    Analysis of the productive and immune response of lambs infected with gastrointestinal nematodes and fed with saccharin

    Objective: To determine the productive traits and immune response in Blackbelly lambs infected with gastrointestinal nematodes (GIN) fed with saccharin. Design / methodology / approach: Three treatments (T) were randomly assigned to 18 Blackbelly lambs, with an initial live weight (LW) of 13.9 + 3.2 kg. In T1 anthelmintic + base diet (CTah) was used; T2 without anthelmintic + base diet (STah) and T3 were grazing lambs without anthelmintic (STPS). The experimental design was completely randomized with measures repeated over time and the averages of the treatments were compared using Lsmeans. Evaluations included live weight (LW), egg per gram of feces (EPG), packed cell volume (PCV), total plasma protein (TPP), differential leukocyte count (WBC), and IgA level by ELISA with Haemonchus contortus and Trichostrongylus colubriformis antigens Results: The STah and CTah lambs showed a higher number of EPG (885 ± 142) and LW (29.73 ± 5.06 kg). While grazing lambs (STPS) showed the lowest PCV (26.4 ± 0.5%) compared to STah and CTah lambs (27.4 to 28.4%), caused by the high prevalence of H. contortus. The IgA level in grazing lambs was from 20.2 to 24.5% with respect to the positive standard, RPS, while the stabled lambs (STah and CTah) showed values ??close to 5% RPS. Study limitations / implications: Due to anthelmintic resistance problems, it was not possible to keep lambs free of infection in grazing, so this group was not included.     Antibodies of Trypanosoma cruzi, Leishmania mexicana and Leishmania braziliensis in domiciled dogs in Tabasco, Mexico

    ABSTRACT Objective. To determine Trypanosoma cruzi (T. cruzi), Leishmania mexicana (L.mexicana) and Leishmania braziliensis (L.braziliensis) circulating antibodies in dogs from Chontalpa region in Tabasco, Mexico using ELISA diagnostic techniques Fe-SOD and Western blot. Materials and methods. For this study, 119 serums were obtained from domiciled dogs. Serums were tested for antibodies against T. cruzi, L. mexicana and L. braziliensis, using ELISA and Western Blot sod as diagnostic test. The antigenic fraction used in both tests was the Fe-SOD excreted by the species of Leishmania and Trypanosoma. Results. The obtained frequency in this study was 3.36% for T. cruzi, 9.24% for L. mexicana and 10.08% for L. braziliensis. Conclusions. The present study has demonstrated the presence of antibodies to these parasites in Chontalpa region from Tabasco, Mexico.RESUMEN Objetivo. Determinar la frecuencia de anticuerpos circulantes de Trypanosoma cruzi (T. cruzi), Leishmania mexicana (L. mexicana) y Leishmania braziliensis (L. braziliensis) en una población de perros usando ELISA Fe-SOD y Western blot en la región Chontalpa del estado de Tabasco, México. Materiales y métodos. Para este estudio se obtuvieron 119 sueros de perros domiciliados, con el consentimiento previo de los propietarios. Los sueros fueron analizados para detectar anticuerpos contra T. cruzi, L. mexicana, y L. braziliensis, usando como prueba diagnóstica ELISA-sod y Western Blot. La fracción antigénica utilizada en las dos pruebas fue la Fe-SOD excretada por las especies de Trypanosoma y Leishmania. Resultados. La frecuencia obtenida en este estudio fue de 3.36% para T. cruzi, 9.24% para L. mexicana y 10.08% L. braziliensis. Conclusiones.     Resumen: En la búsqueda de nuevas alternativas para el mejoramiento de la salud animal se evaluó la actividad antihelmíntica de tres plantas de uso tradicional (Cydista aequinoctialis L., Heliotropium indicum L. y Momordica charantia L.) en el área rural del estado de Tabasco contra Haemonchus contortus. Los extractos acuosos crudos se obtuvieron del material deshidratado. Cada uno de ellos fue disuelto en agua destilada a una concentración de 20 mg ml-1 y la actividad antihelmíntica se evaluó en placas de ELISA de 96 pozos se colocaron alícuotas de 100 μL de extracto y 50 larvas L3 desvainadas de H. contortus. Como control negativo se utilizó agua destilada y como control positivo Febendazol, evaluando a las 24, 48 y 72 h de exposición. Los extractos de las tres plantas estudiadas presentaron diferente actividad antihelmíntica y la mayor mortalidad se presentó con el fruto de M. charantia con 65.5% a las 72 h.Abstract: Antihelmintic activity against Haemonchus contortus was searched as a new alternative for animal health improvement using three plants (Cydista aequinoctialis L., Heliotropium indicum L. and Momordica charantia L.) of traditional usage in the rural area of Tabasco State, Mexico.     Molecular detection of Dirofilaria immitis in dogs and mosquitoes in Tabasco, Mexico

    Background & objectives: Dirofilaria immitis is a filarial nematode that causes heartworm disease in domestic as well as wild canines and felines; and cutaneous or pulmonary infections in humans. The purpose of the study was to estimate the prevalence of D. immitis in domestic dogs in Tabasco, Mexico and to assay mosquitoes temporally and spatially associated with dogs for evidence of infection. Methods: Blood was collected from 1050 dogs in 1039 houses during a random household survey performed in 2016 and 2017. Genomic DNA was extracted and assayed by polymerase chain reaction (PCR) using pan-filarial primers and various species-specific primers. Dog owners were interviewed using a structured questionnaire designed to collect information on factors that may impact the occurrence of filarial infection. The association between canine dirofilariasis prevalence and factors likely to impact infection was determined by univariate logistic regression analysis, followed by multivariate binomial logistic regression analysis. Indoor and outdoor resting mosquitoes were collected from houses by manual aspiration. Mosquitoes were identified according to species, homogenized and tested by PCR for filarial nematodes. Results: A total of 84 (8%) dogs were positive for D. immitis DNA, while 3 (0.3%) dogs contained Acanthocheilonema reconditum DNA. Several factors were significantly associated with D. immitis infection. For example, dogs that lived <100 m from a large source of open standing water were significantly more likely (p = 0.002) to become infected with D. immitis than other dogs. Additionally, dogs with infrequent or no anthelmintic treatment were significantly more likely (p = 0.0) to become infected than dogs that were regularly treated. The entomologic investigation yielded 2618 female mosquitoes from 14 species. Four pools of Culex quinquefasciatus were positive for D. immitis DNA and the minimum infection rate, calculated as the number of positive pools per 1000 mosquitoes tested, was 2.9. Interpretation & conclusion: The study identified several factors positively associated with an increased risk of D. immitis infection in domestic dogs in Tabasco and provides evidence that Cx. quinquefasciatus is potentially an important vector in this region. This information can be used by local veterinarians and dog owners to reduce the burden of D. immitis on canine health

    Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide.
