    Tato bakalářská práce se zabývá modelováním systémů pro řízení vozidla s využitím prostředí Mathworks Matlab/Simulink. První kapitoly jsou věnovány teoretické rešerši z oblasti řídicích prvků automobilu. Jsou zde popsány systémy, kterými řidič ovlivňuje chod vozidla. Další část práce obsahuje modelování a simulace těchto systémů. Cílem této bakalářské práce je seznámení s jednotlivými řídicími systémy, jejich modelováním v softwarovém programu a porovnání výsledků získaných na modelu, s výsledky z reálných měření na vozidle.This bachelor thesis deals with modelling of vehicle’s control systems in software program Matlab-Simulink. First parts of the work are dedicated to a theoretical research of vehicle control systems. There are descripted systems that can be affected by a driver. Second part is aimed to modelling and simulating these systems.     Different view on PQ theory used in the control algorithm of active power filters

    The improvement of power quality is a frequently discussed issue, which still requires a considerable research effort to be devoted to the study of the problem. The aim of this paper is to describe some problems related to the control of switching compensators, commonly known as active power filters. It also includes some shortcomings of pq theory regarded as three phase instantaneous power theory. The term “shortcomings” means that the pq theory does not provide a proper description of power properties. Moreover the control algorithm based on this theory only achieves satisfactory results for sinusoidal balanced voltage system. Nevertheless it can still be considered a helpful approach to the problem under study. The simulation results presented in this paper illustrate the weaknesses of the pq theory

    Glycine N-methyltransferases: A comparison of the crystal structures and kinetic properties of recombinant human, mouse and rat enzymes

    Glycine N-methyltransferases (GNMTs) from three mammalian sources were compared with respect to their crystal structures and kinetic parameters. The crystal structure for the rat enzyme was published previously. Human and mouse GNMT were expressed in Escherichia coli in order to determine their crystal structures. Mouse GNMT was crystallized in two crystal forms, a monoclinic form and a tetragonal form. Comparison of the three structures reveals subtle differences, which may relate to the different kinetic properties of the enzymes. The flexible character of several loops surrounding the active site, along with an analysis of the active site boundaries, indicates that the observed conformations of human and mouse GNMTs are more open than that of the rat enzyme. There is an increase in kcat when going from rat to mouse to human, suggesting a correlation with the increased flexibility of some structural elements of the respective enzymes. © 2004 Wiley-Liss, Inc

    5-Methyltetrahydrofolate is bound in intersubunit areas of rat liver folate-binding protein glycine N-methyltransferase

    Glycine N-methyltransferase (GNMT) is a key regulatory enzyme in methyl group metabolism. It is abundant in the liver, where it uses excess S-adenosylmethionine (AdoMet) to methylate glycine to N-methylglycine (sarcosine) and produces S-adenosylhomocysteine (AdoHcy), thereby controlling the methylating potential of the cell. GNMT also links utilization of preformed methyl groups, in the form of methionine, to their de novo synthesis, because it is inhibited by a specific form of folate, 5-methyltetrahydrofolate. Although the structure of the enzyme has been elucidated by x-ray crystallography of the apoenzyme and in the presence of the substrate, the location of the folate inhibitor in the tetrameric structure has not been identified. We report here for the first time the crystal structure of rat GNMT complexed with 5-methyltetrahydrofolate. In the GNMT-folate complex, two folate binding sites were located in the intersubunit areas of the tetramer. Each folate binding site is formed primarily by two 1-7 N-terminal regions of one pair of subunits and two 205-218 regions of the other pair of subunits. Both the pteridine and p-aminobenzoyl rings are located in the hydrophobic cavities formed by Tyr 5, Leu207, and Met215 residues of all subunits. Binding experiments in solution also confirm that one GNMT tetramer binds two folate molecules. For the enzymatic reaction to take place, the N-terminal fragments of GNMTmust have a significant degree of conformational freedom to provide access to the active sites. The presence of the folate in this position provides a mechanism for its inhibition

    Analysis of problems, consequences and solutions for BIM application in reconstruction projects

    The main aim of analysis presented in this paper is based on the complex evaluation of Building Information Modelling (BIM) application and decision support in reconstruction projects. The reconstruction projects face the high level uncertainty during implementation mostly due inappropriate information management, insufficient quality control and low competence of participants. The application of advanced Computer-Aided tools such as BIM uses both with advanced Information and Communication Technologies (ICT) can be the way to solve the problems of reconstruction projects. The fourteen reconstruction projects have been analysed and the list of thirteen main problems has been identi­fied. The pair wise judgment of problems has been made based on in the expert survey using the Analytic Hierarchy Pro­cess (AHP) method. The occurrence, influence to project and significance of the problems have been calculated and were prioritised according to the importance and consequences. After the complex analysis of problems, it is suggested to use combined list of solutions of the BIM tool application in reconstruction projects to support decision making process

    Designing surveys for radiography and radiation therapy.

    Surveys using standardised questionnaires are frequently used in the health area for research, to gather information to inform or improve clinical practice and to gage perceptions or attitudes towards particular issues. As we begin to conduct more studies in the areas of radiography and radiation therapy we need to ensure that it is essential that we use reliable and valid research methods so that findings are robust and, importantly, useable in clinical practice. This review focuses on how to design the questionnaire and highlights some key rules to help avoid the many pitfalls in survey design. The importance of the planning stage and time spent reviewing and revising questions is emphasised and the benefits and challenges of using existing instruments are discussed. Questionnaire layout is addressed and also the wording of individual questions as both of these will affect response rates and how people interpret questions. Wording can be difficult and may take a few drafts to get right. Several examples are given to demonstrate the concepts. The key messages in this review are: spend time planning and make the questionnaire brief, easy and safe. Issues such as response rates, types of questions, how to test for reliability and validity; and how to analyse the data will be covered in a further review for The Radiographer
