Differential Gene Transcription in Honeybee (Apis cerana) Larvae Challenged by Chinese Sacbrood Virus (CSBV)
Honey bees are economically important social insect. They are suffering from all kinds of pathogens, especially the virus. In response to pathogens, different immune pathways such as Toll, Imd, Jak-Stat and RNAi are involved. In the present study, the transcription analysis of 32 immune-related genes from Apis cerana challenged by Chinese sacbrood virus (CSBV), the most widely distributed virus in A. cerana, was carried out by qRT-PCR to provide cues for the antiviral mechanism and the effective control of bee viruses. The expression level of 22 genes were statistically changed, including 11 up-regulated genes (catus-2, lys-2, vir, s3a, mta1, faa, vhdl, co-1-iv, ago-1, ago-3, aub) in which 3 (ago-1, ago-3, aub) were related to RNAi pathway, and 11 down-regulated genes (kenny, pgrp-lc, pgrp-s2, abaecin, lys-1, lys-3, domeless, tepa, mlc, dscam, rpl8) related to Toll, Imd, and Jak-Stat pathways. The results indicated CSBV infection in A. cerana may activate a RNA-based antiviral immunity system. This work constituted the first report, under laboratory conditions, about induction of immune related genes in response to CSBV
TUBB, a robust biomarker with satisfying abilities in diagnosis, prognosis, and immune regulation via a comprehensive pan-cancer analysis
PurposeTUBB can encode a beta-tubulin protein. At present, the role of TUBB has not been ascertained in cancers. Hence, the importance of further systematic pan-cancer analyses is stressed to explore its value in the diagnosis, prognosis, and immune function of cancers.MethodsBy collecting and handling integrative data from the TCGA, Firehose, UCSC Xena, cBioPortal, GEO, CPTAC, TIMER2.0, TISCH, CellMiner, GDSC, and CTRP databases, we explored the potential diagnostic and prognostic roles of TUBB in pan-cancers from multiple angles. Moreover, the GSEA analysis was conducted to excavate the biological functions of TUBB in pan-cancers. In addition, survival profiles were described, and the differential expressions of TUBB in different molecular subtypes were discussed. Also, we utilized the cMAP function to search drugs or micro-molecules that have an impact on TUBB expressions.ResultsBased on the TCGA data, we found that TUBB was differentially expressed in a variety of tumors and showed an early-diagnostic value. Mutations, somatic copy number alterations, and DNA methylation would lead to its abnormal expression. TUBB expressions had relations with many clinical features. What’s more, TUBB expressions were validated to be related to many metabolism-related, metastasis-related, and immune-related pathways. High TUBB expressions were proved to have a great impact on the prognosis of various types of cancers and would affect the sensitivity of some drugs. We also demonstrated that the expression of TUBB was significantly correlated to immunoregulator molecules and biomarkers of lymphocyte subpopulation infiltration.ConclusionTUBB and its regulatory genes were systemically analyzed in this study, showing that TUBB had satisfying performances in disease diagnosing and prognosis predicting of multiple cancers. It could remodel the tumor microenvironment and play an integral role in guiding cancer therapies and forecasting responses to chemotherapy
Assessment of Material Dislocation Damage by Nonlinear Ultrasound
Under harsh environment or during service, the mechanical properties of materials or structure will deteriorate. Most of the simulations exhibit the phenomenon of nonlinearity by introducing the actual small defects, without considering dislocation. In this manuscript, subroutines are written to change the mechanical constitutive behaviour of materials. When the mechanical constitutive behaviour of the material is not linear any more, it is found that the propagation of ultrasonic wave in the material will show more obvious nonlinear phenomenon. Furthermore, the nonlinear detection coefficient is used to characterize the increase of harmonic components. This work provides a new idea for nonlinear ultrasonic testing
Simulation of Three Constitutive Behaviors Based on Nonlinear Ultrasound
Nonlinear ultrasound has attracted more and more attention. In classical acoustic nonlinear theory, the source of nonlinearity is the change of constitutive relation of materials. Structure response that distorts after a single tone ultrasound wave is important to detect imperfection. This is rarely found in current simulations. The current simulation always introduces defects which do not match to the classical acoustic nonlinear theory. In this manuscript, the recurrence expressions of three kinds of imperfect materials for subroutine are given. The verifying simulation model that is used for verifying recurrence equations and wave propagation model that are used for analysing the process of ultrasonic propagation are established. The results show that the two constitutive models are effective in the verifying simulation and the hysteresis material has some special characteristics. Finally, ultrasonic propagation in two types of materials produce the expected harmonics, which build foundations for simulations of nonlinear ultrasound
