    In vitro antiatherogenicity of extracts from Halimeda incrassata seaweed: antioxidant activity and smooth muscle cell migration studies

    Aim: The aim of this work was to evaluate the in vitro atheroprotective potential of the seaweed Halimeda incrassata in smooth muscle cell migration and lipoprotein oxidation in relation to its antioxidant activity. Material and methods: Antioxidant activity was determinate by DPPH• radical scavenging assay and ORAC method. The inhibitory effect of the aqueous extract on LDL oxidation mediated by Cu2+ ions was determinate by TBARS and conjugated diene quantification. The effect of the seaweed aqueous extract on smooth muscle cell migration was evaluated in MOVAS-1 mouse aortic smooth muscle cell. Results: The inhibitory effect of the aqueous extract on lipoprotein oxidation mediated by Cu2+ was demonstrated. Seaweed extract caused dose-dependent inhibition of TBARS (IC50 = 0.8 mg/mL) and conjugated dienes formation. The seaweed had a high antioxidant activity in the assays performed. The activity could be related to the phenolic content of Halimeda incrassata. Conclusions: In summary, the results of this study represent a further step in the characterization of the atheroprotective action of Halimeda incrassata and indicate the seaweed could be used for a nutraceutical and/or phytoterapeutic application.Objetivos: El objetivo de este trabajo fue evaluar el potencial ateroprotector in vitro del alga Halimeda incrassata en la migración de células de músculo liso de ratón y la oxidación de lipoproteínas en relación con su actividad antioxidante. Material y métodos: La actividad antioxidante fue determinada mediante los métodos de inhibición de radicales DPPH y la Capacidad antioxidante total (ORAC). La actividad inhibitoria de la oxidación de LDL mediada por iones Cu2+ se determinó por la cuantificación de TBARS y dienos conjugados. El efecto del extracto acuoso sobre la migración de las células de músculo liso se evaluó en la línea de células de músculo liso aórtica de ratón MOVAS-1. Resultados: Se demostró el efecto inhibidor del extracto sobre la oxidación de LDL mediada por Cu2+. El extracto del alga causa inhibición dosis-dependiente de la formación de TBARS (IC50 = 0,8 mg/mL) y dienos conjugados. Las algas tuvieron una alta actividad antioxidante en los ensayos realizados y podría estar relacionada con el contenido de compuestos fenólicos. Conclusiones: Los resultados de este trabajo representan un paso más en la caracterización de la acción ateroprotectora de Halimeda incrassata y evidencian sus posibles aplicaciones como nutracéutico y/o fitofármaco.The research was funded by IFS grant F/4897-1.     Comparation of antioxidants properties and polyphenols content of aqueous extract from seaweeds Bryothamnion triquetrum and Halimeda opuntia

    Objetivos. Evaluar y comparar las propiedades antioxidantes mediante ensayos in vitro de extractos acuosos de las algas roja Bryothamnion triquetrum y verde Halimeda opuntia y su relación con el contenido de polifenoles. Material y Métodos. Se utilizaron las técnicas in vitro: DPPH, Capacidad reductora, Inhibición de la peroxidación lipídica e inhibición de la hemólisis inducida por AAPH. Resultados. B. triquetrum: DPPH; CI50=1,15 ± 0,06, capacidad reductora a concentración; 128 mg/mL, DO=2,798, inhibición de la peroxidación lipídica; CI50=5,09± 0,25 e inhibición de la hemólisis con 12 mg/ mL; 35 %. H. opuntia: DPPH; CI50=12,34 ± 0,30 mg/mL, capacidad reductora; DO=0,800, inhibición de la peroxidación lipídica; CI50=1,25± 0,31 mg/mL e inhibición de la hemólisis; 82%. Conclusiones. Los resultados evidencian que B triquetrum resulto mucho más eficiente en los ensayos de DPPH y Capacidad reductora mientras H opuntia resulta más eficiente en Inhibición de la peroxidación lipídica e inhibición de la hemólisis. Se discuten algunos aspectos acerca de sus posibles mecanismos de acción.Objective. To evaluate and compare the antioxidant activity displayed by seaweed H. opuntia and B. triquetrum using different experimental in vitro antioxidant assessment models. Material and Methods. The following techniques are utilized: DPPH, Reducing power, inhibition of lipid peroxidation and inhibition of haemolysis. Results. B. triquetrum: DPPH; IC50=1.15 ± 0.06, Reducing power (concentration 128 mg/mL), OD=2.798, inhibition of lipid peroxidation; IC50=5.09± 0.25 and inhibition of haemolysis; with 12 mg/mL; 35 %. H. opuntia: DPPH; IC50=12.34 ± 0.30 mg/mL, reducing power; OD=0.800, inhibition of lipid peroxidation; IC50=1.25± 0.31 mg/mL, inhibition of lipid peroxidation; IC50=1.25± 0.31 mg/mL and inhibition of haemolysis; 82%. Conclusion. It was demonstrated that B.triquetrum extract was more effective than H. opuntia in reducing power and DPPH assays while H.opuntia was more effective in inhibition of lipid peroxidation in rat brain homogenates and the inhibition of red blood cell (RBC) haemolysis induced by AAPH.     Desenvolvimento de um roteiro conceitual para a gestão da biodiversidade e dos serviços ecossistêmicos no Caribe mexicano

    Coral reefs and mangroves support rich biodiversity and provide ecosystem services that range from food, recreational benefits and coastal protection services, among others. They are one of the most threatened ecosystems by urbanization processes. In this context, we developed a conceptual framework for the management of biodiversity and ecosystem services for these coastal environments. We based our workflow on two sections: “Information base” and “Governance” and use the Puerto Morelos Coastal region as a case study for coastal protection. Puerto Morelos is between two of the most touristic destinations of Mexico (Playa del Carmen and Cancun) that has experienced an increase of population in the past four decades resulting in an intensification of multiple threats to its ecosystems. We characterized the two ecosystems with a “Management Units” strategy. An expert-based ecosystem services matrix was also described in order to connect mangroves and coral reef ecosystems with the multiple beneficiaries. Then an ecosystem model (conceptual model and Global Biodiversity model) was developed. The conceptual model was useful in understanding the interplay processes between systems regarding the ecosystem service of “Coastal Protection”. The Global Biodiversity model evidenced the human-induced shifts in the biodiversity for mangrove and coral reefs ecosystems. Also, a projection for 2035 of “best” and “worst” scenarios was applied using GLOBIO3. A DPSIR conceptual framework was used to analyze environmental problems regarding ecosystem services maintenance. Finally, we evaluated a set of policies associated with these ecosystems that favor coastal protection integrity. This framework facilitates the identification of the most relevant processes and controls about the provision of coastal protection service. It can also be useful to better target management actions and as a tool to identify future management needs to tackle the challenges preventing more effective conservation of coastal environments.Recifes de coral e manguezais possuem rica biodiversidade e fornecem serviços ecossistêmicos, tais como, alimento, recreação, proteção costeira, entre outros. Esses ecossistemas encontram-se entre os mais ameaçados pelos processos de urbanização. Nesse contexto, desenvolvemos um roteiro conceitual para a gestão da biodiversidade e dos serviços ecossistêmicos desses ambientes costeiros. Organizamos nossa sequência de passos de trabalho em duas seções: “Base de informações” e “Governança” e usamos a região costeira da cidade de Puerto Morelos (México) como um estudo de caso para analisar o serviço de proteção de costa. Puerto Morelos encontra-se entre dois dos destinos mais turísticos do México (Playa del Carmen e Cancún), e portanto sua população vem aumentando nas últimas quatro décadas, resultando na intensificação de múltiplas ameaças para os ecossistemas. Primeiramente, caracterizamos os dois ecossistemas identificando-os como “Unidades de Gestão”, detalhando seus principais componentes e processos. Através de uma “Matriz de serviços ecossistêmicos”, construída com base na opinião de especialistas, foram sistematizados os principais serviços ecossistêmicos prestados pelos manguezais e recifes de corais aos múltiplos beneficiários. Em seguida, foi desenvolvida uma modelagem do sistema (e ecossistemas) através de sua representação na forma de um modelo conceitual e um modelo numérico de Biodiversidade Global. O modelo conceitual facilitou a compreensão dos processos de interação entre sistemas em relação ao serviço “Proteção Costeira”. O modelo numérico evidenciou as mudanças induzidas pelo homem na biodiversidade dos ecossistemas de manguezal e recifes de coral. Além disso, uma projeção dos cenários “melhor” e “pior” foi desenvolvida para 2035 usando GLOBIO3. A Estrutura conceitual DPSIR foi aplicada para analisar problemas ambientais relacionados à manutenção dos serviços ecossistêmicos. Finalmente, avaliamos um conjunto de políticas públicas associadas a esses ecossistemas e que favorecem a integridade da proteção costeira. Portanto, o roteiro facilitou a identificação dos principais processos e controles para a provisão de um serviço ecossistêmico. Além disso, pode ser útil para direcionar melhor as ações de gerenciamento, bem como, uma ferramenta para identificar necessidades futuras de planejamento e gestão para enfrentar desafios que permitam uma conservação mais eficaz dos ambientes costeiros.Fil: Sánchez Quinto, Andrés.     Germination of Crotalaria and Lupinus (Fabaceae) seeds submitted to different pre-germination treatments and their effect on enzymatic activity during early germination

    Abstract Most of the wild and native legume seeds has a hard and impermeable testa, which causes physical dormancy and prevents them from germinating even when environmental conditions are favorable. The study evaluated the effect of scarification treatments on germination and enzymatic activity of Crotalaria longirostrata (Cl) and Lupinus exaltatus (Le) seeds. After scarification treatments, germination percentage (GP) and rate (GR) were assessed during 30 days after seeding (DAS); and water absorption (WA) and specific enzymatic activity (SEA) during early germination (0, 6, 18, 36, 72, 120 h) in a growing chamber at 25 °C and photoperiod of 12 h. Scarification with 98% H2SO4 15 min increased GP and GR in both species. At 30 DAS, GP and GR of Le seeds were 34% and 0.97 seeds day-1, respectively. In Cl seeds, GP was 64% and GR 0.90 seeds day-1. Scarification with H2O at 80 °C 1 min also promoted germination in Cl (52%). At 120 h after seeding, Le and Cl seeds showed already a high GP with acid scarification (31% and 48%, respectively). In seeds of both species, scarification treatments affected WA and SEA during early germination. During this period, scarification treatments that increased GP also showed a higher α-D-galactosidase activity. The maximum enzyme activity was observed 72 h after hot water scarification in Cl (82.6 U/mg total protein), followed by acid scarification (54.5 U/mg total protein). In Le, the activity peak was 36 h after acid scarification (9.5 U/mg total protein). No relationship was observed between β-glucosidase activity and GP in both species. In conclusion, during early germination of both species, the increase in GP is accompanied by a rise in α-D-galactosidase activity between 36 and 72 h after seeding; and in Cl seeds, an alternative scarification treatment to increase GP may be the use of hot water

    The 2016 south Alboran earthquake (Mw= 6.4): A reactivation of the Ibero-Maghrebian region?

    On 25 January 2016, an earthquake of magnitude Mw = 6.4 occurred at the southern part of the Alboran Sea, between southern Spain and northern Morocco. This shock was preceded by a foreshock (Mw = 5.1) and fol- lowed by a long aftershock sequence. Focal mechanism of main shock has been estimated from slip inver- sion of body waves at teleseismic distances. Solution corresponds to left-lateral strike-slip motion, showing a complex bilateral rupture, formed by two sub-events, with most energy propagating along a plane oriented N30°E plane dipping to the NW. Relocation of larger events of the aftershock series, show two alignments of epicentres in NE-SW and NNE-SSW direction that intersect at the epicentre of the main shock. We have estimated the focal mechanisms of the largest aftershocks from moment tensor inversion at regional distances. We have obtained two families of focal mechanisms corresponding to strike slip for the NNE-SSW alignment and thrusting motion for the NE-SW alignment. Among the faults present in the area the Al Idrisi fault (or fault zone) may be a good candidate for the source of this earthquake. The study of Coulomb Failure Stress shows that it is possible that the 2016 earthquake was triggered by the previous nearby earthquakes of 1994 (Mw = 5.8) and 2004 (Mw = 6.3). The possible seismic reactivation of the central part of the Ibero-Maghrebian region is an open question, but it is clear that the occurrence of the 2016 earthquake confirms that from 1994 the seismicity of central part of IMR is increasing and that focal mechanism of largest earthquakes in this cen- tral part correspond to complex ruptures (or zone of fault)
