    The lethal concentration of water-borne copper in Chinese mitten crab Eriocheir sinensis larvae was tested by exposing the animals to 0, 0.1, 0.2, 0.3, 0.5 and 0.8 mg Cu L-1 at 20°C for 96 h. The 96-h median lethal concentration (LC50) and its corresponding 95% confident interval was estimated on zoea 1 larvae and megalopa larvae, respectively. Acute dissolved copper toxicity was higher for zoea 1 larvae (0.16 mg L-1) than for megalopa larvae (0.21 mg L-1). The antioxidant status, metallothionein-I mRNA expression and physiological response of the crab to copper toxicity was further investigated by exposing the megalopa larvae to 0, 0.08 and 0.16 mg Cu L-1 for 96 h. The superoxide dismutase activity, catalase activity, glutathione S-transferase (GST) activity and lipid peroxidation content of megalopa larvae increased concomitantly with the exposure time and copper concentration. MT-I mRNA expression levels were positively correlated with both the concentration and duration of copper exposure. The oxygen consumption and respiratory quotient of megalopa larvae in response to 0.16 mg L-1 copper were significantly higher than those in the control group after 96 h of exposure (P < 0.05). The results of this study highlight the potential effects of copper as a common stressor in E. sinensis larvae. La concentración letal media (LC50) y su correspondiente intervalo de confianza del 95% fueron estimados en los estadios larvales zoea I y megalopa. La toxicidad aguda del cobre disuelto fue mayor sobre larvas de zoea I (0.16 mg L–1) que sobre las de megalopa (0.21 mg L–1). El estado antioxidante, la expresión del mRNA de la metalotionina–I y la respuesta fisiológica del cangrejo a la toxicidad del cobre fueron posteriormente investigados exponiendo las larvas de megalopa a dosis de 0, 0.08 y 0.16 mg Cu L–1 durante 96 h. La actividad de la superóxido dismutasa (SOD), de la catalasa (CAT), de la glutatión-S-transferasa (GST) y la peroxidación lipídica de las larvas de megalopa se incrementó en relación con el tiempo de exposición y la concentración de cobre. Los niveles de expresión de MT-1 mRNA se correlacionaron positivamente con la concentración de cobre y el tiempo de exposición. El consumo de oxígeno y la tasa respiratoria de las larvas de megalopa fueron significativamente más elevadas que las del grupo control (P < 0.05) en respuesta a dosis de 0.16 mg Cu L–1, durante 96 h de exposición. Los resultados presentados en este estudio ponen de manifiesto los efectos potenciales del cobre como factor de estrés sobre las larvas de E. sinensis. MT-I y GST parecen ser adecuados biomarcadores de estrés a la exposición de Cu ambiental en larvas de E. sinensis

    Effects of Antibacterial Peptide Extracted from Bacillus subtilis fmbJ on the Growth, Physiological Response and Disease Resistance of Megalobrama amblycephala

    The effects of an antibacterial peptide obtained from Bacillus subtilis fmbJ on growth, serum lysozyme complements 3 and 4, total protein content, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total antioxidative capacity, superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, and disease resistance of Wuchang bream (Megalobrama amblycephala) were examined. Fish were randomly divided into five groups: a control group which was fed a basic diet, and four groups fed the basic diet supplemented with 0.1%, 0.2%, 0.4%, or 0.8% antibacterial peptide. At eight weeks, M. amblycephala fed the diet containing 0.2% antibacterial peptide had higher serum lysozyme activity, complement 3 and 4 contents, and SOD activity than the control fish, but lower serum MDA content and AST activity. Fish fed the 0.4% diet had higher weight gain rate, serum lysozyme activity, complement 4 content, total antioxidative capacity, and total protein than the control, and lower serum ALT activity. Feed conversion ratios of fish fed the 0.2% or 0.4% diets were lower than those of control fish. Artificial infection with Aeromonas hydrophila resulted in 93% cumulative mortality in the control group, and 61-84% in the groups fed the 0.2% or 0.4% diets. The present study suggests that feed supplementation with 0.2-0.4% antibacterial peptides can stimulate immunity, increase resistance to pathogenic infection, and promote growth in M. amblycephala

    Effects of Dietary Protein Level on Growth Performance, Muscle Composition, Blood Composition, and Digestive Enzyme Activity of Wuchang Bream (Megalobrama amblycephala) Fry

    The purpose of this study was to determine the dietary protein requirement and effects of dietary protein level on growth performance, muscle composition, blood composition, and digestive enzyme activity in Wuchang bream fry. Five isoenergetic and isolipidic semi-purified diets were formulated to contain 28%, 30%, 32%, 34%, or 36% (dry matter) dietary protein. Diets were fed to triplicate groups of 25 fishes (16.08±0.03 g) to near satiation three times a day in a closed recirculation system for 10 weeks. Weight gain, specific growth rate, and feed conversion ratio significantly improved as the dietary protein content increased up to 34%. The protein efficiency ratio, hepatosomatic index, and viscerosomatic index significantly dropped as the dietary protein rose while the Fulton condition factor was positively correlated to the dietary protein level. Increased dietary protein resulted in increased muscle protein content and decreased lipid content. Red blood cell, hemoglobin, and hematocrit counts increased significantly with the increase in dietary protein. Serum triiodothyronine and thyroxine significantly rose as the dietary protein rose but serum aspartate aminotransferase significantly dropped. Intestinal protease and amylase activity rose significantly with the increase in dietary protein while lipase tended to drop.     Prevalence, associated factors and outcomes of pressure injuries in adult intensive care unit patients: the DecubICUs study

Epidemiologic data is however unavailable. We aimed to provide an international picture of the extent of pressure injuries and factors associated with ICU-acquired pressure injuries in adult ICU patients. Methods: International 1-day point-prevalence study; follow-up for outcome assessment until hospital discharge (maximum 12 weeks). Factors associated with ICU-acquired pressure injury and hospital mortality were assessed by generalised linear mixed-effects regression analysis. Results: Data from 13,254 patients in 1117 ICUs (90 countries) revealed 6747 pressure injuries; 3997 (59.2%) were ICU-acquired. Overall prevalence was 26.6% (95% confidence interval [CI] 25.9–27.3). ICU-acquired prevalence was 16.2% (95% CI 15.6–16.8). Sacrum (37%) and heels (19.5%) were most affected. Factors independently associated with ICU-acquired pressure injuries were older age, male sex, being underweight, emergency surgery, higher Simplified Acute Physiology Score II, Braden score 3 days, comorbidities (chronic obstructive pulmonary disease, immunodeficiency), organ support (renal replacement, mechanical ventilation on ICU admission), and being in a low or lower-middle income-economy. Gradually increasing associations with mortality were identified for increasing severity of pressure injury: stage I (odds ratio [OR] 1.5; 95% CI 1.2–1.8), stage II (OR 1.6; 95% CI 1.4–1.9), and stage III or worse (OR 2.8; 95% CI 2.3–3.3). Conclusion: Pressure injuries are common in adult ICU patients. ICU-acquired pressure injuries are associated with mainly intrinsic factors and mortality. Optimal care standards, increased awareness, appropriate resource allocation, and further research into optimal prevention are pivotal to tackle this important patient safety threat
