser detectada) e de identificar, agarrar e transportar objetos até uma área específica, apresentando resultados  confiáveis, com qualidade e baixo custo

    Functional assessment of the pelvic floor muscles by electromyography: is there a normalization in data analysis? A systematic review

    This study aims to evaluate the method of analysis of electromyographic data considering the functional assessment of pelvic floor muscles (PFM). We have included in our search strategy the following databases: Medline, PubMed, Cochrane Central Register of Controlled Trials and Cochrane Database of Systematic Reviews, PEDro, and IBECS, considering articles published in the last ten years (2004-2014). The identified articles were independently examined by two evaluators, according to these inclusion criteria: (1) population: female adults; (2) PFM assessment by electromyography (EMG) with vaginal/anal probe; and (3) description of how electromyographic data analysis is performed. The Newcastle-Ottawa Scale (NOS) was used to assess the risk of bias. We identified 508 articles, of which 23 were included in the review. The data showed differences between the collection protocols, and a significant number of studies did not normalize the electromyographic data. Physiotherapists are among the clinicians who most frequently use EMG to evaluate the function and dysfunction of the neuromuscular system. Although some previous studies have provided an overview to guide the evaluator in the assessment, few succeeding studies followed their recommendations.Este estudo objetiva analisar o método de análise dos dados eletromiográficos na avaliação funcional dos músculos do assoalho pélvico (MAP). As bases de dados consultadas foram: Medline, PubMed, Cochrane Central Register of Controlled Trials e Cochrane Database of Systematic Reviews, PEDro e IBECS, considerando-se artigos publicados nos últimos dez anos (2004-2014). Os artigos identificados foram avaliados independentemente por dois avaliadores, de acordo com os critérios de inclusão: (1) população: mulheres adultas; (2) avaliação dos MAP utilizando a eletromiografia (EMG) com sonda vaginal ou anal; (3) descrição de como é feita a análise dos dados eletromiográficos. O instrumento Newcastle-Ottawa Scale (NOS) foi usado para avaliação da qualidade metodológica dos estudos. Foram identificados 508 artigos, dos quais 23 foram incluídos na revisão. Os dados mostraram diferenças entre os protocolos de coleta, e um número significativo de estudos não utilizaram a normalização na análise dos dados eletromiográficos.     Relação entre velocidade crítica, limiar anaeróbio, parâmetros associados ao VO2max, capacidade anaeróbia e custo de O2 submáximo

    The purposes of the study were to compare and correlate the running velocities associated with the maximal oxygen consumption (Vmax), anaerobic threshold (LL), ventilatory threshold (LV), critical velocity (CV), anaerobic work capacity (AWC) and oxygen cost during sub-maximal exercise (C). METHODS: Eight male physically active subjects (20.8±1.6 years; 74.3±14.9 kg), performed the following tests: 1) 500m and 3km time trial (Vm500 and Vm3km); 2) Incremental test on treadmill for of LL, LV, VO2max and Vmax identification; 3) sustained running at Vmax (Tmax) and identification of the time to VO2max and time at VO2max. The CV and AWC were obtained from linear regression (distance x time on 500m and 3km test). The C was determined as a ratio between sub-maximal running velocity and its VO2. RESULTS: A high correlation was verified for Vm3km and Vmax (r=0.83), Vm3km and VC (r=0.98), Vm500 and VC (r=0.90), Vm500 and Vm3km (r=0.92), and between Vm500 and Time to VO2max (r=0.69). No differences were observed between LL and LV (178.7±20.0 e 180.7±21.8m.min –1) (p>0.05). Also a high correlation between LL and Vmax¬ (r=0.91), CV (r=0.96), and Vm3km (r=0.96) were verified. The AWC was negatively correlated to running economy for those with higher aerobic capacity. CONCLUSION: The CV, AWC, Vm500 an Vm3km can be used for exercise evaluation and training prescription. Due the correlation with the C, the AWC may be an indicative of mechanical inefficiency.Os objetivos do estudo foram analisar as relações existentes entre a velocidade de corrida associada ao VO2max (Vmax), tempo para obtenção do VO2max na Vmax, tempo no VO2max, limiar de lactato (LL), limiar ventilatório (LV), velocidade crítica (VC), capacidade de trabalho anaeróbio (CTAn), desempenho em corrida de 500m e 3km e custo de O2 durante exercício submáximo (C). MÉTODOS: 8 homens fisicamente ativos (20,8±1,6 anos; 74,3±14,9kg) realizaram: 1) Corrida 3km e 500m máximos (Vm3km e Vm500). 2) Teste incremental em esteira para determinação do LL, LV, VO2max e Vmax. 3) Tempo de permanência na Vmax (Tmax), Tempo para atingir o VO2max e Tempo no VO2max. A VC e a CTAn foram obtidas (método linear) a partir dos testes de 3km e 500m. O C foi determinado dividindo-se a velocidade de corrida submáxima pelo respectivo VO2. RESULTADOS: Observou-se alta correlação entre Vm3km e Vmax (r=0,83), Vm3km e VC (r=0,98), Vm500 e VC (r=0,90) e entre Vm500 e Tempo no VO2max (r=0,69). Não foram verificadas diferenças entre LL e LV (178,7±20,0 e 180,7±21,8 m.min-1), sendo o LL altamente correlacionado com Vmax (r=0,91), VC (r=0,96) e Vm3km (r=0,96). Correlação positiva foi observada entre CTAn e C para indivíduos com maior aptidão aeróbia (r=0,79). CONCLUSÕES: As correlações apresentadas entre Vm500 e tempo no VO2max e entre Vm500 e VC indicam que exercícios de curta duração e alta intensidade são positivamente influenciados pela aptidão aeróbia. Devido a correlação significativa com o C, a CTAn se mostra como um possível indicador de ineficiência mecânica no exercício aeróbio

    Lipid profile in schoolchildren in Vitória – Brazil

    Introduction: The growing prevalence of obesity is currently considered the most important nutritional disorder. It is characterized, according to the World Health Organization (WHO) as a worldwide epidemic in developing and developed countries. In an associated form, there is an increasing prevalence of dyslipidaemia. Aiming to improve the current situation and prevent the progression of the epidemic, the American Academy of Pediatrics recently reinforced the need for cholesterol screening in overweight children older than two years. Objective: To determine overweight and lipid profi le in children aged between six and nine years. Methods: A descriptive, cross-sectional study with children from Vitoria, ES. For nutritional classifi cation the Z-score > + 1 SD body mass index-for-age (according to WHO / 2007) was used; for the lipid profi le the Atherosclerosis Prevention Guidelines in Childhood were used. Anthropometric measurements (weight, height, waist circumference and triceps skinfold thickness) followed the standard techniques described by WHO. Data were organized and analysed using SPSS, version 8.5 and calculated the absolute, relative and mean (SD) frequencies and the association between overweight, lipid profile and other variables is adopted as signifi cant when p < 0.05. Results: The sample comprised a total of 511 children of both sexes (46.7% male), with a mean age of 101.6 ± 11.1 months. Overweight was found in 197 (38.5%) children: overweight in 71 (13.9%) and obesity in 126 (24.6%). Total cholesterol was elevated in 167 (32.7%) as were high LDL-C (136–27%). High triglycerides were found in 21 participants (4.1%). Signifi cant association was found between waist circumference and high levels of triglycerides (p = 0.019) and HDL-C (p = 0.033). Conclusion: Excess weight of the sample investigated is considered high and its health effects are important, with an increased total cholesterol greater than 32%. The high levels of HDL-C are protectivefactors for coronary heart disease, although the lipid profi le had been changed
