    Molecular characters and recombinant expression of the carboxylesterase gene of the meadow moth Loxostege sticticalis L. (Lepidoptera: Pyralidae)

    Insect carboxylesterases are enzymes that catalyze the hydrolysis of ester and amide moieties, which play important roles in insecticide resistance, specifically allelochemical tolerance and developmental regulation. We obtained the cDNA encoding carboxylesterase gene of Loxostege sticticalis (LstiCarE) by a cDNA library screen. The full cDNA of LstiCarE is 1,980 bp in length, containing an open reading frame (ORF) of 1,875 bp, which encodes a preprotein of 625 amino acid residues. The LstiCarE contains the catalytic triad (Ser-His-Glu), the pentapeptide GxSxG motif and GxxHxxD/E motif, which are typical characteristic of esterases. The GxSxG and GxxHxxD/E motifs of LstiCarE are modified as GCSAG and GxxHxxQ, respectively. The 3-D model structure of LstiCarE showed that Ser197, His440 and Glu321 are aggregated together, which form the catalytic triad. The recombinant LstiCarE were successfully expressed in BL21 cells using recombinant plasmid DNA, and showed high carboxylesterase activity. However, the biochemical and physiological functions of carboxylesterase gene in L. sticticalis requires further investigation.Key words: Carboxylesterase gene, Loxostege sticticalis, recombinant expression

    Polymorphism of the pig-implantation protein 3 (preis3) gene and its association with litter size traits

    The pre-implantation protein 3 (prei3), which might play a role in pre-implantation embryogenesis, is one of the promising candidate genes for litter size traits in pigs. In this study, a single nucleotide polymorphism (SNP: T802G) in intron 6 of the pig prei3 gene was detected and a genotyping assay for this SNP was developed. An association study for this SNP with litter size was performed in two independent populations. One population consisted of crossbred sows derived from Landrace, Large White, Chinese Tongcheng and/or Chinese Meishan (Line DIV). The other population constituted of crossbred animals derived from Chinese Qingping and Duroc (QD). Statistical analysis demonstrated that, in first parity, 2.65 more piglets were born and 3.82 more piglets were born alive in sows in Line DIV with genotype TT than with genotype GG. For second and subsequent litters, in both the DIV and QD lines there were significant differences in the number of piglets born alive between TG and GG sows, with the TG sows producing more piglets born alive than the GG sows. These results suggest that the prei3 SNP is significantly associated with litter size in the two populations studied, and could be useful in selection for increasing litter size in pigs.     Compensation defects in annealed undoped liquid encapsulated Czochralski InP

    As-grown undoped n-type semiconducting and annealed undoped semi-insulating (SI) liquid encapsulated Czochralski (LEC) InP has been studied by temperature dependent Hall measurement, photoluminescence spectroscopy, infrared absorption, and photocurrent spectroscopy. P-type conduction SI InP can frequently be obtained by annealing undoped LEC InP. This is caused by a high concentration of thermally induced native acceptor defects. In some cases, it can be shown that the thermally induced n-type SI property of undoped LEC InP is caused by a midgap donor compensating for the net shallow acceptors. The midgap donor is proposed to be a phosphorus antisite related defect. Traps in annealed SI InP have been detected by photocurrent spectroscopy and have been compared with reported results.     PRED_PPI: a server for predicting protein-protein interactions based on sequence data with probability assignment

    <p>Abstract</p> <p>Background</p> <p>Protein-protein interactions (PPIs) are crucial for almost all cellular processes, including metabolic cycles, DNA transcription and replication, and signaling cascades. Given the importance of PPIs, several methods have been developed to detect them. Since the experimental methods are time-consuming and expensive, developing computational methods for effectively identifying PPIs is of great practical significance.</p> <p>Findings</p> <p>Most previous methods were developed for predicting PPIs in only one species, and do not account for probability estimations. In this work, a relatively comprehensive prediction system was developed, based on a support vector machine (SVM), for predicting PPIs in five organisms, specifically humans, yeast, <it>Drosophila</it>, <it>Escherichia coli</it>, and <it>Caenorhabditis elegans</it>. This PPI predictor includes the probability of its prediction in the output, so it can be used to assess the confidence of each SVM prediction by the probability assignment. Using a probability of 0.5 as the threshold for assigning class labels, the method had an average accuracy for detecting protein interactions of 90.67% for humans, 88.99% for yeast, 90.09% for <it>Drosophila</it>, 92.73% for <it>E. coli</it>, and 97.51% for <it>C. elegans</it>. Moreover, among the correctly predicted pairs, more than 80% were predicted with a high probability of ≥0.8, indicating that this tool could predict novel PPIs with high confidence.</p> <p>Conclusions</p> <p>Based on this work, a web-based system, Pred_PPI, was constructed for predicting PPIs from the five organisms. Users can predict novel PPIs and obtain a probability value about the prediction using this tool.     Formation of P In defect in annealed liquid-encapsulated Czochralski InP

    Fourier transform infrared spectroscopy measurements have been carried out on liquid-encapsulated Czochralski-grown undoped InP wafers, which reproducibly become semi-insulating upon annealing in an ambient of phosphorus at 800-900°C. The measurements reveal a high concentration of hydrogen complexes in the form V InH 4 existing in the material before annealing in agreement with recent experimental studies. It is argued that the dominant and essential process producing the semi-insulating behavior is the compensation produced by an EL 2-like deep donor phosphorus antisite defect, which is formed by the dissociation of the hydrogen complexes during the process of annealing. The deep donor compensates acceptors, the majority of which are shallow residual acceptor impurities and deep hydrogen associated V In and isolated V In levels, produced at the first stage of the dissociation of the V InH 4 complex. The high concentration of indium vacancies produced by the dissociation are the precursor of the EL 2-like phosphorus antisite. These results show the importance of hydrogen on the electrical properties of InP and indicate that this largely results from low formation energy of the complex V InH 4 in comparison with that of an isolated V In. © 1998 American Institute of Physics.published_or_final_versio
