    Long non-coding RNA SNHG1 promotes bladder cancer progression by upregulating EZH2 and repressing KLF2 transcription

    Objective: Long Non-Coding RNAs (LncRNAs) act as an indispensable role in cancer development. The study aimed to investigate the role and mechanism of lncRNA Small Nucleolar RNA Host Gene 1 (SNHG1) in Bladder Cancer (BC) progression. Method: The expression, prognostic value, diagnostic value, and correlation of SNHG1, Enhancer of Zeste 2 polycomb repressive complex 2 subunit (EZH2), and Kruppel Like Factor 2 (KLF2) were analyzed through bioinformatics analysis. The expression was also validated in BC tissues and cell lines. Besides, their regulation and binding were tested via qPCR, Western blot, Dual-Luciferase Reporter Assay (DLRA), Argonaute RISC catalytic component 2-RNA Immunoprecipitation (AGO2-RIP), and Chromatin Immunoprecipitation (ChIP). A xenograft model in nude mice was also established. Results: SNHG1 was significantly overexpressed in BC tissues and cells. Importantly, SNHG1 was associated with poor survival, and ROC curves revealed high diagnostic values. Moreover, by CCK8, wound healing, transwell, and Western blot analysis, SNHG1 knockdown significantly inhibited the proliferation, migration, invasion, and epithelial-mesenchymal transition of BC cells. Additionally, in vivo experiments showed that silencing SNHG1 hindered tumorigenesis and tumor growth. Regarding mechanism, the results of AGO2-RIP, ChIP or DLRA showed that SNHG1 played different roles at diverse subcellular sites. In the cytoplasm, SNHG1 acted as a competing endogenous RNA for miR-137-3p to promote EZH2 expression. In the nucleus, SNHG1 could interact with EZH2 to inhibit KLF2 transcription. Conclusion: Our study elucidated that SNHG1 formed a regulatory network and played an oncogenic role in BC, which provided a novel therapeutic target for BC treatment

    Joint 3D Deployment and Resource Allocation for UAV-assisted Integrated Communication and Localization

    In this paper, we investigate an unmanned aerial vehicle (UAV)-assisted integrated communication and localization network in emergency scenarios where a single UAV is deployed as both an airborne base station (BS) and anchor node to assist ground BSs in communication and localization services. We formulate an optimization problem to maximize the sum communication rate of all users under localization accuracy constraints by jointly optimizing the 3D position of the UAV, and communication bandwidth and power allocation of the UAV and ground BSs. To address the intractable localization accuracy constraints, we introduce a new performance metric and geometrically characterize the UAV feasible deployment region in which the localization accuracy constraints are satisfied. Accordingly, we combine Gibbs sampling (GS) and block coordinate descent (BCD) techniques to tackle the non-convex joint optimization problem. Numerical results show that the proposed method attains almost identical rate performance as the meta-heuristic benchmark method while reducing the CPU time by 89.3%.Comment: The paper has been accepted for publication by IEEE Wireless Communications Letter

    MelodyGLM: Multi-task Pre-training for Symbolic Melody Generation

    Pre-trained language models have achieved impressive results in various music understanding and generation tasks. However, existing pre-training methods for symbolic melody generation struggle to capture multi-scale, multi-dimensional structural information in note sequences, due to the domain knowledge discrepancy between text and music. Moreover, the lack of available large-scale symbolic melody datasets limits the pre-training improvement. In this paper, we propose MelodyGLM, a multi-task pre-training framework for generating melodies with long-term structure. We design the melodic n-gram and long span sampling strategies to create local and global blank infilling tasks for modeling the local and global structures in melodies. Specifically, we incorporate pitch n-grams, rhythm n-grams, and their combined n-grams into the melodic n-gram blank infilling tasks for modeling the multi-dimensional structures in melodies. To this end, we have constructed a large-scale symbolic melody dataset, MelodyNet, containing more than 0.4 million melody pieces. MelodyNet is utilized for large-scale pre-training and domain-specific n-gram lexicon construction. Both subjective and objective evaluations demonstrate that MelodyGLM surpasses the standard and previous pre-training methods. In particular, subjective evaluations show that, on the melody continuation task, MelodyGLM gains average improvements of 0.82, 0.87, 0.78, and 0.94 in consistency, rhythmicity, structure, and overall quality, respectively. Notably, MelodyGLM nearly matches the quality of human-composed melodies on the melody inpainting task

    Viable but nonculturable bacteria and their resuscitation: implications for cultivating uncultured marine microorganisms

    Culturing has been the cornerstone of microbiology since Robert Koch first successfully cultured bacteria in the late nineteenth century. However, even today, the majority of microorganisms in the marine environment remain uncultivated. There are various explanations for the inability to culture bacteria in the laboratory, including lack of essential nutrients, osmotic support or incubation conditions, low growth rate, development of micro-colonies, and the presence of senescent or viable but nonculturable (VBNC) cells. In the marine environment, many bacteria have been associated with dormancy, as typified by the VBNC state. VBNC refers to a state where bacteria are metabolically active, but are no longer culturable on routine growth media. It is apparently a unique survival strategy that has been adopted by many microorganisms in response to harsh environmental conditions and the bacterial cells in the VBNC state may regain culturability under favorable conditions. The resuscitation of VBNC cells may well be an important way to cultivate the otherwise uncultured microorganisms in marine environments. Many resuscitation stimuli that promote the restoration of culturability have so far been identified; these include sodium pyruvate, quorum sensing autoinducers, resuscitation-promoting factors Rpfs and YeaZ, and catalase. In this review, we focus on the issues associated with bacterial culturability, the diversity of bacteria entering the VBNC state, mechanisms of induction into the VBNC state, resuscitation factors of VBNC cells and implications of VBNC resuscitation stimuli for cultivating these otherwise uncultured microorganisms. Bringing important microorganisms into culture is still important in the era of high-throughput sequencing as their ecological functions in the marine environment can often only be known through isolation and cultivation

    The Efficacy and Safety of Chinese Herbal Decoction in Type 2 Diabetes: A 5-Year Retrospective Study

    Background. The study was designed to assess the efficacy and safety of Chinese herbal decoction in treating outpatients with T2DM. Methods. All patients enrolled received decoction for at least 6 months. The primary outcome was the control rate of HbA1c and the change in HbA1c. FPG, 2hPG, HOMA-IR, and HOMA-β were also collected and evaluated. Results. The control rates after treatment at months 6, 12, 18, 24, 36, 48, and 60 were 45.07%, 52.78%, 47.22%, 45.83%, 50.00%, 57.14%, and 40.00%. Multiple linear regression showed the change of HbA1c has a significant relationship with the baseline HbA1c and duration of DM and BMI (p<0.05). Both FPG and 2hPG levels significantly decreased compared to the baseline (p<0.05). Chinese herbal decoction also improved islet cell function with decreased HOMA-IR and increased HOMA-β (p<0.05). 19 and 4 subjects deactivated the antidiabetes drugs or insulin, respectively, after taking decoction. One subject developed DKD and one developed DPN, and another subject showed abnormal liver function which was irrelevant to decoction treatment. Conclusions. Chinese herbal decoction significantly enhanced the hypoglycemic action and had certain effect on protecting islet cell function. As a candidate diabetes therapy, it may reduce the use of antidiabetes drugs and slow the progression to diabetes complications

    Modeling seismic wave propagation in the Loess Plateau using a viscoacoustic wave equation with explicitly expressed quality factor

    The thick Quaternary loess on the Loess Plateau of China produces strong seismic attenuation, resulting in weak reflections from subsurface exploration targets. Accurately simulating seismic wavefield in the Loess Plateau is important for guiding subsequent data processing and interpretation. We present a 2D/3D wavefield simulation method for the Loess Plateau using a viscoacoustic wave equation with explicitly expressed quality factor. To take into account the effect of irregular surface, we utilize a vertically deformed grid to represent the topography, and solve the viscoacoustic wave equation in a regular computational domain that conforms to topographic surface. Grid deformation introduces the partial derivatives such as ∂vx/∂z and ∂vy/∂z in the wave equation, which is difficult to be accurately computed using traditional staggered-grid finite-difference method. To mitigate this issue, a finite-difference scheme based on a fully staggered-grid is adopted to solve the viscoacoustic wave equation. Numerical experiments for a simple layer model and 2D/3D realistic Loess Plateau models demonstrate the feasibility and adaptability of the proposed method. The 3D modeling results show comparable amplitude and waveform characteristics to the field data acquired from the Chinese Loess Plateau, suggesting a good performance of the proposed modeling method

    Autonomous detection of damage to multiple steel surfaces from 360° panoramas using deep neural networks

    Structural health assessments are essential for infrastructure. By using an autonomous panorama vision‐based inspection system, the limitations of the human cost and safety factors of previously time‐consuming tasks have been overcome. The main damage detection challenges to panorama images are (1) the lack of annotated panorama defect image data, (2) detection in high‐resolution images, and (3) the inherent distortion disturbance for panorama images. In this paper, a new PAnoramic surface damage DEtection Network (PADENet) is presented to solve the challenges by (a) using an unmanned aerial vehicle to capture panoramic images and a distorted panoramic augmentation method to expand the panoramic dataset, (b) employing the proposed multiple projection methods to process high‐resolution images, and (c) modifying the faster region‐based convolutional neural network and training via transfer learning on VGG‐16, which improves the precision for detecting multiple types of damage in distortion. The results show that the proposed method is optimal for surface damage detection

    Measurement and correlation of the solubility of telmisartan (form A) in nine different solvents from 277.85 to 338.35 K

    The solubility of telmisartan (form A) in nine organic solvents (chloroform, dichloromethane, ethanol, toluene, benzene, 2-propanol, ethyl acetate, methanol and acetone) was determined by a laser monitoring technique at temperatures from 277.85 to 338.35 K. The solubility of telmisartan (form A) in all of the nine solvents increased with temperature as did the rates at which the solubility increased except in chloroform and dichloromethane. The mole fraction solubility in chloroform is higher than that in dichloromethane, which are both one order of magnitude higher than those in the other seven solvents at the experimental temperatures. The solubility data were correlated with the modified Apelblat equation and λh equations. The results show that the λh equation is in better agreement with the experimental data than the Apelblat equation. The relative root mean square deviations (σ) of the λh equation are in the range from 0.004 to 0.45 %. The dissolution enthalpies, entropies and Gibbs energies of telmisartan in these solvents were estimated by the Van’t Hoff equation and the Gibbs equation. The melting point and the fusion enthalpy of telmisartan were determined by differential scanning calorimetry
