    Ribosomes synthesizing nascent secretory proteins are targeted to the membrane by the signal recognition particle (SRP), a small ribonucleoprotein that binds to the signal peptide as it emerges from the ribosome. SRP arrests further elongation, causing ribosomes to stack behind the arrested ribosome. Upon interaction of SRP with its receptor on the ER membrane, the translation arrest is released and the ribosome becomes bound to the ER membrane. We have examined the distribution of unattached and membrane-bound ribosomes during the translation of mRNAs encoding two secretory proteins, bovine preprolactin and rat preproinsulin I. We find that the enhancement of ribosome stacking that occurs when SRP arrests translation of these proteins is relaxed in the presence of microsomal membranes. We also demonstrate that two previously described populations of membrane-associated ribosomes, distinguished by their sensitivity to high salt or EDTA extraction, correspond to ribosomes that have synthesized differing lengths of the nascent polypeptide. This analysis has revealed that nascent chain insertion into the membrane begins at distinct points for different presecretory proteins

    Trauma Informed Community Building Evaluation: A Formative Evaluation of the TICB Model and its Implementation in Potrero Hill

    This formative evaluation of the TICB model and its implementation in the PTA public housing community was conducted between September 2014 and July 2015 by an evaluation team from the HOPE SF Learning Center. This evaluation was designed to support the further development of the TICB model as well as inform efforts to implement the model in Potrero Terrace and Annex and other communities. This evaluation seeks to examine the implementation and impact of the TICB model at PTA in order to: * Understand the impact of ongoing TICB-informed programming through analysis of outputs and outcomes prioritized by stakeholder partners [see Appendix A]. * Identify facilitators and barriers to implementation of the TICB model in community-building work within PTA and the surrounding Potrero Hill neighborhood. * Inform BRIDGE Housing's work to improve programming, and guide future program priorities and structures. * Generate information to better understand the impact of the financial investment in helping to build community with and between public housing residents and residents of the surrounding neighborhood. * Assess implications for replicability/reproducibility at other public housing communities, including the additional HOPE SF sites, and beyond

    Correlates of quality of life in overweight or obese breast cancer survivors at enrollment into a weight loss trial

    OBJECTIVE: To examine the correlates of the physical and psychosocial domains of quality of life (QOL) in a cohort of breast cancer survivors participating in a weight loss intervention trial. Available data included information on weight and physical activity, as well as demographic and medical characteristics. METHODS: Correlates of QOL and psychosocial functioning were examined in 692 overweight/obese breast cancer survivors at entry into a weight loss trial. QOL was explored with three measures: Short-form 36 (SF-36); Impact of Cancer Scale (IOCv2); and the Breast Cancer Prevention Trial (BCPT) Symptom Scales. Bivariate and multivariate analyses were used to identify correlates and associations adjusted for other characteristics. RESULTS: In multivariate analysis, younger age was associated with higher negative impact scores (p<0.01). Hispanic, African-American and Asian women had higher IOC positive impact scores compared to white non-Hispanic women (p<0.01). Higher education was associated with lower scores on mental QOL and the IOC positive impact scale (p<0.01). BMI was not independently associated with QOL measures. Physical activity was directly associated with physical and mental QOL and IOC positive impact, and inversely related to IOC negative impact and BCPT symptom scales. CONCLUSIONS: QOL measures in breast cancer survivors are differentially associated with demographic and other characteristics. When adjusted for these characteristics, degree of adiposity among overweight/obese women does not appear to be independently associated with QOL. Among overweight/obese breast cancer survivors, higher level of physical activity is associated with higher QOL across various scales and dimensions

    Encountering violence: terrorism and horrorism in war and citizenship

    This article introduces Adriana Cavarero's concept of “horrorism” into International Relations (IR) discussions of the relationship between war and citizenship. Horrorism refers to a violent violation of vulnerable humans who are defined by their simultaneous openness to the other's care and harm. With its motif of physical and ontological denigration, horrorism offends the human condition by making its victims gaze upon and/or experience repugnant violence and bodily disfiguration precisely when the vulnerable are most in need of care. The article argues that horrorism complicates disciplinary understandings of contemporary violence which tend to see terrorism, but not horrorism, in war and which generally neglect to theorize how violence—and particularly horrorism—is embedded in, and exchanged, through state/citizen relationships. To elaborate these arguments, the article analyses three pieces of war art: Jeremy Deller's “Baghdad, 5 March 2007,” Donald Gray's mural, “Operation Iraqi Freedom,” and a still image from Cynthia Weber's film, “Guadalupe Denogean: ‘I am an American.’” By taking the War on Terrorism as their subject, these pieces demonstrate how war makes visible the terror and horror in state/citizen relationships. The article concludes by reconsidering how encountering signs of horrorism might broaden our frames of war and further our empathic vision toward the precarious victims of horrorism or, alternatively, might confirm the patriotic allegiances of imperial citizens in ways that further bind their citizenship to state political and economic violence and narrow the scope for genuine empathy

    Human Influence Past and Present – Relationship of Nutrient and Hydrologic Conditions to Urban Wetland Macrophyte Distribution

They provide habitat for wildlife, green space, and recreational opportunities for nearby human populations. We investigated an isolated section of an urban wetland in the Cleveland metropolitan area to determine the overall quality of the vegetation and to evaluate the site for possible rehabilitation. We also researched the distribution of plant species in relation to existing hydrologic, soil, and nutrient conditions in order to identify possible impacts of historic or present human activities in the surrounding watershed. Vegetation composition and physical/chemical parameters were measured in 1.0 × 1.0 m2 plots along three transects. Canonical correspondence analysis (CCA) was used to directly correlate species distributions to nutrient concentrations, soil carbon content, and water depth. Our sample area was dominated by Typha angustifolia, Leersia oryzoides, and Sparganium eurycarpum. A few high quality species were present, but the overall macrophyte community was indicative of human disturbance. Historic information revealed a long history of disturbance at the site and continuing anthropogenic impact. Patchiness in nutrient and water depth gradients results from historic and current human impacts in the study area. Our results indicate any rehabilitation efforts of the site need to take into account past and current anthropogenic stressors. We recommend aggressive removal of invasive species and re-introduction of nutrient-tolerant native taxa to achieve successful rehabilitation at the site
