    A Search for Gamma-ray Burst Subgroups in the SWIFT and RHESSI Databases

    A sample of 286 gamma-ray bursts (GRBs) detected by the Swift satellite and 358 GRBs detected by the RHESSI satellite are studied statistically. Previously published articles, based on the BATSE GRB Catalog, claimed the existence of an intermediate subgroup of GRBs with respect to duration. We use the statistical chi^2 test and the F-test to compare the number of GRB subgroups in our databases with the earlier BATSE results. Similarly to the BATSE database, the short and long subgroups are well detected in the Swift and RHESSI data. However, contrary to the BATSE data, we have not found a statistically significant intermediate subgroup in either Swift or RHESSI data.Comment: Published in Proceedings of the 2008 Nanjing Gamma-ray Burst Conferenc

    Nonsolar astronomy with the Reuven Ramaty High Energy Solar Spectroscopic Imager (RHESSI)

    The Reuven Ramaty High Energy Solar Spectroscopic Imager (RHESSI) is a NASA Small Explorer satellite designed to study hard x-ray and gamma-ray emission from solar flares. In addition, its high-resolution array of germanium detectors can see photons from high-energy sources throughout the Universe. Here we discuss the various algorithms necessary to extract spectra, lightcurves, and other information about cosmic gamma-ray bursts, pulsars, and other astrophysical phenomena using an unpointed, spinning array of detectors. We show some preliminary results and discuss our plans for future analyses. All RHESSI data are public, and scientists interested in participating should contact the principal author

    RHESSI Spectral Fits of Swift GRBs

    One of the challenges of the Swift era has been accurately determining Epeak for the prompt GRB emission. RHESSI, which is sensitive from 30 keV to 17 MeV, can extend spectral coverage above the Swift-BAT bandpass. Using the public Swift data, we present results of joint spectral fits for 26 bursts co-observed by RHESSI and Swift-BAT through May 2007. We compare these fits to estimates of Epeak which rely on BAT data alone. A Bayesian Epeak estimator gives better correspondence with our measured results than an estimator relying on correlations with the Swift power law indices.Comment: 4 pages, 1 figure.     Observations of the Prompt Gamma-Ray Emission of GRB 070125

    The long, bright gamma-ray burst GRB 070125 was localized by the Interplanetary Network. We present light curves of the prompt gamma-ray emission as observed by Konus-WIND, RHESSI, Suzaku-WAM, and \textit{Swift}-BAT. We detail the results of joint spectral fits with Konus and RHESSI data. The burst shows moderate hard-to-soft evolution in its multi-peaked emission over a period of about one minute. The total burst fluence as observed by Konus is 1.79×10−41.79 \times 10^{-4} erg/cm2^2 (20 keV--10 MeV). Using the spectroscopic redshift z=1.548z=1.548, we find that the burst is consistent with the ``Amati'' Epeak,i−EisoE_{peak,i}-E_{iso} correlation. Assuming a jet opening angle derived from broadband modeling of the burst afterglow, GRB 070125 is a significant outlier to the ``Ghirlanda'' Epeak,i−EγE_{peak,i}-E_\gamma correlation. Its collimation-corrected energy release Eγ=2.5×1052E_\gamma = 2.5 \times 10^{52} ergs is the largest yet observed.Comment: 25 pages, 6 figures; accepted for publication in ApJ.     Acid ceramidase (Ac) is a lysosomal enzyme catalyzing the generation of sphingosine from ceramide, and Ac inhibitors are currently being investigated as potential cancer therapeutics. Yet, the role of the Ac in immune responses, particularly anti-viral immunity, is not fully understood. To investigate the impact of Ac expression on various leukocyte populations, we generated a tamoxifen-inducible global knockout mouse model for the Ac (iAc-KO). Following tamoxifen administration to healthy mice, we extracted primary and secondary lymphoid organs from iAc-KO and wild-type (wt) littermates and subsequently performed extensive flow cytometric marker analysis. In addition, we isolated CD4 + T cells from the spleen and lymph nodes for sphingolipid profiling and restimulated them in vitro with Dynabeadsℱ Mouse T-activator CD3/CD28. Intracellular cytokine expression (FACS staining) was analyzed and secreted cytokines detected in supernatants. To study cell-intrinsic effects, we established an in vitro model for iAc-KO in isolated CD4 + T and B cells. For CD4 + T cells of iAc-KO versus wt mice, we observed reduced Ac activity, an increased ceramide level, and enhanced secretion of IFNγ upon CD3/CD28 costimulation. Moreover, there was a marked reduction in B cell and plasma cell and blast numbers in iAc-KO compared to wt mice. To study cell-intrinsic effects and in line with the 3R principles, we established in vitro cell culture systems for iAc-KO in isolated B and CD4 + T cells. Our findings pinpoint to a key role of the Ac in mature B and antibody-secreting cells and in IFNγ secretion by CD4 + T cells

    Hippocampal biomarkers of fear memory in an animal model of generalized anxiety disorder

    Generalized anxiety disorder (GAD) is highly prevalent and incapacitating. Here we used the Carioca High-Conditioned Freezing (CHF) rats, a previously validated animal model for GAD, to identify biomarkers and structural changes in the hippocampus that could be part of the underlying mechanisms of their high-anxiety profile. Spatial and fear memory was assessed in the Morris water maze and passive avoidance test. Serum corticosterone levels, immunofluorescence for glucocorticoid receptors (GR) in the dentate gyrus (DG), and western blotting for hippocampal brain derived neurotrophic factor (BDNF) were performed. Immunohistochemistry for markers of cell proliferation (bromodeoxiuridine/Ki-67), neuroblasts (doublecortin), and cell survival were undertaken in the DG, along with spine staining (Golgi) and dendritic arborization tracing. Hippocampal GABA release was assessed by neurochemical assay. Fear memory was higher among CHF rats whilst spatial learning was preserved. Serum corticosterone levels were increased, with decreased GR expression. No differences were observed in hippocampal cell proliferation/survival, but the number of newborn neurons was decreased, along with their number and length of tertiary dendrites. Increased expression of proBDNF and dendritic spines was observed; lower ratio of GABA release in the hippocampus was also verified. These findings suggest that generalized anxiety/fear could be associated with different hippocampal biomarkers, such as increased spine density, possibly as a compensatory mechanism for the decreased hippocampal number of neuroblasts and dendritic arborization triggered by high corticosterone. Disruption of GABAergic signaling and BDNF impairment are also proposed as part of the hippocampal mechanisms possibly underlying the anxious phenotype of this model
