Radioprotective profile of Urtica dioica L. seed extract on oxidative DNA-damage in liver tissue and whole blood of radiation-administered rats
It was aimed to investigate the radioprotective activity of Urtica dioica L. seed extract (UDSE) in the whole blood and liver of radiation-administered rats, both biochemically and immunohistochemically. 32 rats were divided into 4 groups (n:8). Control group (C): no administration for 10 days. Radiation group (IR): fed pellets for 10 days after exposure to radiation. Radiation + UDSE (IR+UDSE) group: exposed to radiation and fed UDSE for 10 days. UDSE group (UDSE): fed UDSE for 10 days. Radiation (5Gy) was given as a single fraction. 8-hydroxy-2-deoxyguanosine (8-OHdG) and deoxyguanosine (dG) levels were analyzed by biochemical method and glutathione peroxidase 1 (GPx-1) analyses were performed by immunohistochemical method in the liver and blood tissues of the rats. The increased 8-OHdG rates and decreased GPx-1 immunoreactivity was observed in the IR group. Those parameters were ameliorated in the IR+UDSE group when compared to the IR group. UDSE is likely to be a valuable radioprotector against the harmful effects of radiation
Effect of selenium against doxorubicin-induced oxidative stress, inflammation, and apoptosis in the brain of rats: Role of TRPM2 channel
Doxorubicin (DOX) is widely used as an anticancer drug in humans' various solid and haematological tumours. Although many studies on the toxic effect of DOX are used in different organs, its impact on brain tissue has yet to be adequately studied. This study investigated the protective effect of selenium (Se) and the role of transient receptor potential melastatin‐2 (TRPM2) channel activation against brain damage caused by DOX administration. Sixty rats were randomly divided into the sham, dimethyl sulfoxide (DMSO), DOX, DOX + Se, DOX + N-(p-amylcinnamoyl) anthranilic acid (ACA), and DOX + Se + ACA groups. The reactive oxygen species (ROS), poly [ADP-ribose] polymerase 1 (PARP1), and TRPM2 channel levels in brain tissues were measured by ELISA. In addition, a histopathological examination was performed in the cerebral cortex and hippocampal areas, and the TRPM2 channel, NF-κB, and caspase-3 expression were determined immunohistochemically. The levels of ROS, PARP1 and TRPM2 channel in the DOX group were higher than in the sham and DMSO groups (P < 0.05). However, these parameters were decreased in the in DOX+Se and DOX+ACA groups by the treatments of Se and ACA (P < 0.05). Also, we determined that Se and ACA treatment decreased the NF-κB, caspase-3, and TRPM2 channel expression in the cerebral cortex and hippocampal areas in the DOX-induced rats. The data showed that Se and/or ACA administration together with DOX administration could be used as a protective agent against DOX-induced brain damage
Hepatoprotective effect of Foeniculum vulgare essential oil: A carbon-tetrachloride induced liver fibrosis model in rats
Hepatoprotective activity of Foeniculum vulgare (fennel) essential oil was studied using a carbon tetrachloride- induced liver fibrosis model in rats. The hepatotoxicity produced by chronic carbon tetrachloride administration was found to be inhibited by Foeniculum vulgare essential oil with evidence of decreased levels of serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and bilirubin. Histopathological findings also suggest that Foeniculum vulgare essential oil prevents the development of chronic liver damage. The changes in body weights in the rats assigned to the study groups supported these biochemical and histopathological findings. The results of this study clearly indicate that Foeniculum vulgare essential oil has a potent hepatoprotective action against carbon tetrachloride-induced liver fibrosis in rats.
Polikistik over sendromlu hastalarda insülin direnci ve eser elementlerin ilişkisi
Objective: The study was investigate serum zinc (Zn),chromium (Cr), cobalt (Co) and manganese (Mn) concentrations in relation to hormone levels and polycystic ovarysyndrome (PCOS).Materials and Methods: Fourty women between 18-40years of age diagnosed with PCOS. We also seleceted 40healty volunteers as the control group. Serum Zn, Cr, Mn,follicular stimulating hormone (FSH), luteinizing hormone(LH), Dehydroepiandrosterone sulfate (DHEA-S), totaltestosterone (TT), sex hormone binding globulin (SHBG),insulin, glucose, cholesterol, triglyceride, High DensityLipoprotein (HDL-C) and Low density lipoprotein (LDLC) concentrations were determined.Results: Insulin, glucose, triglyceride, DHEA-S levelswere significantly higher in the PCOS group than in thecontrol group, while FSH and Mn levels were significantly lower in the HOMA-IR group than in the controlgroup (p< 0.05).Conclusions: A possible association is perceptible in ourstudy between some of serum trace elements and womenwith PCOS. The association attains a connotative levelwith the involvement of IR in it. Additional studies evaluating the effects of these elements supplementation wouldbe required to confirm the hypothesis as well as to ascertain whether insulin resistance in the presence of manganese deficiency can directly lead to oxidative stress.Amaç: Bu çalışma, polikistik over sendromunda (PKOS) hormon düzeyleriyle çinko (Zn), krom (Cr), kobalt (Co) ve mangan (Mn) konsantrasyonlarının ilişkisini incelemek için tasarlanmıştır. Materyal ve Metot: Çalışmaya endokrinoloji polikliniğine başvuran 18-40 yaş arası PKOS teşhisi konulan 40 kadın ve aynı sayıda sağlıklı gönüllü alındı. Serum Zn, Cr, Mn, Foliküler stimüle edici hormon (FSH), Lüteinleştirici Hormon (LH), Dehidroepiandrosteron (DHEA-S), Total Testosteron (TT), Seks hormonu bağlayıcı globulin (SHBG), insülin, glukoz, kolesterol, trigliserit, yüksek yoğunluklu lipoproteinler (HDL) ve düşük yoğunluklu lipoproteinler (LDL) konsantrasyonları analiz edildi. Bulgular: PKOS grubunda insülin, glukoz, trigliserit, DHEA-S düzeyleri kontrol grubuna göre anlamlı derecede yüksek iken, FSH ve Mn düzeyleri HOMA-IR grubunda kontrol grubuna göre anlamlı derecede düşüktü (p <0.05). Sonuç: Çalışmamızda serum eser elementleri ile PKOS arasında bir ilişki saptanırken, IR’nin dahil edilmesiyle ilişki düzeyi artmaktadır. Ayrıca, mangan eksikliği varlığında insülin direncinin doğrudan oksidatif strese yol açıp açamayacağını belirlemek için bu elementin takviyesinin etkilerini değerlendiren ek çalışmalara ihtiyaç olacağını da düşünmekteyiz
Do Avanafil and Zaprinast Change Some Selected Cytokine Levels In Ovariectomized Rat’s Liver?
Studies reported that phosphodiesterase-5 inhibitors (PDE-5Is) positively contributed to bone-mineral-density and thickness in rats with ovariectomy, which have the same condition with postmenapozal period. To explain the positive contribution mechanism on bone mineral density of PDE-5Is, we investigated the effect of zaprinast and avanafil on levels of some pro -or anti-resorptive cytokines in ovariectomized-rats. Albino female rats (8 months and 250-350 g) were used and four groups of equal-number were randomly assigned (n=6). Groups; was the sahm operated, positive control (OVX), Zaprinast and OVX, Avanafil and OVX groups, respectively. The levels of Estrogen, IL-1β, IL-6, IL-8, IL-10 and TNF-α were measured by ELISA kits, in liver of rats. IL-1β, IL-6, IL-8 and TNF-α levels were high in groups with OVX compared to sham group, while IL-10 levels were low. Also, IL-1β, IL-6, IL-8 and TNF-α levels were low in zaprinast and especially avanafil-treated groups with OVX and were similar to the sahm group values (p=0.001 for IL-1β, p=0.045 for IL-6, p=0.008 for IL-8, p=0.006 for IL-10, p=0.026 for TNF-α). Zaprinast and especially avanafil inhibited IL-1β, 8 and TNF-α and increased the IL-10 levels compared to the OVX group. This may support opinion that PDE-5Is enhance bone mineralization by inhibiting proresorptive cytokines
Relationship with Nephrotoxicity of Abemaciclib in Rats: Protective Effect of Curcumin
Abemaciclib (ABE) has been reported to cause gastrointestinal toxicity. Therefore, it is important to investigate the question of whether abemaciclib administration causes nephrotoxicity in the gastrointestinal tract, and if so, what pathophysiological pathways it follows. In this study, the relationship between ABE administration and nephrotoxicity and the protective effect of Curcumin (CMN) was investigated. Forty female rats were equally divided into the sham, dimethyl sulfoxide (DMSO), CMN, abemaciclib and ABE+CMN groups. Aquaporin (AQP) 1-7, TNF-α, IL-1β, intercellular adhesion molecule (ICAM)-1, IL-10 and IL-37 levels in serum and kidney tissue homogenates were measured by ELISA. In addition, Urea and Creatinine were measured in serum samples. Furthermore, histopathological examination was performed in kidney tissues and Bax, Caspase-3 and Bcl-2 expression levels were determined immunohistochemically. The levels of AQP1-7 and IL-10 in the ABE group were partially lower than in the other groups, while the ratio of TNFα, IL-1β, MDA, caspase-3 and Bax/Bcl2 were high. In addition, kidney tissue was examined histopathologically. However, AQP1 and 7 levels in the ABE+CMN group were higher than in the ABE group, while TNF-α, IL-1β, MDA, Caspase-3 levels and Bax/Bcl2 ratio were low. In addition, the poor histopathological changes in the ABE group were largely restored in the ABE+CMN. The data presented that ABE in rats can adversely affect functions and histology of kidneys through the increase in oxidative stress, pro-inflammatory cytokines and apoptosis, but CMN therapy may be protective against the nephrotoxic effects of AB
Rosmarinic acid inhibits some metabolic enzymes including glutathione S-transferase, lactoperoxidase, acetylcholinesterase, butyrylcholinesterase and carbonic anhydrase isoenzymes
Rosmarinic acid (RA) is a natural polyphenol contained in many aromatic plants with promising biological activities. Carbonic anhydrases (CAs, EC 4.2.1.1) are widespread and intensively studied metalloenzymes present in higher vertebrates. Acetylcholinesterase (AChE, E.C. 3.1.1.7) is intimately associated with the normal neurotransmission by catalysing the hydrolysis of acetylcholine to acetate and choline and acts in combination with butyrylcholinesterase (BChE) to remove acetylcholine from the synaptic cleft. Lactoperoxidase (LPO) is an enzyme involved in fighting pathogenic microorganisms, whereas glutathione S-transferases (GSTs) are dimeric proteins present both in prokaryotic and in eukaryotic organisms and involved in cellular detoxification mechanisms. In the present study, the inhibition effects of rosmarinic acid on tumour-associated carbonic anhydrase IX and XII isoenzymes, AChE, BChE, LPO and GST enzymes were evaluated. Rosmarinic acid inhibited these enzymes with K(i)s in the range between micromolar to picomolar. The best inhibitory effect of rosmarinic acid was observed against both AChE and BChE
The Status of Antioxidants and Oxidative Damage in Patients with COVID-19
Purpose: COVID-19 is a viral disease that has recently caused a pandemic and significantly affects human health. In this study, superoxide dismutase, glutathione peroxidase, glutathione, total thiol, natural thiol, disulfide, oxidative DNA damage and malondialdehyde levels in COVID-19 were investigated. Materials and Methods: Thirty-five patients and 35 healthy volunteers were included in this study. The diagnosis of COVID-19 was made by reverse transcriptase-polymerase chain reaction. Serum glutathione, glutathione peroxidase, superoxide dismutase, natural thiol, total thiol and disulphide levels by enzyme-linked immunosorbent assay and malondialdehyde and 8-hydroxy-2-deoxyguanosine/10⁶ deoxyguanosine levels by high-pressure liquid chromatography measured. Results: While serum superoxide dismutase, glutathione peroxidase, malondialdehyde, 8-hydroxy-2-deoxyguanosine/10⁶ deoxyguanosine, disulfide levels were higher in the COVID-19 patient group than in the healthy control group, glutathione, total thiol, natural thiol levels were lower. In addition, there was a negative correlation between 8-hydroxy-2-deoxyguanosine/10⁶ deoxyguanosine and glutathione, natural thiol and total thiol, and a positive correlation with disulfide. Conclusion: This study revealed that serum superoxide dismutase, glutathione peroxidase, malondialdehyde, 8-hydroxy-2-deoxyguanosine/10⁶ deoxyguanosine, and disulfide levels increased and glutathione, thiol and natural thiol levels decreased in COVID-19 patients. These results revealed that there was a decrease in antioxidant marker levels and an increase in oxidative stress markers in COVID-19 patients
The Effects of Some Phosphodiesterase 5 Inhibitors on Oxidative Stress, VEGF, BMP 2 and 9 in the Liver Tissue of Ovariectomized Rats
INTRODUCTION: Osteoporosis is an important health problem and there is no effective treatment yet. phosphodiesterase 5 inhibitors are promising agents for the treatment of osteoporosis. In this study, we aimed to determine the effects of phosphodiesterase 5 inhibitors (vardenafil, tadalafil, and udenafil) on bone morphogenic protein-2 and 9 (BMP-2 and 9), vascular endothelial growth factor (VEGF), and oxidative stress markers (malondialdehyde and CoQ10) in liver tissue of rats with ovariectomy-induced osteoporosis. METHODS: 50 Albino wistar female rats were randomly divided into 5 groups of 10 rats per group. Groups were the sham-operated, ovariectomise (OVEX), OVEX + Tadalafil, OVEX + udenafil and OVEX + vardenafil, respectively. VEGF, BMP-2 and 9 levels were measured by ELISA kits. To detect levels of MDA and CoQ10, we used high pressure liquid chromatography method. RESULTS: The levels of VEGF, BMP-2 and 9 levels in the groups that applied PDE-5 inhibitors were significantly higher than in the sham and OVEX groups. There was no significant difference between the OVEX+vardenafil and OVEX+udenafil groups in terms of VEGF, BMP-2 and 9 levels. The levels of MDA and CoQ10 were significantly lower in the groups that applied PDE5 inhibitors than in the OVEX group. When the histological and immunohistochemical results were examined, it was seen that angiogenesis was significantly higher in PDE-5 inhibitor groups. DISCUSSION AND CONCLUSION: In conclusion, we can say that these inhibitors may have positi ve effects on bone mineralization and remodelling by inducing the expression of VEGF, BMP-2 and 9 in liver tissue
Antioxidant and Antiradical Properties of Selected Flavonoids and Phenolic Compounds
Phenolic compounds and flavonoids are known by their antioxidant properties and one of the most important sources for humans is the diet. Due to the harmful effects of synthetic antioxidants such as BHA and BHT, natural novel antioxidants have become the focus of attention for protecting foods and beverages and reducing oxidative stress in vivo. In the current study, we investigated the total antioxidant, metal chelating, Fe3+ and Cu2+ reduction, and free radical scavenging activities of some phenolic and flavonoid compounds including malvin, oenin, ID-8, silychristin, callistephin, pelargonin, 3,4-dihydroxy-5-methoxybenzoic acid, 2,4,6-trihydroxybenzaldehyde, and arachidonoyl dopamine. The antioxidant properties of these compounds at different concentrations (10–30 μg/mL) were compared with those of reference antioxidants such as BHA, BHT, α-tocopherol, and trolox. Each substance showed dose-dependent antioxidant activity. Furthermore, oenin, malvin, arachidonoyl dopamine, callistephin, silychristin, and 3,4-dihydroxy-5-methoxybenzoic acid exhibited more effective antioxidant activity than that observed for the reference antioxidants. These results suggest that these novel compounds may function to protect foods and medicines and to reduce oxidative stress in vivo
