    The effect of titanium (Ti) and titanium 500 (ti 500) implantation on the activation of rat macrophage subgroups

Introduction: Nowadays it is necessary to make new researches in order to solve the problems related to the prolongation of life and related health problems, especially fractures and spinal degeneration. The biocompatibility, mechanical compatibility, morphological compatibility and osseointegration properties of the implant material are very important. In order to prevent unwanted side effects in the use of biomaterials, new strategies need to be developed. Implants, where they will be implanted and their functions will vary according to the characteristics of the material used. The most commonly used metallic materials are 316L stainless steel, Co-Cr alloys and Ti alloys. Objective: To demostrate the effect of Titanium and Titanium 500 on activation of macropages Material and Methods: Our research was performed in the Laboratory of Cytokines and Receptors in the Department of Physiology of Cerrahpasa Medical Faculty, Istanbul University-Cerrahpasa. Our research has been approved by the Animal Experiments Local Ethics Committee of Bezmialem Vakif University (Approval Number:2017/218). In order to control the rejection of the patient with specific inflammation caused by titanium implantation, we analyzed the first triggered cells of the innate immun system, especially macrophages and sub-groups (M1, M2a, M2b, M2c), by implanting Titanium and Titanium 500 into the spinal region in Wistar albino male rats. According to the Power Analysis statistic program, 3 different groups of Wistar albino species male rats with a weight of 250 - 300 grams and 10-12 weeks of age were formed. Group I (n: 8, Sham group (Control)), Group II (n: 8, Titanium alloy), Group III (n: 8, Titanium 500). No implant was used in Group I (sham group). Only surgical stress was applied to the rats and they were closed again. In Group II and Group III, the rods were placed on the lamina. Peripheral blood samples were collected on the 1st, 3rd, 5th and 7th days following the implantation phase. M1 macrophages (CCL3, CCL4, CXCL9, IL-23), M2a macrophages (CD163, CD206), M2b macrophages (CCL1), M2c macrophage (SLAM) markers were examined by ELISA (Enzyme-Linked ImmunoSorbent Assay) method. Groups were compared with one-way analysis of variance (ANOVA). For the paired comparison of significant variables, a statistically significant difference (HSD) test was used for Tukey's homogeneous variance variables. Statistical significance was set at p < 0.05. Results and Conclusions: We believe that our analysis results will be helpful in the control, prevention, immunological and therapeutic methods of the reactions (inflammation, rejection, etc.) that may occur in patients who are implanted with Titanium (Ti) and Titanium 500 (Ti 500) implants (spinal, orthopedic, dental etc.)

    Genotypic identification and technological characterization of lactic acid bacteria isolated from traditional Turkish Kargi tulum cheese

    Kargi tulum cheese is an artisanal cheese produced through the spontaneous fermentation of raw milk ripened inside a goat-skin bag. The objective of this study was to characterize the dominant species of natural lactic acid bacteria (LAB) found in Kargi tulum cheese. Some technologically important properties of LAB isolates were also determined. Seven samples of cheese of different levels of ripeness were taken from local producers and 97 isolates were obtained from these samples. Non-spore forming, Gram-positive, catalase- and oxidase-negative isolates were assessed as LAB. Based on genotypic characterization, the dominant LAB were identified as Lactobacillus paracasei (43.3%), Lactobacillus plantarum (23.7%), Enterococcus durans (6.2%), Streptococcus thermophilus (6.2%), Lactobacillus brevis (5.2%), Enterococcus faecium (5.2%), Lactobacillus fermentum (4.1%) and Lactobacillus pentosus (1%). Homofermentative strains such as L. paracasei, L. plantarum, S. thermophilus; E. durans and E. faecium were selected as probable starter cultures. These strains were tolerant to 6.5% salt. They are also characterized by acidification ability (from pH = 6.6 to 4.7 to 6.0, in 6 to 8 h), low proteinase and high peptidase, esterase, esterase-lipase, β-galactosidase and β- glucosidase activities.     The anatomical studies on two Biarum (Araceae) species in Turkey

    In this study, anatomical properties of Biarum marmarisense (P. C. Boyce) P. C. Boyce and Biarum pyrami (Schott) Engl. var. pyrami were examined. The specimens were collected from Antalya province, South West of Anatolia. Degree of spathe tube margin connection, distribution of pistillodes and staminate-pistillate flower zone interstice length characters are important in Biarum Schott. taxonomy at specific level. The cross-sections of root, stem and leaf parts of the plant were examined and demonstrated by photographs. Although thicknesses of the cell wall of the endodermis and xylem strands are distinguishing features, raphide crystals are same feature in both species. B. marmarisense has observed paracytic stoma types while B. pyrami var. pyrami has anomocytic stoma types. Most of the anatomical properties were similar to the other members of the Araceae family

    Overcoming cross-cultural group work tensions: mixed student perspectives on the role of social relationships

    As universities worldwide rapidly internationalise, higher education classrooms have become unique spaces for collaboration between students from different countries. One common way to encourage collaboration between diverse peers is through group work. However, previous research has highlighted that cross-cultural group work can be challenging and has hinted at potential social tensions. To understand this notion better, we have used robust quantitative tools in this study to select 20 participants from a larger classroom of 860 students to take part in an in-depth qualitative interview about cross-cultural group work experiences. Participant views on social tensions in cross-cultural group work were elicited using a unique mediating artefact method to encourage reflection and in-depth discussion. In our analysis of emergent interview themes, we compared student perspectives on the role of social relationships in group work by their academic performance level. Our findings indicated that all students interviewed desired the opportunity to form social relationships with their group work members, but their motivations for doing so varied widely by academic performance level

    The effects of prenatal exposure to a 'junk food' diet on offspring food preferences and fat deposition can be mitigated by improved nutrition during lactation

    Exposure to a maternal junk food (JF) diet in utero and during the suckling period has been demonstrated to increase the preference for palatable food and increase the susceptibility to diet-induced obesity in adult offspring. We aimed to determine whether the effects of prenatal exposure to JF could be ameliorated by cross-fostering offspring onto dams consuming a standard rodent chow during the suckling period. We report here that when all offspring were given free access to the JF diet for 7 weeks from 10 weeks of age, male offspring of control (C) or JF dams that were cross-fostered at birth onto JF dams (C-JF, JF-JF), exhibited higher fat (C-C: 12.3 ± 0.34 g/kg/day; C-JF: 14.7 ± 1.04 g/kg/day; JF-C: 11.5 ± 0.41 g/kg/day; JF-JF: 14.0 ± 0.44 g/kg/day; P < 0.05) and overall energy intake (C-C: 930.1 ± 18.56 kJ/kg/day; C-JF: 1029.0 ± 82.9 kJ/kg/day; JF-C: 878.3 ± 19.5 kJ/kg/day; JF-JF: 1003.4 ± 25.97 kJ/kg/day; P < 0.05) than offspring exposed to the JF diet only before birth (JF-C) or not at all (C-C). Female offspring suckled by JF dams, despite no differences in food intake, had increased fat mass as percentage of body weight (C-C: 19.9 ± 1.33%; C-JF: 22.8 ± 1.57%; JF-C: 17.4 ± 1.03%; JF-JF: 22.0 ± 1.0%; P < 0.05) after 3 weeks on the JF diet. No difference in fat mass was observed in male offspring. These findings suggest that the effects of prenatal exposure to a JF diet on food preferences in females and susceptibility to diet-induced obesity in males can be prevented by improved nutrition during the suckling period.J.     An improved constraint satisfaction adaptive neural network for job-shop scheduling

    Copyright @ Springer Science + Business Media, LLC 2009This paper presents an improved constraint satisfaction adaptive neural network for job-shop scheduling problems. The neural network is constructed based on the constraint conditions of a job-shop scheduling problem. Its structure and neuron connections can change adaptively according to the real-time constraint satisfaction situations that arise during the solving process. Several heuristics are also integrated within the neural network to enhance its convergence, accelerate its convergence, and improve the quality of the solutions produced. An experimental study based on a set of benchmark job-shop scheduling problems shows that the improved constraint satisfaction adaptive neural network outperforms the original constraint satisfaction adaptive neural network in terms of computational time and the quality of schedules it produces. The neural network approach is also experimentally validated to outperform three classical heuristic algorithms that are widely used as the basis of many state-of-the-art scheduling systems.     Exposure of neonatal rats to maternal cafeteria feeding during suckling alters hepatic gene expression and DNA methylation in the insulin signalling pathway

    Nutrition in early life is a determinant of lifelong physiological and metabolic function. Diseases that are associated with ageing may, therefore, have their antecedents in maternal nutrition during pregnancy and lactation. Rat mothers were fed either a standard laboratory chow diet (C) or a cafeteria diet (O) based upon a varied panel of highly palatable human foods, during lactation. Their offspring were then weaned onto chow or cafeteria diet giving four groups of animals (CC, CO, OC, OO n=9-10). Livers were harvested 10 weeks post-weaning for assessment of gene and protein expression, and DNA methylation. Cafeteria feeding post-weaning impaired glucose tolerance and was associated with sex-specific altered mRNA expression of peroxisome proliferator activated receptor gamma (PPARg) and components of the insulin-signalling pathway (Irs2, Akt1 and IrB). Exposure to the cafeteria diet during the suckling period modified the later response to the dietary challenge. Post-weaning cafeteria feeding only down-regulated IrB when associated with cafeteria feeding during suckling (group OO, interaction of diet in weaning and lactation P=0.041). Responses to cafeteria diet during both phases of the experiment varied between males and females. Global DNA methylation was altered in the liver following cafeteria feeding in the post-weaning period, in males but not females. Methylation of the IrB promoter was increased in group OC, but not OO (P=0.036). The findings of this study add to a growing evidence base that suggests tissue function across the lifespan a product of cumulative modifications to the epigenome and transcriptome, which may be both tissue and sex-specific

    Topical Review: Development of overgrown semi-polar GaN for high efficiency green/yellow emission

    The most successful example of large lattice-mismatched epitaxial growth of semiconductors is the growth of III-nitrides on sapphire, leading to the award of the Nobel Prize in 2014 and great success in developing InGaN-based blue emitters. However, the majority of achievements in the field of III-nitride optoelectronics are mainly limited to polar GaN grown on c-plane (0001) sapphire. This polar orientation poses a number of fundamental issues, such as reduced quantum efficiency, efficiency droop, green and yellow gap in wavelength coverage, etc. To date, it is still a great challenge to develop longer wavelength devices such as green and yellow emitters. One clear way forward would be to grow III-nitride device structures along a semi-/non-polar direction, in particular, a semi-polar orientation, which potentially leads to both enhanced indium incorporation into GaN and reduced quantum confined Stark effects. This review presents recent progress on developing semi-polar GaN overgrowth technologies on sapphire or Si substrates, the two kinds of major substrates which are cost-effective and thus industry-compatible, and also demonstrates the latest achievements on electrically injected InGaN emitters with long emission wavelengths up to and including amber on overgrown semi-polar GaN. Finally, this review presents a summary and outlook on further developments for semi-polar GaN based optoelectronics

    Winter Survival of Individual Honey Bees and Honey Bee Colonies Depends on Level of Varroa destructor Infestation

    Background: Recent elevated winter loss of honey bee colonies is a major concern. The presence of the mite Varroa destructor in colonies places an important pressure on bee health. V. destructor shortens the lifespan of individual bees, while long lifespan during winter is a primary requirement to survive until the next spring. We investigated in two subsequent years the effects of different levels of V. destructor infestation during the transition from short-lived summer bees to long-lived winter bees on the lifespan of individual bees and the survival of bee colonies during winter. Colonies treated earlier in the season to reduce V. destructor infestation during the development of winter bees were expected to have longer bee lifespan and higher colony survival after winter. Methodology/Principal Findings: Mite infestation was reduced using acaricide treatments during different months (July, August, September, or not treated). We found that the number of capped brood cells decreased drastically between August and November, while at the same time, the lifespan of the bees (marked cohorts) increased indicating the transition to winter bees. Low V. destructor infestation levels before and during the transition to winter bees resulted in an increase in lifespan of bees and higher colony survival compared to colonies that were not treated and that had higher infestation levels. A variety of stress-related factors could have contributed to the variation in longevity and winter survival that we found between years. Conclusions/Significance: This study contributes to theory about the multiple causes for the recent elevated colony losses in honey bees. Our study shows the correlation between long lifespan of winter bees and colony loss in spring. Moreover, we show that colonies treated earlier in the season had reduced V. destructor infestation during the development of winter bees resulting in longer bee lifespan and higher colony survival after winter
