    EFFECT OF COMPOST ON THE MICROCLOVER ESTABLISHMENT AND USE OF COMPOST AND MICROCLOVER TO REDUCE LAWN NUTRIENT RUNOFF

    High volume compost incorporation can reduce runoff from compacted soils but its use also associated with elevated N and P concentrations in runoff making it difficult to assess if this practice will reduce nutrient loading of surface waters. Additionally, little is known about how this practice will effect leguminous species establishment in lawns as means to reduce long term fertilizer use. When 5 cm of compost was incorporated into soil a reduction in runoff of 40 and 59% was needed for N and P losses from a tall fescue + microclover lawn to be equivalent to a non-compost amended soil supporting a well fertilized tall fescue lawn. Use of compost as a soil amendment resulted in quicker lawn establishment and darker color, when compared to non-amended soil receiving a mineral fertilizer.     The Reading Palaeofire Database : an expanded global resource to document changes in fire regimes from sedimentary charcoal records

    Sedimentary charcoal records are widely used to reconstruct regional changes in fire regimes through time in the geological past. Existing global compilations are not geographically comprehensive and do not provide consistent metadata for all sites. Furthermore, the age models provided for these records are not harmonised and many are based on older calibrations of the radiocarbon ages. These issues limit the use of existing compilations for research into past fire regimes. Here, we present an expanded database of charcoal records, accompanied by new age models based on recalibration of radiocarbon ages using IntCal20 and Bayesian age-modelling software. We document the structure and contents of the database, the construction of the age models, and the quality control measures applied. We also record the expansion of geographical coverage relative to previous charcoal compilations and the expansion of metadata that can be used to inform analyses. This first version of the Reading Palaeofire Database contains 1676 records (entities) from 1480 sites worldwide.     TCS2 Increases Olaquindox-Induced Apoptosis by Upregulation of ROS Production and Downregulation of Autophagy in HEK293 Cells

    Olaquindox, a feed additive, has drawn public attention due to its potential mutagenicity, genotoxicity, hepatoxicity and nephrotoxicity. The purpose of this study was to investigate the role of tuberous sclerosis complex (TSC2) pathways in olaquindox-induced autophagy in human embryonic kidney 293 (HEK293) cells. The results revealed that olaquindox treatment reduced the cell viability of HEK293 cells and downregulated the expression of TSC2 in a dose- and time-dependent manner. Meanwhile, olaquindox treatment markedly induced the production of reactive oxygen species (ROS), cascaded to autophagy, oxidative stress, and apoptotic cell death, which was effectively eliminated by the antioxidant N-acetylcysteine (NAC). Furthermore, overexpression of TSC2 attenuated olaquindox-induced autophagy in contrast to inducing the production of ROS, oxidative stress and apoptosis. Consistently, knockdown of TSC2 upregulated autophagy, and decreased olaquindox-induced cell apoptosis. In conclusion, our findings indicate that TSC2 partly participates in olaquindox-induced autophagy, oxidative stress and apoptosis, and demonstrate that TSC2 has a negative regulation role in olaquindox-induced autophagy in HEK293 cells

    Fabrication of Microglass Nozzle for Microdroplet Jetting

    An ejection aperture nozzle is the essential part for all microdrop generation techniques. The diameter size, the flow channel geometry, and fluid impedance are the key factors affecting the ejection capacity. A novel low-cost fabrication method of microglass nozzle involving four steps is developed in this work. In the first heating step, the glass pipette is melted and pulled. Then, the second heating step is to determine the tip cone angle and modify the flow channel geometry. The desired included angle is usually of 30~45 degrees. Fine grind can determine the exact diameter of the hole. Postheating step is the final process and it can reduce the sharpness of the edges of the hole. Micronozzles with hole diameters varying from 30 to 100 µm are fabricated by the homemade inexpensive and easy-to-operate setup. Hydrophobic treating method of microglass nozzle to ensure stable and accurate injection is also introduced in this work. According to the jetting results of aqueous solution, UV curing adhesive, and solder, the fabricated microglass nozzle can satisfy the need of microdroplet jetting of multimaterials

    GADD45a Regulates Olaquindox-Induced DNA Damage and S-Phase Arrest in Human Hepatoma G2 Cells via JNK/p38 Pathways

    Olaquindox, a quinoxaline 1,4-dioxide derivative, is widely used as a feed additive in many countries. The potential genotoxicity of olaquindox, hence, is of concern. However, the proper mechanism of toxicity was unclear. The aim of the present study was to investigate the effect of growth arrest and DNA damage 45 alpha (GADD45a) on olaquindox-induced DNA damage and cell cycle arrest in HepG2 cells. The results showed that olaquindox could induce reactive oxygen species (ROS)-mediated DNA damage and S-phase arrest, where increases of GADD45a, cyclin A, Cdk 2, p21 and p53 protein expression, decrease of cyclin D1 and the activation of phosphorylation-c-Jun N-terminal kinases (p-JNK), phosphorylation-p38 (p-p38) and phosphorylation-extracellular signal-regulated kinases (p-ERK) were involved. However, GADD45a knockdown cells treated with olaquindox could significantly decrease cell viability, exacerbate DNA damage and increase S-phase arrest, associated with the marked activation of p-JNK, p-p38, but not p-ERK. Furthermore, SP600125 and SB203580 aggravated olaquindox-induced DNA damage and S-phase arrest, suppressed the expression of GADD45a. Taken together, these findings revealed that GADD45a played a protective role in olaquindox treatment and JNK/p38 pathways may partly contribute to GADD45a regulated olaquindox-induced DNA damage and S-phase arrest. Our findings increase the understanding on the molecular mechanisms of olaquindox

    Postglacial fire history and interactions with vegetation and climate in southwestern Yunnan Province of China

    A high-resolution, continuous 18.5 kyr (1 kyr D1000 cal yr BP) macroscopic charcoal record from Qinghai Lake in southwestern Yunnan Province, China, reveals postglacial fire frequency and variability history. The results show that three periods with high-frequency and high-severity fires occurred during the periods 18.5-15.0, 13.0-11.5, and 4.3-0.8 ka, respectively. This record was compared with major pollen taxa and pollen diversity indices from the same core, and tentatively related to the regional climate proxy records with the aim to separate climate-from human-induced fire activity, and discuss vegetation-fire-climate interactions. The results suggest that fire was mainly controlled by climate before 4.3 ka and by the combined actions of climate and humans after 4.3 ka. Before 4.3 ka, high fire activity corresponded to cold and dry climatic conditions, while warm and humid climatic conditions brought infrequent and weak fires. Fire was an important disturbance factor and played an important role in forest dynamics around the study area. Vegetation responses to fire after 4.3 ka are not consistent with those before 4.3 ka, suggesting that human influence on vegetation and fire regimes may have become more prevalent after 4.3 ka. The comparisons between fire activity and vegetation reveal that evergreen oaks are flammable plants and fire-tolerant taxa. Alnus is a fire-adapted taxon and a nonflammable plant, but density of Alnus forest is a key factor to decide its fire resistance. The forests dominated by Lithocarpus/Castanopsis and/or tropical trees and shrubs are not easy to ignite, but Lithocarpus/Castanopsis and tropical trees and shrubs are fire-sensitive taxa.     New evidence on deglacial climatic variability from an alpine lacustrine record in northwestern Yunnan Province, southwestern China

    New deglacial pollen and conifer stoma records from Tiancai Lake, northwestern Yunnan Province, southwestern China, an alpine lake in the southwest monsoon region, are presented in this study. Based on these records, the lithology of core TCYL1, and PCA analysis of pollen data between ~21 and 11.5kaBP (calibrated 14C years), the deglacial vegetation and climate changes are discussed in detail. The results show that Tiancai Lake was above the upper limit of Picea/Abies forest (a treeline in the study area) between ~21 and 11.5kaBP, and the climate was colder and drier than that of the Holocene. During this period eight significant vegetation changes are recorded that are considered to be responses to changing temperatures and variations in the southwest monsoon in southwestern China. The Heinrich Event 1 (H1), the Bølling/Allerød warm period (BA) and the Younger Dryas cold event (YD) are all clearly detected in this record. In addition, this study finds that the initial late glacial warming in northwestern Yunnan Province was at ~18.7kaBP, which is coincident with the climate records in monsoonal Central Asia, the Indian Ocean, the tropical and subtropical Pacific Ocean, and Antarctica, and is a response to solar insolation changes. A noted temperature increase between 15.8 and 14.4kaBP occurred at the end of the H1 and before the BA, which indicates a strong pre-Bølling warming. Based on the study, we consider that the hypothesis about a slowdown of the ocean's thermohaline circulation is sufficient to explain these late glacial abrupt events
