    EVALUATION OF ANALGESIC, ANTI-INFLAMMATORY AND ANTIPYRETIC ACTIVITIES OF THE ETHANOL EXTRACT FROM SPERANSKIA TUBERCULATA

    Background: Speranskia tuberculata (Bunge) Baill, has been used to prevent and treat many diseases in Chinese folk medicine, nevertheless, few investigations had been reported. Materials and Methods: Animals were orally administered STE at the doses of 125, 250, 500 mg/kg. The analgesic effect was estimated in mice by hot-plate test and the acetic acid-induced writhing test. The anti-inflammatory effect was assessed using rat paw edema model elicited by fresh egg white and the mouse ear edema model caused by dimethylbenzene. The antipyretic effect was determined using the lipopolysaccharide (LPS)-induced rat fever model. In addition, the acute oral toxicity of STE was studied. Results: STE significantly and dose-dependently reduced the number of writhing responses in mice, prolonged reaction time of mice against heat stimulation, depressed egg white-induced paw edema in rats and the dimethylbenzene-caused ear edema in mice, but did not alleviate LPS-induced pyrexia in rats. No death of mice was observed when orally administered STE up to 52.8 g/kg (approximately 2080 times of clinical dose used). Conclusion: STE possesses evident analgesic and anti-inflammatory activities, but has no antipyretic effect. Furthermore STE has a favorable safety. These findings support the applications of Speranskia tuberculata as an analgesic and anti-inflammatory drug in folk medicine

    Spatial transmission and meteorological determinants of tuberculosis incidence in Qinghai Province, China: a spatial clustering panel analysis

    BACKGROUND: Tuberculosis (TB) is the notifiable infectious disease with the second highest incidence in the Qinghai province, a province with poor primary health care infrastructure. Understanding the spatial distribution of TB and related environmental factors is necessary for developing effective strategies to control and further eliminate TB. METHODS: Our TB incidence data and meteorological data were extracted from the China Information System of Disease Control and Prevention and statistical yearbooks, respectively. We calculated the global and local Moran's I by using spatial autocorrelation analysis to detect the spatial clustering of TB incidence each year. A spatial panel data model was applied to examine the associations of meteorological factors with TB incidence after adjustment of spatial individual effects and spatial autocorrelation. RESULTS: The Local Moran's I method detected 11 counties with a significantly high-high spatial clustering (average annual incidence: 294/100 000) and 17 counties with a significantly low-low spatial clustering (average annual incidence: 68/100 000) of TB annual incidence within the examined five-year period; the global Moran's I values ranged from 0.40 to 0.58 (all P-values < 0.05). The TB incidence was positively associated with the temperature, precipitation, and wind speed (all P-values < 0.05), which were confirmed by the spatial panel data model. Each 10 °C, 2 cm, and 1 m/s increase in temperature, precipitation, and wind speed associated with 9 % and 3 % decrements and a 7 % increment in the TB incidence, respectively. CONCLUSIONS: High TB incidence areas were mainly concentrated in south-western Qinghai, while low TB incidence areas clustered in eastern and north-western Qinghai. Areas with low temperature and precipitation and with strong wind speeds tended to have higher TB incidences

    Molecular Mechanisms of Hepatocellular Carcinoma Related to Aflatoxins: An Update

    Hepatocellular carcinoma (hepatocarcinoma) is a major type of primary liver cancer and one of the most frequent human malignant neoplasms. Aflatoxins are I-type chemical carcinogen for hepatocarcinoma. Increasing evidence has shown that hepatocarcinoma induced by aflatoxins is the result of interaction between aflatoxins and hereditary factor. Aflatoxins can induce DNA damage including DNA strand break, adducts formation, oxidative DNA damage, and gene mutation and determine which susceptible individuals feature cancer. Inheritance such as alterations may result in the activation of proto-oncogenes and the inactivation of tumor suppressor genes and determine individual susceptibility to cancer. Interaction between aflatoxins and genetic susceptible factors commonly involve in almost all pathologic sequence of hepatocarcinoma: chronic liver injury, cirrhosis, atypical hyperplastic nodules, and hepatocarcinoma of early stages. In this review, we discuss the biogenesis, toxification, and epidemiology of aflatoxins and signal pathways of aflatoxin-induced hepatocarcinoma. We also discuss the roles of some important genes related to cell apoptosis, DNA repair, drug metabolism, and tumor metastasis in hepatocarcinogenesis related to aflatoxins

    The conscious processing of emotion in depression disorder: a meta-analysis of neuroimaging studies

    BackgroundDepression is generally accompanied by a disturbed conscious processing of emotion, which manifests as a negative bias to facial/voice emotion information and a decreased accuracy in emotion recognition tasks. Several studies have proved that abnormal brain activation was responsible for the deficit function of conscious emotion recognition in depression. However, the altered brain activation related to the conscious processing of emotion in depression was incongruent among studies. Therefore, we conducted an activation likelihood estimation (ALE) analysis to better understand the underlying neurophysiological mechanism of conscious processing of emotion in depression.MethodElectronic databases were searched using the search terms “depression,” “emotion recognition,” and “neuroimaging” from inceptions to April 10th, 2023. We retrieved trials which explored the neuro-responses of depressive patients to explicit emotion recognition tasks. Two investigators independently performed literature selection, data extraction, and risk of bias assessment. The spatial consistency of brain activation in conscious facial expressions recognition was calculated using ALE. The robustness of the results was examined by Jackknife sensitivity analysis.ResultsWe retrieved 11,365 articles in total, 28 of which were included. In the overall analysis, we found increased activity in the middle temporal gyrus, superior temporal gyrus, parahippocampal gyrus, and cuneus, and decreased activity in the superior temporal gyrus, inferior parietal lobule, insula, and superior frontal gyrus. In response to positive stimuli, depressive patients showed hyperactivity in the medial frontal gyrus, middle temporal gyrus, and insula (uncorrected p &lt; 0.001). When receiving negative stimuli, a higher activation was found in the precentral gyrus, middle frontal gyrus, precuneus, and superior temporal gyrus (uncorrected p &lt; 0.001).ConclusionAmong depressive patients, a broad spectrum of brain areas was involved in a deficit of conscious emotion processing. The activation of brain regions was different in response to positive or negative stimuli.     In situ Raman spectroscopic evidence for oxygen reduction reaction intermediates at platinum single crystal surfaces

    铂基催化剂表面的氧还原反应(ORR)是燃料电池重最重要的阴极反应。虽然经过几十年的研究,ORR的具体反应路径并没有真正解析清楚。在本研究中,李剑锋教授课题组首次利用其发展的电化学壳层隔绝纳米粒子增强拉曼光谱(SHINERS)技术原位研究了Pt(hkl)单晶表面的ORR反应过程,成功捕获到在1150 cm-1、1080 cm-1和732 cm-1区域O2-、OH*和HO2*等重要反应中间物种的直接拉曼光谱证据,并通过相应的同位素取代实验(氘和O-18同位素实验)及DFT理论模拟确认了中间物种的谱峰归属和吸附构型。总的来说,该项研究首次在Pt(hkl)单晶表面原位获得ORR反应重要中间物种的直接拉曼光谱证据,提出合理的ORR反应路径,加深了人们对ORR反应机理的认识,也为其他界面催化反应机理的研究提供了一条可行的研究思路。 该研究工作由校内外多个课题组共同努力完成,化学化工学院李剑锋教授课题组负责相关电化学原位拉曼光谱实验,Alicante大学Juan Feliu教授课题组负责单晶电化学实验,化学化工学院吴德印教授课题组负责DFT理论模拟,物理系杨志林教授课题组负责3D-FDTD的理论模拟。该研究工作的第一作者董金超和第二作者张霞光分别为化学化工学院2014级博士生(已毕业)和2015级博士生。【Abstract】Developing an understanding of structure-activity relationships and reaction mechanisms of catalytic processes is critical to the successful design of highly efficient catalysts. As a fundamental reaction in fuel cells, elucidation of the oxygen reduction reaction (ORR) mechanism at Pt(hkl) surfaces has remained a significant challenge for researchers. Here, we employ in situ electrochemical surface-enhanced Raman spectroscopy (SERS) and density functional theory (DFT) calculation techniques to examine the ORR process at Pt(hkl) surfaces. Direct spectroscopic evidences for ORR intermediates indicates that under acid conditions, the pathway of ORR at Pt(111) occurs through the formation of HO2, while at Pt(110) and Pt(100) it occurs via the generation of OH*. However, we propose that the pathway of ORR under alkaline conditions at Pt(hkl) surfaces mainly occurs through the formation of O2- .     Two New Cytotoxic Indole Alkaloids from a Deep-Sea Sediment Derived Metagenomic Clone

    An atlas of DNA methylomes in porcine adipose and muscle tissues

    It is evident that epigenetic factors, especially DNA methylation, have essential roles in obesity development. Here, using pig as a model, we investigate the systematic association between DNA methylation and obesity. We sample eight variant adipose and two distinct skeletal muscle tissues from three pig breeds living within comparable environments but displaying distinct fat level. We generate 1,381 Gb of sequence data from 180 methylated DNA immunoprecipitation libraries, and provide a genome-wide DNA methylation map as well as a gene expression map for adipose and muscle studies. The analysis shows global similarity and difference among breeds, sexes and anatomic locations, and identifies the differentially methylated regions. The differentially methylated regions in promoters are highly associated with obesity development via expression repression of both known obesity-related genes and novel genes. This comprehensive map provides a solid basis for exploring epigenetic mechanisms of adipose deposition and muscle growth
