Simulation ausgewählter Heuristiken zur Tourenplanung in manuellen Kommissionierstationen

Im Folgenden werden die Auswirkungen des Einsatzes verschiedener Heuristiken zur Tourenplanung in manuellen Kommissionierstationen bei einem Pharmagroßhandel untersucht. Dabei wird nicht nur auf die Läange der Touren eingegangen, sondern zusätzlich die Auswirkung von Ressourcenkonflikten während der Kommissionierung auf die Dauer der Bedienzeiten betrachtet. Nach einer Beschreibung des Systems werden dazu zuerst der Entwurf und die Implementierung eines agentenbasierten Modells dargestellt. Daraufhin werden einige häufig verwendete Heuristiken eingeführt und für den Anwendungsfall evaluiert. Am Schluss steht eine kurze Zusammenfassung und ein Ausblick auf das weitere Vorgehen

Reducing blocking effects in multi-block layouts

Tour planning in multi-block layouts is a common exercise in logistics. In those systems, blocking effects result from conflicting agents competing for resources. Although clearly exceptional in real world applications, most methods of tour planning assume only one active agent, and thus do not consider blocking effects. In this paper we examine heuristic methods of tour planning in multi-block layouts with multiple agents, finding that blocking effects have a significant impact on system performance. We show that methods devised for the mentioned special case do not scale very well when applied to scenarios with multiple agents. We propose a heuristic method which is capable of reducing blocking effects. It generates tours of equal or shorter length than those produced by the other examined methods

Critical role of reactive oxygen species (ROS) for synergistic enhancement of apoptosis by vemurafenib and the potassium channel inhibitor TRAM-34 in melanoma cells

Inhibition of MAP kinase pathways by selective BRAF inhibitors, such as vemurafenib and dabrafenib, have evolved as key therapies of BRAF-mutated melanoma. However, tumor relapse and therapy resistance have remained as major problems, which may be addressed by combination with other pathway inhibitors. Here we identified the potassium channel inhibitor TRAM-34 as highly effective in combination with vemurafenib. Thus apoptosis was significantly enhanced and cell viability was decreased. The combination vemurafenib/TRAM-34 was also effective in vemurafenib-resistant cells, suggesting that acquired resistance may be overcome. Vemurafenib decreased ERK phosphorylation, suppressed antiapoptotic Mcl-1 and enhanced proapoptotic Puma and Bim. The combination resulted in enhancement of proapoptotic pathways as caspase-3 and loss of mitochondrial membrane potential. Indicating a special mechanism of vemurafenib-induced apoptosis, we found strong enhancement of intracellular ROS levels already at 1 h of treatment. The critical role of ROS was demonstrated by the antioxidant vitamin E (α-tocopherol), which decreased intracellular ROS as well as apoptosis. Also caspase activation and loss of mitochondrial membrane potential were suppressed, proving ROS as an upstream effect. Thus ROS represents an initial and independent apoptosis pathway in melanoma cells that is of particular importance for vemurafenib and its combination with TRAM-34

This paper describes the development and application of a simulation software representing their logistic processes, built in cooperation with Gehe's Troisdorf branch. We start with an outline of the Gehe facility, describe the process of implementing and validating the software application, and show some sample results

Lost in the City? - A Scoping Review of 5G Enabled Location-Based Urban Scenarios

5G mobile network technologies and scenarios with the associated innovations receive growing interest among academics and practitioners. Current literature on 5G technologies discusses several scenarios and specific chances and challenges. However, 5G literature is fragmented and not systematically reviewed. We conducted a scoping review on 5G applications in urban scenarios. We reviewed 1,394 papers and identified 20 studies about urban logistics and emergency indoor localization. Our review accumulates current academic knowledge on these scenarios and identifies six further research directions in four research fields. It reveals several further research opportunities, e.g., regarding trust and privacy concerns. We review and discuss 5G literature for academics and practitioners, contribute towards more tailored 5G research and reflect on cost- efficient 5G applications in urban scenarios

QUANTIFYING DISPERSAL AND ESTABLISHMENT LIMITATION IN A POPULATION OF AN EPIPHYTIC LICHEN

Dispersal is a process critical for the dynamics and persistence of metapopulations, but it is difficult to quantify. It has been suggested that the old-forest lichen Lobaria pulmonaria is limited by insufficient dispersal ability. We analyzed 240 DNA extracts derived from snow samples by a L. pulmonaria-specific real-time PCR (polymerase chain reaction) assay of the ITS (internal transcribed spacer) region allowing for the discrimination among propagules originating from a single, isolated source tree or propagules originating from other locations. Samples that were detected as positives by real-time PCR were additionally genotyped for five L. pulmonaria microsatellite loci. Both molecular approaches demonstrated substantial dispersal from other than local sources. In a landscape approach, we additionally analyzed 240 snow samples with real-time PCR of ITS and detected propagules not only in forests where L. pulmonaria was present, but also in large unforested pasture areas and in forest patches where L. pulmonaria was not found. Monitoring of soredia of L. pulmonaria transplanted to maple bark after two vegetation periods showed high variance in growth among forest stands, but no significant differences among different transplantation treatments. Hence, it is probably not dispersal limitation that hinders colonization in the old-forest lichen L. pulmonaria, but ecological constraints at the stand level that can result in establishment limitation. Our study exemplifies that care has to be taken to adequately separate the effects of dispersal limitation from a limitation of establishment
