    Study of CP violation in Ξ›c+\Lambda_c^+ decay

    In this paper, we study CP violation in Ξ›c+β†’BP\Lambda_c^+\to BP and Ξ›c+β†’BV\Lambda_c^+\to BV decays, where B,PB,P and VV denote a light spin-1/2 baryon, pseudoscalar and a vector meson respectively. In these processes the TT odd CP violating triple-product (TP) correlations are examined. The genuine CP violating observables which are composed of the helicity amplitudes occurring in the angular distribution are constructed. Experimentally, by performing a full angular analysis it is shown how one may extract the helicity amplitudes and then obtain the TP asymmetries. We estimate the TP asymmetries in \lam decays to be negligible in the standard model making these processes an excellent place to look for new physics. Taking a two Higgs doublet model, as an example of new physics, we show that large TP asymmetries are possible in these decays. Finally, we discuss how BES-III and Super Ο„\tau-Charm experiments will be sensitive to these CP violating signals in Ξ›c+\Lambda_c^+ decays.Comment: 7 pages, 2 tables. Journal reference is adde

    Effect of cytochalasin types on the production of heterozygous parthenogenetic porcine embryos and the isolation of putative parthenogenetic embryonic stem cells

    Parthenogenetic embryos have been suggested as an alternative source of embryonic stem cells (ESCs). The present study was undertaken to determine the efficiency with which porcine heterozygous parthenotes could be produced using cytochalasin B (CB) or cytochalasin D (CD) and whether parthenogenetic ESCs (pESCs) could be isolated from these. Cleavage rate was lower and fewer embryos developed to the blastocyst stage in the CB group compared with the CD group. The number of primary outgrowths obtained was also lower in the CB compared with the CD group. No primary lines were isolated from embryonal outgrowths in the CB group. In contrast, primary cell lines were derived from these in the CD group. These lines survived vitrification and warming, resulting in established cell lines, which maintained a characteristic ESC morphology and expressed the pluripotent markers Oct4 and Nanog following repeated passaging.     Functional Synchronization of Biological Rhythms in a Tritrophic System

    In a tritrophic system formed by a plant, an herbivore and a natural enemy, each component has its own biological rhythm. However, the rhythm correlations among the three levels and the underlying mechanisms in any tritrophic system are largely unknown. Here, we report that the rhythms exhibited bidirectional correlations in a model tritrophic system involving a lima bean, a pea leafminer and a parasitoid. From the bottom-up perspective, the rhythm was initiated from herbivore feeding, which triggered the rhythms of volatile emissions; then the rhythmic pattern of parasitoid activities was affected, and these rhythms were synchronized by a light switch signal. Increased volatile concentration can enhance the intensity of parasitoid locomotion and oviposition only under light. From the top-down perspective, naive and oviposition-experienced parasitoids were able to utilize the different volatile rhythm information from the damaged plant to locate host leafminers respectively. Our results indicated that the three interacting organisms in this system can achieve rhythmic functional synchronization under a natural light-dark photoperiod, but not under constant light or darkness. These findings provide new insight into the rhythm synchronization of three key players that contribute to the utilization of light and chemical signals, and our results may be used as potential approaches for manipulating natural enemies

    Peginterferon Alfa-2a, Lamivudine, and the Combination for HBeAg-Positive Chronic Hepatitis B

    Background: Current treatments for chronic hepatitis B are suboptimal. In the search for improved therapies, we compared the efficacy and safety of pegylated interferon alfa plus lamivudine, pegylated interferon alfa without lamivudine, and lamivudine alone for the treatment of hepatitis B e antigen (HBeAg)–positive chronic hepatitis B. Methods: A total of 814 patients with HBeAg-positive chronic hepatitis B received either peginterferon alfa-2a (180 Β΅g once weekly) plus oral placebo, peginterferon alfa-2a plus lamivudine (100 mg daily), or lamivudine alone. The majority of patients in the study were Asian (87 percent). Most patients were infected with hepatitis B virus (HBV) genotype B or C. Patients were treated for 48 weeks and followed for an additional 24 weeks. Results: After 24 weeks of follow-up, significantly more patients who received peginterferon alfa-2a monotherapy or peginterferon alfa-2a plus lamivudine than those who received lamivudine monotherapy had HBeAg seroconversion (32 percent vs. 19 percent [

    Geldanamycin Derivative Ameliorates High Fat Diet-Induced Renal Failure in Diabetes

    Diabetic nephropathy is a serious complication of longstanding diabetes and its pathogenesis remains unclear. Oxidative stress may play a critical role in the pathogenesis and progression of diabetic nephropathy. Our previous studies have demonstrated that polyunsaturated fatty acids (PUFA) induce peroxynitrite generation in primary human kidney mesangial cells and heat shock protein 90Ξ²1 (hsp90Ξ²1) is indispensable for the PUFA action. Here we investigated the effects of high fat diet (HFD) on kidney function and structure of db/db mice, a widely used rodent model of type 2 diabetes. Our results indicated that HFD dramatically increased the 24 h-urine output and worsened albuminuria in db/db mice. Discontinuation of HFD reversed the exacerbated albuminuria but not the increased urine output. Prolonged HFD feeding resulted in early death of db/db mice, which was associated with oliguria and anuria. Treatment with the geldanamycin derivative, 17-(dimethylaminoehtylamino)-17-demethoxygeldanamycin (17-DMAG), an hsp90 inhibitor, preserved kidney function, and ameliorated glomerular and tubular damage by HFD. 17-DMAG also significantly extended survival of the animals and protected them from the high mortality associated with renal failure. The benefit effect of 17-DMAG on renal function and structure was associated with a decreased level of kidney nitrotyrosine and a diminished kidney mitochondrial Ca2+ efflux in HFD-fed db/db mice. These results suggest that hsp90Ξ²1 is a potential target for the treatment of nephropathy and renal failure in diabetes

    Regulatory Effect of Connexin 43 on Basal Ca2+ Signaling in Rat Ventricular Myocytes

    Background: It has been found that gap junction-associated intracellular Ca 2+ [Ca 2+]i disturbance contributes to the arrhythmogenesis and hyperconstriction in diseased heart. However, whether functional gaps are also involved in the regulation of normal Ca 2+ signaling, in particular the basal [Ca 2+] i activities, is unclear. Methods and Results: Global and local Ca 2+ signaling and gap permeability were monitored in cultured neonatal rat ventricular myocytes (NRVMs) and freshly isolated mouse ventricular myocytes by Fluo4/AM and Lucifer yellow (LY), respectively. The results showed that inhibition of gap communication by heptanol, Gap 27 and flufenamic acid or interference of connexin 43 (Cx43) with siRNA led to a significant suppression of LY uptake and, importantly, attenuations of global Ca 2+ transients and local Ca 2+ sparks in monolayer NRVMs and Ca 2+ sparks in adult ventricular myocytes. In contrast, overexpression of rat-Cx43 in NRVMs induced enhancements in the above measurements, and so did in HEK293 cells expressing rat Cx43. Additionally, membrane-permeable inositol 1,4,5-trisphosphate (IP3 butyryloxymethyl ester) and phenylephrine, an agonist of adrenergic receptor, could relieve the inhibited Ca 2+ signal and LY uptake by gap uncouplers, whereas blockade of IP 3 receptor with xestospongin C or 2-aminoethoxydiphenylborate mimicked the effects of gap inhibitors.
