    Numerical Simulations for Nonlinear Waves Interaction with Multiple Perforated Quasi-Ellipse Caissons

    A three-dimensional numerical flume is developed to study cnoidal wave interaction with multiple arranged perforated quasi-ellipse caissons. The continuity equation and the Navier-Stokes equations are used as the governing equation, and the VOF method is adopted to capture the free surface elevation. The equations are discretized on staggered cells and then solved using a finite difference method. The generation and propagation of cnoidal waves in the numerical flume are tested first. And the ability of the present model to simulate interactions between waves and structures is verified by known experimental results. Then cnoidal waves with varying incident wave height and period are generated and interact with multiple quasi-ellipse caissons with and without perforation. It is found that the perforation plays an effective role in reducing wave runup/rundown and wave forces on the caissons. The wave forces on caissons reduce with the decreasing incident wave period. The influence of the transverse distance of multiple caissons on wave forces is also investigated. A closer transverse distance between caissons can produce larger wave forces. But when relative adjacent distance L/D (L is the transverse distance and D is the width of the quasi-ellipse caisson) is larger than 3, the effect of adjacent distance is limited

    Association between Portal Vein Thrombosis and Survival in Non-Liver-Transplant Patients with Liver Cirrhosis: A Systematic Review of the Literature

    A systematic review of the literature was performed to analyze the association between portal vein thrombosis (PVT) and survival in non-liver-transplant patients with liver cirrhosis. PubMed, EMBASE, and Cochrane Library databases were searched for all relevant papers which evaluated the prognostic value of PVT in predicting the survival of liver cirrhosis. Meta-analyses were not conducted because the ways of data expression and lengths of follow-up were heterogeneous among studies. Overall, 13 papers were included. The 5-day, 6-week, and 1-year mortality were investigated in 1, 3, and 1 studies, respectively; and all of them were not significantly different between cirrhotic patient with and without PVT. By comparison, the 3-year mortality was reported in 1 study; and it was significantly increased by the presence of PVT. The overall mortality was analyzed in 5 studies; and the association with overall mortality and PVT was significant in 4 studies, but not in another one. However, as for the cirrhotic patients undergoing surgical or interventional shunts, the overall mortality was not significantly associated with the presence of PVT in 4 studies. In conclusion, the presence of PVT might be associated with the long-term mortality in non-liver-transplant patients with liver cirrhosis, but not with the short-term mortality

    Confinement of carbon dots localizing to the ultrathin layered double hydroxides toward simultaneous triple-mode bioimaging and photothermal therapy

    It is a great challenge to develop multifunctional nanocarriers for cancer diagnosis and therapy. Herein, versatile CDs/ICG-uLDHs nanovehicles for triple-modal fluorescence/photoacoustic/two-photon bioimaging and effective photothermal therapy were prepared via a facile self-assembly of red emission carbon dots (CDs), indocyanine green (ICG) with the ultrathin layered double hydroxides (uLDHs). Due to the J-aggregates of ICG constructed in the self-assembly process, CDs/ICG-uLDHs was able to stabilize the photothermal agent ICG and enhanced its photothermal efficiency. Furthermore, the unique confinement effect of uLDHs has extended the fluorescence lifetime of CDs in favor of bioimaging. Considering the excellent in vitro and in vivo phototherapeutics and multimodal imaging effects, this work provides a promising platform for the construction of multifunctional theranostic nanocarrier system for the cancer treatment

    MiR-24-3p attenuates IL-1β-induced chondrocyte injury associated with osteoarthritis by targeting BCL2L12

    Abstract Background MiR-24-3p has been reported to be involved in an osteoarthritis (OA)-resembling environment. However, the functional role and underlying mechanism of miR-24-3p in chondrocyte injury associated with OA remains unknown. Methods The expression of miR-24-3p was determined using reverse transcription quantitative PCR analysis in OA cases and control patients, as well as IL-1β-stimulated chondrocyte cell line CHON-001. The cell viability was analyzed by CCK-8 assay. Apoptosis status was assessed by caspase-3 activity detection. The pro-inflammatory cytokines (TNF-α and IL-18) were determined using ELISA assay. The association between miR-24-3p and B cell leukemia 2-like 12 (BCL2L12) was confirmed by luciferase reporter assay. Results We first observed that miR-24-3p expression level was lower in the OA cases than in the control patients and IL-1β decreased the expression of miR-24-3p in the chondrocyte CHON-001. Functionally, overexpression of miR-24-3p significantly attenuated IL-1β-induced chondrocyte injury, as reflected by increased cell viability, decreased caspase-3 activity, and pro-inflammatory cytokines (TNF-α and IL-18). Western blot analysis showed that overexpression of miR-24-3p weakened IL-1β-induced cartilage degradation, as reflected by reduction of MMP13 (Matrix Metalloproteinase-13) and ADAMTS5 (a disintegrin and metalloproteinase with thrombospondin motifs-5) protein expression, as well as markedly elevation of COL2A1 (collagen type II). Importantly, BCL2L12 was demonstrated to be a target of miR-24-3p. BCL2L12 knockdown imitated, while overexpression significantly abrogated the protective effects of miR-24-3p against IL-1β-induced chondrocyte injury. Conclusions In conclusion, our work provides important insight into targeting miR-24-3p/BCL2L12 axis in OA therapy

    Dickkopf-1 Expression Is a Novel Prognostic Marker for Gastric Cancer

    Aim. To investigate the involvement of Dickkopf-1 expression in gastric cancer. Methods. Dickkopf-1 mRNA and protein expression were determined by real-time quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) and immunohistochemistry in specimens of primary cancer and their adjacent noncancerous tissues in gastric cancer patients. Results. Dickkopf-1 mRNA and protein expression levels were both significantly upregulated in gastric cancer lesions compared with adjacent noncancerous tissues. Its positive expression was correlated with depth of invasion, vessel invasion, lymph node and distant metastasis, and TNM stage of tumors. Additionally, in stages I, II, and III gastric cancers, the 5- year survival rate of patients with a high expression of Dickkopf-1 was significantly lower than that in patients with low expression. In stage IV, Dickkopf-1 expression did not correlate with the 5-year survival rate. Further multivariate analysis suggested that the up-regulation of Dickkopf-1 was an independent prognostic indicator for gastric cancer. Conclusion. A subset of cases with gastric cancer revealed the up-regulation of Dickkopf-1, which was associated with a progressive pathological feature and an aggressive clinical course. Therefore, Dickkopf-1 expression may be predictor for poor prognosis in patients with gastric cancer. This is the first report describing the involvement of Dickkopf-1 in gastric cancer

    Strength analysis and optimization of alkali activated slag backfills through response surface methodology

    The significant difference in water-to-binder ratio, activator type and concentration between alkali-activated slag (AAS) paste/mortar/concrete and AAS-based cemented paste backfill (AAS-CPB) means that previous results related to the properties and mix optimization of AAS materials cannot be directly translated to AAS-CPB. This study statistically identifies the effect of key influential variables such as silicate modulus, slag fineness and activator concentration on 3- and 28 day unconfined compressive strength (UCS) of AAS-CPB by central composite design (CCD) established in response surface methodology (RSM). In this study, the prominence of independent variables and their relations are investigated by using ANOVA (analysis of variance) having a significant level of 0.05. ANOVA results certify that there is a strong link between the level of variable contribution on UCS performance of AAS-CPB and curing age. Obviously, silicate modulus and activator concentration are the most major variables influencing UCS at 3 and 28 days, respectively. Increased fineness of slag and higher pH of pore solution enhance 3 day UCS, but restrain the further hydration of unreacted slag and subsequent the gain in strength at advanced curing ages.
