    Untersuchungen zur Bedeutung des Lipopolysaccharid-bindenden Proteins (LBP) fĂŒr Mikroorganismen des Magen-Darm-Traktes von BALB/c-LBP+/+ - und BALB/c-LBP-/- (Knock-out)-MĂ€usen

    Durch Forschungsarbeiten der neueren Zeit ist die Bedeutung der Akut-Phase-Proteine als wichtiger Bestandteil der unspezifischen Abwehr im Organismus sichtbar geworden. Die möglichen Wechselwirkungen zwischen der physiologischen Magen-Darm-Flora und diesen Proteinen sind jedoch weitesgehend unbekannt. In dieser Arbeit wurde die Magen-Darm-Flora von einem LBP-/--Knock-out-Maus-Stamm und dem Wildstamm (jeweils 20 Tiere) untersucht. Dabei wurde die aerobe Gesamtkeimzahl, die aerobe und anaerobe Gram-negative Gesamtkeimzahl und die Gesamtkeimzahl der auf MRS-Agar gewachsenen Keime in Darminhaltsproben der Darmabschnitte Jejunum, ZĂ€kum und Kolon von oben genannten MĂ€usen erfasst. ZusĂ€tzlich wurden sieben serologische Untersuchungen bei 40 Tieren durchgefĂŒhrt, um mögliche parallele immunologische Reaktionen des Körpers auf Bestandteile der physiologischen Magen-Darm-Flora zu erfassen und vergleichend zu betrachten. Bei den Untersuchungen konnten hochsignifikante Unterschiede bezĂŒglich der Höhe und Zusammensetzung der aeroben Gesamtkeimzahl, der aeroben Gram-negativen Gesamtkeimzahl, der anaeroben Gram-negativen Gesamtkeimzahl und der Laktobazillen-Gesamtkeimzahl festgestellt werden. Dabei waren bei der LBP-/--Gruppe, den gendeletierten MĂ€usen, die aerobe Gesamtkeimzahl signifikant erhöht, die anderen Keimzahlen, insbesondere die Keimzahlen der aeroben und anaeroben Gram-negativen Bakterien und der Laktobazillen signifikant erniedrigt. ZusĂ€tzlich konnten Abweichungen verschiedener Koloniemerkmale wie HĂ€molyseverhalten, Koloniemorphologie und GenausprĂ€gung zwischen der LBP-/-- und der LBP+/+-Gruppe festgestellt werden. Bei den serologischen Untersuchungen konnten die Ergebnisse der mikrobiologischen Untersuchungen bestĂ€tigt werden. Es wurden signifikante Erhöhungen der relativen Antikörperspiegel gegenĂŒber ausgewĂ€hlten bakteriellen Strukturen, von Mikroorganismen, welche auch signifikant hĂ€ufiger von den jeweiligen Gruppen isoliert werden konnten, festgestellt. Diese Ergebnisse lassen auf einen möglichen Zusammenhang zwischen der Gendeletion, also der Nichtexpression von LBP, und der Zusammensetzung der Magen-Darm-Flora schließen. Inwiefern dies auf direkten oder indirekten Einfluss des LBP auf die Magen-Darm-Flora zurĂŒckzufĂŒhren ist, mĂŒssen weitere Untersuchungen zeigen. Dabei sollten noch andere Einflussfaktoren, wie z. B. die Varianz des LBP in seiner genotypischen AusprĂ€gung, berĂŒcksichtigt werden

    Successful Mobile Application Development: Towards a Taxonomy of Domain-Specific Process Models and Methodologies

    Mobile applications and mobile application development issues receive an increasing attention for practitioners and academics. The development of mobile applications is connected with a number of domain-specific issues and challenges (e.g., fulfilment of customer requirements or the prevention of high development costs). Consequently, the decision of the most effective process model to develop a mobile application plays a crucial role for software and mobile application development teams. With the help of a structured taxonomy-building methodology, we contribute to the extant literature by creating and presenting a taxonomy for process models and methodologies in software engineering and the mobile application development domain. The taxonomy enrich the existing knowledge base and can help mobile application developers to choose the most suitable process model or methodology. Based on our examination, our results indicate new directions for mobile application research and implications for mobile application development

    Impact of c-MYC expression on proliferation, differentiation, and risk of neoplastic transformation of human mesenchymal stromal cells

    Background: Mesenchymal stromal cells isolated from bone marrow (MSC) represent an attractive source of adult stem cells for regenerative medicine. However, thorough research is required into their clinical application safety issues concerning a risk of potential neoplastic degeneration in a process of MSC propagation in cell culture for therapeutic applications. Expansion protocols could preselect MSC with elevated levels of growth-promoting transcription factors with oncogenic potential, such as c-MYC. We addressed the question whether c-MYC expression affects the growth and differentiation potential of human MSC upon extensive passaging in cell culture and assessed a risk of tumorigenic transformation caused by MSC overexpressing c-MYC in vivo. Methods: MSC were subjected to retroviral transduction to induce expression of c-MYC, or GFP, as a control. Cells were expanded, and effects of c-MYC overexpression on osteogenesis, adipogenesis, and chondrogenesis were monitored. Ectopic bone formation properties were tested in SCID mice. A potential risk of tumorigenesis imposed by MSC with c-MYC overexpression was evaluated. Results: C-MYC levels accumulated during ex vivo passaging, and overexpression enabled the transformed MSC to significantly overgrow competing control cells in culture. C-MYC-MSC acquired enhanced biological functions of c-MYC: its increased DNA-binding activity, elevated expression of the c-MYC-binding partner MAX, and induction of antagonists P19ARF/P16INK4A. Overexpression of c-MYC stimulated MSC proliferation and reduced osteogenic, adipogenic, and chondrogenic differentiation. Surprisingly, c-MYC overexpression also caused an increased COL10A1/COL2A1 expression ratio upon chondrogenesis, suggesting a role in hypertrophic degeneration. However, the in vivo ectopic bone formation ability of c-MYC-transduced MSC remained comparable to control GFP-MSC. There was no indication of tumor growth in any tissue after transplantation of c-MYC-MSC in mice. Conclusions: C-MYC expression promoted high proliferation rates of MSC, attenuated but not abrogated their differentiation capacity, and did not immediately lead to tumor formation in the tested in vivo mouse model. However, upregulation of MYC antagonists P19ARF/P16INK4A promoting apoptosis and senescence, as well as an observed shift towards a hypertrophic collagen phenotype and cartilage degeneration, point to lack of safety for clinical application of MSC that were manipulated to overexpress c-MYC for their better expansion

    Activation of Hypoxia Inducible Factor 1 Is a General Phenomenon in Infections with Human Pathogens

    BACKGROUND: Hypoxia inducible factor (HIF)-1 is the key transcriptional factor involved in the adaptation process of cells and organisms to hypoxia. Recent findings suggest that HIF-1 plays also a crucial role in inflammatory and infectious diseases. METHODOLOGY/PRINCIPAL FINDINGS: Using patient skin biopsies, cell culture and murine infection models, HIF-1 activation was determined by immunohistochemistry, immunoblotting and reporter gene assays and was linked to cellular oxygen consumption. The course of a S. aureus peritonitis was determined upon pharmacological HIF-1 inhibition. Activation of HIF-1 was detectable (i) in all ex vivo in biopsies of patients suffering from skin infections, (ii) in vitro using cell culture infection models and (iii) in vivo using murine intravenous and peritoneal S. aureus infection models. HIF-1 activation by human pathogens was induced by oxygen-dependent mechanisms. Small colony variants (SCVs) of S. aureus known to cause chronic infections did not result in cellular hypoxia nor in HIF-1 activation. Pharmaceutical inhibition of HIF-1 activation resulted in increased survival rates of mice suffering from a S. aureus peritonitis. CONCLUSIONS/SIGNIFICANCE: Activation of HIF-1 is a general phenomenon in infections with human pathogenic bacteria, viruses, fungi and protozoa. HIF-1-regulated pathways might be an attractive target to modulate the course of life-threatening infections

    Improving mental well-being in psychocardiology—a feasibility trial for a non-blended web application as a brief metacognitive-based intervention in cardiovascular disease patients

    Background: Many patients with cardiovascular disease also show a high comorbidity of mental disorders, especially such as anxiety and depression. This is, in turn, associated with a decrease in the quality of life. Psychocardiological treatment options are currently limited. Hence, there is a need for novel and accessible psychological help. Recently, we demonstrated that a brief face-to-face metacognitive therapy (MCT) based intervention is promising in treating anxiety and depression. Here, we aim to translate the face-to-face approach into digital application and explore the feasibility of this approach. Methods: We translated a validated brief psychocardiological intervention into a novel non-blended web app. The data of 18 patients suffering from various cardiac conditions but without diagnosed mental illness were analyzed after using the web app over a two-week period in a feasibility trial. The aim was whether a non-blended web app based MCT approach is feasible in the group of cardiovascular patients with cardiovascular disease. Results: Overall, patients were able to use the web app and rated it as satisfactory and beneficial. In addition, there was first indication that using the app improved the cardiac patients’ subjectively perceived health and reduced their anxiety. Therefore, the approach seems feasible for a future randomized controlled trial. Conclusion: Applying a metacognitive-based brief intervention via a non-blended web app seems to show good acceptance and feasibility in a small target group of patients with CVD. Future studies should further develop, improve and validate digital psychotherapy approaches, especially in patient groups with a lack of access to standard psychotherapeutic care

    Untersuchungen zur Bedeutung des Lipopolysaccharid-bindenden Proteins (LBP) fĂŒr Mikroorganismen des Magen-Darm-Traktes von BALB/c-LBP+/+ - und BALB/c-LBP-/- (Knock-out)-MĂ€usen

    Durch Forschungsarbeiten der neueren Zeit ist die Bedeutung der Akut-Phase-Proteine als wichtiger Bestandteil der unspezifischen Abwehr im Organismus sichtbar geworden. Die möglichen Wechselwirkungen zwischen der physiologischen Magen-Darm-Flora und diesen Proteinen sind jedoch weitesgehend unbekannt. In dieser Arbeit wurde die Magen-Darm-Flora von einem LBP-/--Knock-out-Maus-Stamm und dem Wildstamm (jeweils 20 Tiere) untersucht. Dabei wurde die aerobe Gesamtkeimzahl, die aerobe und anaerobe Gram-negative Gesamtkeimzahl und die Gesamtkeimzahl der auf MRS-Agar gewachsenen Keime in Darminhaltsproben der Darmabschnitte Jejunum, ZĂ€kum und Kolon von oben genannten MĂ€usen erfasst. ZusĂ€tzlich wurden sieben serologische Untersuchungen bei 40 Tieren durchgefĂŒhrt, um mögliche parallele immunologische Reaktionen des Körpers auf Bestandteile der physiologischen Magen-Darm-Flora zu erfassen und vergleichend zu betrachten. Bei den Untersuchungen konnten hochsignifikante Unterschiede bezĂŒglich der Höhe und Zusammensetzung der aeroben Gesamtkeimzahl, der aeroben Gram-negativen Gesamtkeimzahl, der anaeroben Gram-negativen Gesamtkeimzahl und der Laktobazillen-Gesamtkeimzahl festgestellt werden. Dabei waren bei der LBP-/--Gruppe, den gendeletierten MĂ€usen, die aerobe Gesamtkeimzahl signifikant erhöht, die anderen Keimzahlen, insbesondere die Keimzahlen der aeroben und anaeroben Gram-negativen Bakterien und der Laktobazillen signifikant erniedrigt. ZusĂ€tzlich konnten Abweichungen verschiedener Koloniemerkmale wie HĂ€molyseverhalten, Koloniemorphologie und GenausprĂ€gung zwischen der LBP-/-- und der LBP+/+-Gruppe festgestellt werden. Bei den serologischen Untersuchungen konnten die Ergebnisse der mikrobiologischen Untersuchungen bestĂ€tigt werden. Es wurden signifikante Erhöhungen der relativen Antikörperspiegel gegenĂŒber ausgewĂ€hlten bakteriellen Strukturen, von Mikroorganismen, welche auch signifikant hĂ€ufiger von den jeweiligen Gruppen isoliert werden konnten, festgestellt. Diese Ergebnisse lassen auf einen möglichen Zusammenhang zwischen der Gendeletion, also der Nichtexpression von LBP, und der Zusammensetzung der Magen-Darm-Flora schließen. Inwiefern dies auf direkten oder indirekten Einfluss des LBP auf die Magen-Darm-Flora zurĂŒckzufĂŒhren ist, mĂŒssen weitere Untersuchungen zeigen. Dabei sollten noch andere Einflussfaktoren, wie z. B. die Varianz des LBP in seiner genotypischen AusprĂ€gung, berĂŒcksichtigt werden
